Purification and partial characterization of phospholipases A2 from Bothrops asper (barba amarilla) snake venom from Chiriguaná (Cesar, Colombia)

Abstract: ABSTRACT.Components with phospholipase A 2 activity were isolated by gel filtration and cationic exchange chromatography from the venom of Bothrops asper snakes from Chiriguaná, Colombia (9°22´N; 73°37´W). Five fractions were obtained by the gel filtration, and PLA 2 activity was found in fraction 3 (F3). In the cationic exchange chromatography, F3showed eight components with PLA 2 activity. Six of these components appeared as one band in polyacrylamide gel electrophoresis (SDS-PAGE). Fractions II and VII exhi… Show more

“…Phospholipase A2 activity was evaluated as described by Habermann and Hardt [ 96 ]. Twenty micrograms of the lyophilized soluble venoms from S. gertschi and the snake Bothrops asper (positive control [ 97 ]) was resuspended in 10 µL of distilled water and deposited into small wells in egg yolk agarose. Samples containing just water were used as negative control.…”

“…Phospholipases A2 (PLA2) (EC.3.1.1.4) catalyze the hydrolysis of the 2-acyl ester linkage of 1, 2-diacyl-3-snphosphoglycerides, with calcium (Ca 2+ ) requirement [1,2] producing fatty acids and lysophospholipids [3]. Snake venoms are particularly rich in PLA2s [4] and PLA2s are able to induce several biological effects, such as pre-synaptic or post-synaptic neurotoxicity, cardiotoxicity, myotoxicity, platelet aggregation, oedema, haemolysis, anticoagulation, convulsion, and hypotension [5]. Their catalytic activity upon cell membranes of tissues suggests an important role of the enzyme in venom toxicity [6].…”
