Methylentetrahydrofolatereductase (rs1801133) polymorphism and psoriasis: contribution to oxidative stress, lipid peroxidation and correlation with vascular adhesion protein 1, preliminary report

Abstract: In the present study, we demonstrated for the first time that the psoriasis patients with MTHFR-677-T (C/T + T/T) allele had higher serum levels of MDA, VAP-1, APOB and ratio of APOB/APOA1 and dominant/recessive model (CC+CT/TT) and T allele of MTHFR-677 are significantly more common in psoriasis and increased risk of psoriasis by 7.45 and 1.76 fold, respectively. These data suggest that psoriasis patients carrying of TT genotypes and T allele of MTHFR-677 may be more susceptible to cardiovascular disease and … Show more

“…To date, a number of casecontrol studies have been performed to investigate the association between MTHFR 677C/T polymorphism and psoriasis risk (Baiqiu et al, 2000;Jie et al, 2007;Weger et al, 2008;Vasku et al, 2009;Dehui et al, 2012;Liew et al, 2012;Asefi et al, 2014). However, the conclusion was controversial, which may have been partly due to a small sample size in individual studies or differences in various ethnic groups.…”

“…Moreover, a lower frequency of the tyrosine phosphatase (SHP-1) gene and p16 gene demethylation has been observed in psoriatic skin lesions (Ruchusatsawat et al, 2006;Zhang et al, 2007). However, several case-control studies carried out subsequently have reported inconsistent results (Weger et al, 2008;Vasku et al, 2009;Liew et al, 2012;Asefi et al, 2014). Therefore, we performed the present meta-analysis to comprehensively evaluate the association between the MTHFR C677T polymorphism and psoriasis risk based on all eligible case-control studies.…”

“…In China, where the first study on the subject was conducted, the prevalence of TT genotype was found to be high in patients with psoriasis (n=39) 37 . Similarly, the prevalence of T allele containing genotypes was found to be significantly higher in psoriatic patients in Hatay (n=96) and in Iran (n=100) compared to controls 36,48 . It was shown in a study conducted in Istanbul in 60 patients without a control group that the blood homocysteine levels were high in the psoriatic patients with TT and CT genotypes 47 .…”

“…Conditions causing elevated homocysteine include genetic defects of enzymes such as methylenetetrahydrofolate reductase (MTHFR), methionine synthase and cystathionine β synthase 11 . The most common defect seen worldwide is mild or moderate hyperhomocysteinemia accompanied by single nucleotide polymorphism in the MTHFR enzyme, which has been found to be associated with psoriasis in some studies 6,11,36,37 . Since plasma homocysteine level is affected by eating habits 38 and the genetic polymorphisms of the enzymes involved in its metabolism differ depending on the geographic region and ethnicity 15 , we aimed at investigating plasma homocysteine and homocysteine-related vitamin levels in psoriatic patients living in our region.…”

“…In many native and foreign publications there was stated that all above mentioned systems are closely connected with each other and the shifts in any of them directly react with certain changes in other systems which intended for compensation of initial systems. When possibilities of compensations on the part of other systems are exhausted the development of pathological processes with appropriate clinical presentations shall start [13].…”
