2‐(1H‐Tetrazol‐5‐yl)‐4,5‐dihydro‐4‐oxo‐indeno[1,2‐b] pyran

Abstract

Es wird uber die Synthese der Titelverbindung berichtet. Die Ergebnisse der Priifung auf antiallergische Wirksamkeit fir die bioisosteren Heterocyclen 8b und 8c wird mitgeteilt. Versuche, durch ON-Austausch zu 4-Pyridonen zu gelangen, werden beschrieben. Zur Darstellung von 8a wurden zwei Wege A und B eingeschlagen, die ausgehend vondem 4-Pyron-2-carbonsaureethylester 13) zur Schliisselverbindung, dem Nitril 7, fiihren sollten.Auf dem kurzeren Weg A bestand das Problem lediglich darin, das aus 1 gut zugangliche Amid 93) zu 7 zu dehydratisieren. 9 ist in den iiblichen Losungsmitteln sehr schwer loslich, so daB z.T. in Suspensionen gearbeitet werden muate. Alle Versuche (1.4-Toluolsulfonylchlorid/Pyridin/DMF4); 2. Acetanhydrid'); 3. POCl,/DMF6); 4. DiglymeA'aBH:);5. Oxalylchlorid/DMF8) und 6. HMPT9)) scheiterten jedoch.++ Herrn Prof. Dr. A.
