In order to do an in vitro propagation of Casearia sylvestris, a tree native to Brazilian forests, studies of in vitro seed germination were performed and aseptic seedlings obtained were used as explant donors for micropropagation. Mature and immature seeds, stored for 10 days at 10 or 25°C were inoculated in medium containing 10% of MS salts and kept in the light (a 16-hour photoperiod) or continuous dark, at 25°C. For immature seeds, gibberellic acid (GA 3 ) was or was not tested at a concentration of 1.5 mg l -1 . From the seedlings obtained, the cotyledon and apical segments were removed and cultured in complete MS medium with five combinations of 6-benzylaminopurine (BAP) and naphthalene acetic acid (NAA): 0.0; 0.25; 0.5; 1.0; 2.0 and 0.0; 0025; 0.05; 0.1; 0.2 mg L-1, respectively. The aerial shoots formed in vitro were cultured in MS medium containing 0.0 or 1.5 mg L -1 indole-3-butyric acid (IBA) for rooting. The highest percentage of germination (60%) occurred in mature seeds exposed to light, regardless of other treatments, and may be considered positive photoblastic. The highest percentage of germination of immature seeds (27%) occurred when previously stored at 10 ° C with or without GA 3 . The induction of aerial shoots was more efficient in apical segments in a medium containing the lowest combination of BAP and NAA or free of these growth regulators. The higher number of roots occurred in aerial shoots obtained from cotyledon segments, with or without IBA. After acclimatization, 83% of the plants survived. RESUMOCom o objetivo obter a propagação in vitro de Casearia sylvestris, árvore nativa das florestas brasileiras, estudos de germinação in vitro das sementes foram realizados e as plântulas assépticas obtidas foram usadas como doadoras de explantes na micropropagação. Sementes maduras e imaturas, armazenadas por 10 dias a 10 ou 25°C foram inoculadas em meio contendo 10% de sais de MS e mantidas na luz (fotoperíodo de 16 horas) ou escuro contínuo, a 25°C. Para sementes imaturas, testou-se ou não ácido giberélico (GA 3 ) na concentração de 1,5 mg l -1 . Das plântulas obtidas, segmentos cotiledonares e apicais foram retirados e cultivados em meio MS completo com cinco combinações de 6-benzilaminopurina (BAP) e ácido naftalenoacético (ANA): 0,0; 0,25; 0,5; 1,0; 2,0 e 0,0; 0025; 0,05; 0,1; 0,2 mg L -1 , respectivamente. As brotações aéreas formadas in vitro foram cultivadas em meio MS contendo 0,0 ou 1,5 mg L -1 de ácido Paranhos, J. T. et al. 1192indol-3-butírico (IBA) para o enraizamento. O maior percentual de germinação (60%) ocorreu em sementes maduras expostas à luz, independentemente dos demais tratamentos, podendo ser consideradas fotoblásticas positivas. RESUMOO objetivo deste trabalho foi avaliar a produtividade de microcepas de grápia (Apuleia leiocarpa) mantidas em diferentes condições de cultivo in vitro, visando maximizar a taxa de multiplicação. Microcepas oriundas da poda drástica de plantas assépticas foram cultivadas em meio de cultura WPM acrescido de 0; 2,2; 4,4; 6,6 e 8,8 µM de 6-benzilaminopurina (BAP) e subcultivadas em meio de cultura WPM sem citocinina. Microcepas de grápia também foram cultivadas em meio de cultura WPM, MS ou ½ MS, acrescido ou não de 1,5 g L -1 de carvão ativado e submetidas a três coletas de brotos aos 30, 60 e 90 dias de cultivo. A manutenção das microcepas em meio de cultura WPM ou ½ MS suplementado com 8,8 µM de BAP, seguido do subcultivo em meio de cultura sem citocinina aumenta o número e o crescimento de brotações. O cultivo de microcepas de grápia em meio de cultura com carvão ativado aumenta a produção de microestacas por microcepas, enquanto a ausência de carvão ativado favorece a formação de calos e brotos por organogênese indireta. A manutenção in vitro de microcepas de grápia maximiza a taxa de multiplicação, constituindo-se de uma técnica promissora para a produção de plantas de grápia. ABSTRACTThe aim of this work was to evaluate the in vitro productivity of grapia (Apuleia leiocarpa) micro-stumps cultivated in different media conditions to maximize its multiplication rate. Micro-stumps from aseptic seedlings were cultivated in WPM medium added with 0, 2.2, 4.4, 6.6 and 8.8 µM of 6-benzylaminopurine (BAP) and subcultured in WPM medium without cytokinin. Micro-stumps were also cultivated in WPM, MS or ½ MS base media with or without the addition of 1.5 g L -1 of activated charcoal and submitted to drastic pruning at 30, 60 and 90 days of cultivation. The in vitro cultivation of grapia micro-stumps in WPM or ½ MS media added with 8.8 µM of BAP and the subcultivation in media without cytokinin increase the number and length of shoots. The cultivation of grapia micro-stumps in media with activated charcoal increases the productivity of micro-cuttings per micro-stumps, but in the absence of activated charcoal induces callus formation and indirect organogenesis of shoots. The in vitro conservation of grapia microstumps maximizes the multiplication rate, being a feasible technique for the production of grapia plantlets.

RESUMO:O presente estudo teve por objetivo avaliar o efeito de BAP, na presença e ausência de ANA, sobre a multiplicação in vitro de segmentos apicais caulinares de Satureja hortensis. Os explantes foram isolados de plântulas germinadas in vitro e cultivados em meio nutritivo MS. O delineamento experimental utilizado foi o inteiramente casualizado, em esquema fatorial 2x5, correspondendo às concentrações de ANA (0 e 1 µM) e de BAP (0; 5; 10; 15 e 20 µM), com seis repetições, cada uma composta por três explantes. Para a porcentagem de explantes com brotações houve efeito significativo para o fator BAP, aumentando à medida que cresce a concentração da citocinina. A variável "número de brotos por explante" apresentou interação entre os fatores, havendo a maior formação de brotos na presença de ANA na faixa de 10 -15 µM de BAP. Na presença da auxina, o maior valor ocorreu com 15 µM de BAP tendendo a diminuir independente da presença de ANA. O enraizamento dos segmentos apicais foi maior na presença de ANA e ausência de BAP, diminuindo com o aumento da citocinina. O número de folhas foi influenciado pela maior concentração de BAP sendo que a 15 µM foi observado o maior número, tendendo à queda com aumento da concentração. A concentração de 15 µM de BAP, independente de ANA, proporcionou os melhores resultados na multiplicação da espécie produzindo aumento de brotações e número de folhas, à exceção do enraizamento que foi influenciado pela auxina.Palavras-chave: cultura de tecidos, regulador de crescimento, plantas medicinais, Lamiaceae. ABSTRACT:In vitro multiplication of apical shoot segments of Summer Savory(Satureja hortensis L.).This study aimed to evaluate the effect of BAP in the presence and absence of NAA, on the in vitro multiplication of shoot apical segments of Satureja hortensis. The explants were isolated from seedlings germinated in vitro and cultured in nutrient medium MS. The experimental design was completely randomized in a 2x5 factorial arrangement, corresponding to the concentrations of NAA (0 and 1 µM) and BAP (0, 5, 10, 15 and 20 µM), with six replicates, each consisting of three explants. For the percentage of explants with shoots, there was no significant effect for the factor BAP, increasing as the concentration of BAP grows. The variable number of shoots per explant showed interaction between factors, with the highest shoot formation in the presence of NAA and BAP in the range of 10 -15 µM. In the presence of auxin, the highest value occurred with 15 µM BAP, tending to decline regardless of the presence of NAA. The rooting of the apical segments was higher in the presence of NAA and absence of BAP, decreasing with increasing cytokinin. Leaf number was influenced by the higher concentration of BAP, and the amount of 15 µMhadthe largest number, tending to decrease with increasing concentration.
