Articular inflammation induced by an enzymatically-inactive Lys49 phospholipase A2: activation of endogenous phospholipases contributes to the pronociceptive effect

Abstract: BackgroundArthritis is a set of inflammatory conditions that induce aching, stiffness, swelling, pain and may cause functional disability with severe consequences to the patient’s lives. These are multi-mediated pathologies that cannot be effectively protected and/or treated. Therefore, the aim of this study was to establish a new model of acute arthritis, using a Lys49-PLA2 (Bothrops asper myotoxin II; MT-II) to induce articular inflammation.MethodsThe articular inflammation was induced by MT-II (10 μg/joint)… Show more

“…Joint diseases, such as rheumatoid arthritis and osteoarthritis, can cause functional disability, interfering with the patient’s quality of life. Arthritis symptoms include aching, stiffness, swelling, and pain in the affected joint [ 144 , 145 ]. Effective and/or protective treatments for these pathologies are still a challenge [ 146 , 147 ].…”

“…These can induce inflammation [124]; e.g., MT-II can stimulate the production and release of inflammatory mediators, such as interleukin-6 [125], interleukin-1, tumour necrosis factor α, leukotriene B4, thromboxane A2 and prostaglandins E2 and D2 [126][127][128][129]. Indeed, this activity of MT-II was recently used to establish a new experimental model of acute arthritis [130].…”

“…Among these sources, snake venoms have a wide variety of components, where about 90% of their dry weight is composed of proteins, among which are phospholipases A 2 (PLA 2 ), enzymatic proteins that generally have low molecular weight. These are responsible for catalyzing the hydrolysis of the 3-sn-phosphoglyceride-dependent calcium 2-acyl ester bond, obtaining lysophospholipids and fatty acid products [18][19][20].…”
