A Global eDNA Comparison of Freshwater Bacterioplankton Assemblages Focusing on Large-River Floodplain Lakes of Brazil

With its network of lotic and lentic habitats that shift during changes in seasonal connection, the tropical and subtropical large-river systems represent possibly the most dynamic of all aquatic environments. Pelagic water samples were collected from Brazilian floodplain lakes (total n = 58) in four flood-pulsed systems (Amazon [n = 21], Araguaia [n = 14], Paraná [n = 15], and Pantanal [n = 8]) in 2011–2012 and sequenced via 454 for bacterial environmental DNA using 16S amplicons; additional abiotic field and laboratory measurements were collected for the assayed lakes. We report here a global comparison of the bacterioplankton makeup of freshwater systems, focusing on a comparison of Brazilian lakes with similar freshwater systems across the globe. The results indicate a surprising similarity at higher taxonomic levels of the bacterioplankton in Brazilian freshwater with global sites. However, substantial novel diversity at the family level was also observed for the Brazilian freshwater systems. Brazilian freshwater bacterioplankton richness was relatively average globally. Ordination results indicate that Brazilian bacterioplankton composition is unique from other areas of the globe. Using Brazil-only ordinations, floodplain system differentiation most strongly correlated with dissolved oxygen, pH, and phosphate. Our data on Brazilian freshwater systems in combination with analysis of a collection of freshwater environmental samples from across the globe offers the first regional picture of bacterioplankton diversity in these important freshwater systems.Electronic supplementary materialThe online version of this article (doi:10.1007/s00248-016-0834-5) contains supplementary material, which is available to authorized users.

Since the relative role of local and regional abiotic factors on the Odonata diversity in rainforest streams is still poorly understood, we evaluated the effects of these factors on adult Odonata (Insecta) from preserved and altered streams in the Amazonian region. Adult Odonata were sampled in 98 streams in the Eastern Amazon, Pará, Brazil. Six variables were used to measure local environmental factors: habitat integrity index; mean canopy over the channel; and four physical and chemical descriptors of the water. To measure regional environmental factors, six variables were also used: altitude gradient, three bioclimatic variables and two percentage forest variables. In partial redundancy analysis, both abiotic factors (local and regional) were important to explain the variation in the Odonata community. The Odonata community can be influenced by regional and local factors. The relationship between Odonata and the local (e.g., integrity, canopy cover, and physical and chemical descriptors of the water) and regional (e.g., bioclimatic and forest cover variables) environmental variables recorded in this study has important implications for the use of these organisms to monitor small streams of the Eastern Amazon. The scale at which habitat is measured is an important issue in community structuring studies considering the rapid environmental changes. It is of great importance to consider the different scales in studies assessing community structure, once an adequate habitat must meet the ecological needs of all stages of the life of the Odonata.

Identifying Key Stressors Driving Biological Impairment in Freshwater Streams in the Chesapeake Bay Watershed, USA

Biological communities in freshwater streams are often impaired by multiple stressors (e.g., flow or water quality) originating from anthropogenic activities such as urbanization, agriculture, or energy extraction. Restoration efforts in the Chesapeake Bay watershed, USA seek to improve biological conditions in 10% of freshwater tributaries and to protect the biological integrity of existing healthy watersheds. To achieve these goals, resource managers need to better understand which stressors are most likely driving biological impairment. Our study addressed this knowledge gap through two approaches: 1) reviewing and synthesizing published multi-stressor studies, and 2) examining 303(d) listed impairments linked to biological impairment as identified by jurisdiction regulatory agencies (the states within the watershed and the District of Columbia). Results identified geomorphology (i.e., physical habitat), salinity, and toxic contaminants as important for explaining variability in benthic community metrics in the literature review. Geomorphology (i.e., physical habitat and sediment), salinity, and nutrients were the most reported stressors in the jurisdictional impairment analysis. Salinity is likely a major stressor in urban and mining settings, whereas geomorphology was commonly reported in agricultural settings. Toxic contaminants, such as pesticides, were rarely measured; more research is needed to quantify the extent of their effects in the region. Flow alteration was also highlighted as an important urban stressor in the literature review but was rarely measured in the literature or reported by jurisdictions as a cause of impairment. These results can be used to prioritize stressor monitoring by managers, and to improve stressor identification methods for identifying causes of biological impairment.
