To develop a montelukast sodium-loaded clear oral solution with enhanced stability that was bioequivalent to commercial granules in rats for the treatment of asthma, various montelukast sodium-loaded solutions were prepared with various amounts of solubilizers and stabilizer, and their solubility and stability were investigated. Furthermore, the dissolution and pharmacokinetic studies were carried out in rats with the optimised formulation and the commercial montelukast sodium-loaded granules. Among the solubilizers tested in this study, hydroxypropyl-b-cyclodextrin (HP-b-CD) was selected because it greatly improved the drug solubility and stability. Furthermore, EDTA sodium was used as a stabilizer because it gave excellent stability. In particular, the montelukast-loaded oral solution, an aqueous clear solution containing montelukast sodium, HP-b-CD, methylparaben sodium, propylparaben sodium and EDTA sodium at w/v percentages of 1.04/156/1.8/0.2/1, gave similar dissolution to the commercial granules in 0.5 % (w/v) sodium lauryl sulphate in water, FDA-regulated dissolution medium. This oral solution gave similar plasma concentrations and pharmacokinetic parameters to the commercial granules, suggesting that it was bioequivalent to the commercial granules in rats. Moreover, it was physically and chemically stable at 25°C/60 % RH and 40°C/75 % RH for at least 12 months. Thus, this oral solution is strongly recommended as an alternative to the oral montelukast-loaded product for the treatment of asthma.

Introdução: Alergia é um quadro de hipersensibilidade onde o organismo responde de maneira excessiva em contato com um antígeno. Na prática odontológica as mais comuns são as do tipo I e IV. As reações do tipo I na cavidade oral se manifestam como angioedema, sensação de ardor ou parestesia oral, enquanto nas reações de hipersensibilidade tipo IV, os sintomas são as reações liquenóides, eritema e estomatite. O presente estudo se propõe a realizar uma revisão integrativa sobre as muitas causas de reações alérgicas no âmbito odontológico e suas interrelações com outros alérgenos. Metodologia: Foram realizadas buscas nas bases de dados PubMed e LILACS utilizando os termos “allergy”, “dentistry” “materials”, “allergic reaction” e “dentist” que foram combinados usando os operadores booleanos “AND”. Foram incluídos artigos no intervalo de tempo de 2002 a 2022 na língua inglesa, portuguesa e espanhola e a busca resultou em 21 artigos. Resultados e Discussão: A odontologia vem sofrendo grandes inovações e, com isso, uma gama de materiais são desenvolvidos e disponibilizados no mercado. Mesmo criados com princípios de biocompatibilidade, há casos onde se pressupõe falhas nesse princípio, ocasionando as reações alérgicas. Essas reações são causadas por diversos materiais, sendo os mais comuns às luvas de látex, anestésicos, materiais de moldagem e metais. Conclusão: Sempre que houver suspeita de alergia a algum material, mesmo que remota, o profissional deverá alterar o protocolo de atendimento lançando mão de materiais alternativos para evitar emergências médicas no consultório odontológico.
