Frequency of and risk factors associated to Leptospira spp. seropositivity in goats in the state of Sergipe, Northeastern... Ciência Rural, v.47, n.7, 2017. INTRODUCTIONLeptospirosis is a major public health problem in Brazil with an annual average rate of 3,780 human cases between 2000 and 2015, 9.6% (364) of which were lethal. In the country's Northeastern region and in the state of Sergipe, lethality is as high as 12.8% (87/681) and 24.1% (10.6/44), respectively (SINAN/SVS/MS, 2017a, b). Leptospirosis is caused by spirochetes of the genus Leptospira. Twenty-five serogroups and approximately 300 serovars belonging to 25 serogroups can infect domestic animals, wildlife, and humans. On rural properties, transmission occurs primarily via the urine of infected animals (GOMES, 2013;HIGINO & AZEVEDO, 2014). Accordingly, rural workers in Jamaica who have contact with goats have a 3.52 times higher chance of presenting symptoms of the disease (KEENAN et al., 2010).Goats usually present chronic infection, with a predominance of embryonic absorption, miscarriage, neonatal mortality, and hemorrhagic jaundice. In the acute form of the disease, a sudden onset of fever, anemia, jaundice, and hemoglobinuria occur, leading to death (MARTINS et al., 2012;HIGINO & AZEVEDO, 2014).In the Northeastern region of Brazil, surveys were performed in the states of Rio Grande do Norte, Ceará, Paraíba, and Pernambuco. The transit of infected animals may be a route for the introduction and dispersal of diseases, which necessitates protocols for the controlled entry of infected animals into herds. The objective of this study was to determine the prevalence of anti-Leptospira and anti-Brucella ovis antibodies in small ruminants sold at an animal fair in the Sertão region of the Pernambuco State, Northeastern Brazil, and to identify the possible risk factors associated with seropositivity. Serum samples were collected from 233 goats and 119 sheep, belonging to twelve different owners from November 2014 to June 2015. Diagnosis of Leptospira spp. infection was established using the microscopic agglutination test (MAT); for B. ovis infection, the agar gel immuno-diffusion (AGID) test was conducted. A total of 16/233 (6.87%; 95% CI = 4.27% -10.83%) goats and 16/119 (13.45%; 95% CI = 8.67% -21.24%) sheep were identified as seropositive for Leptospira spp. The most frequent serotype was Icterohaemorrhagiae. In goats, intensive management system (odds ratio = 15.8; p < 0.001) and consorted rearing of equines and goats (odds ratio = 7.3; p = 0.005) were identified as risk factors. There were no risk factors identified for sheep. Seven (5.88%; 95% CI = 2.88% -11.65%) sheep were seropositive for B. ovis, and the intensive management system was identified as the risk factor (odds ratio = 11.5; p = 0.005). This suggests that Leptospira spp. and B. ovis infections occur in small ruminants commercialized in the Sertão region of the Pernambuco State. Thus, improvements to the sanitary conditions of the facilities and the control of synanthropic rodents are recommended. ResumoO trânsito de animais infectados pode ser uma forma de introdução e dispersão de enfermidades, de maneira que o bloqueio da entrada de animais infectados nos rebanhos é uma importante ferramenta de controle. O objetivo desse estudo foi determinar a frequência de anticorpos anti-Leptospira spp. e antiBrucella ovis em pequenos ruminantes comercializados em feira de animais no Sertão de Pernambuco, Nordeste do Brasil, e identificar os possíveis fatores de risco associados às soropositividades. Foram coletadas amostras de soro de 233 caprinos e 119 ovinos, pertencentes a doze proprietários diferentes, no período de novembro de 2014 a junho de 2015. O diagnóstico da infecção por Leptospira spp. foi realizado pela técnica de soroaglutinação microscópica (SAM), e para B. ovis o teste de imunodifusão em gel de ágar (IDGA) foi aplicado. Foram identificados 16 (6,87%; IC 95% = 4,27% -10,83%) caprinos e 16 (13,45%; IC 95% = 8,67% -21,24%) ovinos soropositivos para Leptospira spp. O sorotipo mais frequente foi o Icterohaemorrhagiae. The objective of this study was to determine the frequency of animals that tested positive for brucellosis and leptospirosis and the risk factors for these diseases in the State of Paraíba. A total of 136 buffaloes from 14 herds were examined. For brucellosis, we used the buffered acidified plate antigen (BAPA) test as screening method and 2-mercaptoethanol as confirmatory test. For leptospirosis, we conducted a microscopic agglutination test (MAT), with a cut-off point of 1:100. Of the animals examined, two [1.5%; 95% CI = 0.4%-5.2%] were positive for brucellosis, and 38 (27.9%; 95% CI = 21.1%-36.0%) were positive for leptospirosis. The brucellosis-positive animals were from two (14.3%) herds, while nine (64.3%) herds had leptospirosis-seropositive animals. The more frequent Leptospira spp. serotypes were Bratislava, Pomona, and Canicola. We concluded that leptospirosis was widespread in buffaloes in the state of Paraíba and suggested that breeding alongside horses and pigs might be an important factor in the spread of leptospirosis-positive animals. The presence of brucellosispositive animals indicated the possibility of negative-impacting measures on disease control in bovines, and it is therefore recommended that greater attention be given to these animals for brucellosis control. Key words: Brucella abortus, buffalo, Leptospira spp, Northeast Brazil, seroepidemiology ResumoO objetivo do trabalho foi determinar a frequência de animais reagentes e os fatores de risco para brucelose e leptospirose em búfalos do Estado da Paraíba. Foram utilizados 136 búfalos oriundos de 14 propriedades. Para o diagnóstico da brucelose empregou-se como teste de triagem o antígeno acidificado tamponado (AAT) e o teste do 2-mercaptoetanol (2-ME) como prova confirmatória. Para leptospirose foi realizado o teste de soroaglutinação microscópica (SAM), com ponto de corte 1:100. Dos 136 animais examinados dois (1,5%; IC 95% = 0,4% -5,2%) foram positivos para brucelose e 38 (27,9%; IC 95% = 21,1% -36,0%) para leptospirose. Os animais positivos para brucelose foram procedentes de duas (14,3%) propriedades, enquanto para leptospirose nove (64,3%) propriedades apresentaram animais soropositivos. Os sorovares de Leptospira spp. mais frequentes foram Bratislava, Pomona e Canicola. Conclui-se que a leptospirose encontra-se disseminada em búfalos do Estado da Paraíba, e sugere-se que a criação consorciada com equinos e suínos pode ser um fator importante na ocorrência de animais positivos. A presença de animais positivos para brucelose indica a possibilidade de impacto negativo nas ações de controle da doença em bovinos, e dessa forma recomenda-se que maior atenção seja dada a esses animais do ponto de vista do controle da brucelose.
