Cytotoxic effects of White-MTA and MTA-Bio cements on odontoblast-like cells (MDPC-23)

Abstract: This study evaluated the cytotoxic effects of 2 mineral trioxide aggregate (MTA) cements - White-MTA-Angelus and a new formulation, MTA-Bio - on odontoblast-like cell (MDPC-23) cultures. Twenty-four disc-shaped (2 mm diameter x 2 mm thick) specimens were fabricated from each material and immersed individually in wells containing 1 mL of DMEM culture medium for either 24 h or 7 days to obtain extracts, giving rise to 4 groups of 12 specimens each: G1 - White-MTA/24 h; G2 - White-MTA/7 days; G3 - MTA-Bio/24 h; a… Show more

“…The relatively poor viability seen with MTA compared with the control (49%) is suggestive of at least an initial toxic effect of MTA. While some of the previously published studies have reported higher cell viability in the presence of MTA (16,17), the data obtained during the first 24 h post-setting perhaps reflected the high pH generated by the material during this period (11). The pH values for MTA of around 12.0 have been reported in other studies (18,19).…”

“…In spite of the critical role of calcium ions in the tissue repair process and also its combination with water leading to the setting expantion of material such as MTA and CEM, a high release of these ions in the culture medium may result in irreversible damage to the cell culture 41) . It should be emphasized that the results obtained from this preliminary genotoxicity test had limitations therefore further studies employing CEM are required to demonstrate its genotoxicity in different methods of evaluation such as LDH, analysis of qRT-PCR, micronucleus, necrosis and apoptosis assay by FACS, confocal laser microscope.…”

“…More recently, new products similar to MTA have been introduced including MTA Angelus (Hashem and Hassanien, 2008), MTA-Bio (Lessa et al, 2010) and BioAggregate (Zhang et al, 2009). BioAggregate is a relatively new bioceramic based material which is similar to MTA.…”
