Bovine papillomavirus type 4 L1 gene transfection in a Drosophila S2 cell expression system: absence of L1 protein expression

Abstract: The development of a bovine papillomavirus (BPV) vaccine is an outstanding challenge. BPV protein L1 gene transfection in the Drosophila melanogaster S2 cell expression system failed to produce L1 protein notwithstanding correct L1 gene insertion. Severe genetic inbalance in the host cell line, including cytogenetic alterations, may account for the lack of protein expression.

“…According to these latter authors, Debruyne et al (2005), there are 14 validly described Campylobacter species. More recently, Fernández et al (2008) stated that the genus comprises 20 species and subspecies. However, other authors have stated that there are 16 species with a further six subspecies within the genus Campylobacter (On, 2001; Foster et al, 2004).…”

“…The idea of developing a vaccine to combat BPV began with the infection of Shope papillomas extract in the 1940 decade ( Shope, 1937 ). Since then, different vaccine models were proposed ( Jarret et al , 1991 ; Gaukroger et al , 1996 ; Góes et al , 2008 ; Love et al , 2012 ; Mazzuchelli-de-Souza et al , 2013 ). However, none of them became a commercial product.…”

“…The BPV icosahedral capsid has 360 copies of the L1 protein of 55 kDa, organized in 72 capsomers and approximately 12 copies of the L2 protein with 39 kDa (Campo, 1997;Góes et al, 2008;Meinke and Meinke, 1981) having a molecular weight of 5.2 × 10 6 Da (Orth et al, 1977). The BPV genome is divided into three regions: early, late and long control, separated by two polyadenylation sites http://dx.doi.org/10.1016/j.jviromet.2014.07.029 0166-0934/© 2014 Elsevier B.V.
