Micellar high performance liquid chromatographic determination of Itraconazole in bulk, pharmaceutical dosage forms and human plasma

Abstract: A micellar liquid chromatographic method was developed for determination of Itraconazole in bulk, dosage form and human plasma using µBondapack cyano column and a mobile phase consisting of 0.1 M sodium dodecyl sulphate, 20% 1-propanol, 0.3% triethylamine in 0.02 M orthophosphoric acid (pH = 3.5). The UV detection was achieved at 258 nm. Various chromatographic parameters were studied, e.g. types of columns, pH of mobile phase, concentration of sodium dodecyl sulphate, 1-propanol, triethylamine, etc .The metho… Show more

“…Micellar mobile phases usually need less quantity of organic modifier and generate less amount of toxic waste in comparison to aqueous-organic solvents, so that they are less toxic, non-inflammable, biodegradable and relatively inexpensive [18]. MLC has proved to be a useful technique in the determination of diverse [19][20][21][22][23][24], including food samples [25][26][27][28].…”

“…It has been reported that ITR can undergo a primary dehalogenation process under UV light irradiation to form a biologically reactive aryl radical that may induce phototoxicity or photoallergy side effects. Some basic photochemical properties of ITR have been investigated using UV spectrophotometric, high-performance liquid chromatography and/or chromatography methods, and a photoreaction product analysis has also been done by Miranda and co-workers . However, there is no in-depth time-resolved spectroscopic investigation about the specific mechanism of its photoreaction.…”
