Laelia halbingeriana Salazar & Soto Arenas is a wild epiphytic orchid endemic to Oaxaca, Mexico, that is collected with no plans for its management or conservation. The objective of this study was to propagate and evaluate the in vitro development of this species in the culture media with a variety of mineral salts and growth regulators. Groups of three plants, 1.2 to 1.4 cm tall, were established in receptacles with culture medium containing 30 g L-1 sucrose, 1 mg L-1 thiamine, 100 mg L-1 inositol and 0.5 mg L-1 benzyl aminopurine (BAP). Proportions of Murashige and Skoog (1962) or Knudson (1946) mineral salts were varied (100, 66 and 33%). The growth regulators indoleacetic acid (IAA) and kinetin (Kin) were used at various concentrations (0.5 + 0.5 mg L-1 , 1 + 1 mg L-1 and 2 + 2 mg L-1). pH was adjusted to 5.8, and 5.5 g L-1 agar was added. After 120 days of incubation, the plants that were cultivated in 66% and 100% Knudson media were 2.1 cm tall and had 4.8 roots, while the plants in 100% Murashige and Skoog media were 1.5 cm tall and had 2.5 roots. For 120 days, the plants had a logarithmic trend in vertical growth (R 2 ≥0.96). Several plant characteristics were evaluated simultaneously using a cluster analysis, which found that the best culture medium for Laelia halbingeriana development contains 100% Knudson mineral salts, 0.5 mg L-1 BAP, 0.5 mg L-1 IAA, and 0.5 mg L-1 KIN.

Anathallis adenochila is a small epiphytic orchid, endemic to the Atlantic Forest in the southern and southeastern regions of Brazil. Information on the species is scarce and limited to distribution and occurrence data, with no reports about its development. In the present study, plantlets were propagated in vitro, and the influence of different concentrations of macronutrient salts and sucrose on the survival and development of the species was assessed. The analysis were performed on complete Murashige and Skoog (MS) medium and half MS medium (with half-strength macronutrients) containing 10, 30 or 60 g L -1 of sucrose. Plantlet survival, height of the aerial part, and the length of the longest root were significantly greater on half MS medium containing 30 or 60 g L -1 of sucrose. The number of leaves per plantlet was higher in the presence of 60 g L -1 of sucrose, regardless of macronutrient concentration, and the highest number of roots was observed in plantlets cultured on half MS medium with 60 g L -1 of sucrose. This first report of Anathallis adenochila in vitro propagation may contribute to future studies on the physiological and ecological aspects of the life cycle of this species.

Vriesea incurvata in the natural environment shows some difficulties associated to the low seed germination capacity. Aiming to provide basis for the seedlings production, the results of the seeds germination percentage and the initial growth of V. incurvata seedlings in vitro conditions in different culture media, are reported. Completely randomized design was comprised of eight treatments and eight replications. The treatments were as culture media: MS (Murashige & Skoog); MS½; KC (Knudson); KC½; MS + activated carbon (AC); MS½ + AC; KC + AC; and KC½ + AC. The germination percentage was calculated from the division between the number of seeds with primary root extrusion by the total number of sowed seeds. The initial growth was evaluated considering the values of total fresh biomass, percentage of normal and dead seedlings, number and roots length, stem length, number of leaves and percentage of chlorotic, necrotic and dead leaves, respectively. All the cultures promoted high germination percentages (> 82.8%). In MS and MS½ medium, it was evidenced the highest percentage of normal seedlings, the highest values of fresh biomass production, stem growth and number of leaves. KC and KC½ medium also promoted highest percentages of normal seedlings and low percentages of necrotic and dead leaves. The addition of activated carbon in the culture media was unfavorable to promote the growth of seedlings. This suggests that MS and MS½ are the most suitable culture media for the production of Vriesea incurvata in vitro.
