With the increased demand for high quality maize seeds, the market absorbs innovations that add speed and reliability in the tests that enable differentiation of potential performance of seed lots. This study aims to evaluate the combination of the physiological tests with the expression of enzymes of respiratory route for differences in vigor levels in lots of hybrid seed corn. The percentage of germination, germination first count, emergence on tray, index of emergency speed, accelerated aging and respiration rate were evaluated by Pettenkoffer and Titulation method. Furthermore, the expression of alpha amylase, alcohol dehydrogenase, malate dehydrogenase and pyruvate decarboxylase was analyzed. With the results, it is determined that the vigor tests analyzed, together with the tests of enzymatic expressions of respiratory route were efficient to detect different levels of force in hybrid seed corn. Lot 1 had higher physiological quality while lot 2 had the lowest vigor compared to the other lots. With the vigor tests and expression of enzymes, it was possible to differentiate the quality of the seeds and therefore it can be recommended as reliable tests for defining the quality of seed lots.

-The efficiency of vigor tests in assessing the physiological potential of seeds depends on their standardization for different species. In this context, the research aimed to study specific methodologies to evaluate the vigor of spinach seeds (Tetragonia tetragonoides). For this purpose, five lots of spinach seed cv. New Zealand were submitted to germination, first count of germination, seedling emergence in the field, accelerated aging (with and without use of saturated solution of NaCl at 41and 45 ºC for 24,48 and 72 hours), controlled deterioration(18,21 and 24% seed water content adjustments for 24 hours at 45 ºC) and electrical conductivity tests with variations inwater volume (25, 50 and 75 mL), seed quantity (25 and 50) and soaking period (1, 2, 4, 8,12, 16 and 24 hours) at 25 ºC. The first count test allowed obtaining preliminary information about seed vigor, and other tests in general showed similarity with seedling emergence in the field. Thus, it was concluded that traditional accelerated aging test 24 h /41 ºC and accelerated aging test with saturated salt solution for 24 h at 41 ºC and 45 ºC, and controlled deterioration test with 21% seed water content / 24 h / 45 ºC were all efficient for evaluating the physiological potential of spinach seeds.

O objetivo do estudo foi identificar as variáveis da análise de sementes de abóbora que melhor predizem a emergência de plântulas dessa cultura. O trabalho foi realizado no Departamento de Fitotecnia da Universidade Federal de Santa Maria. Três lotes de sementes de abóbora de cada um dos cultivares 'Caserta', 'De Tronco', 'Caravela' e 'Menina Brasileira' foram utilizados. Foi determinada a percentagem de plântulas normais nos testes de germinação, primeira contagem de germinação, teste de envelhecimento acelerado, temperatura subótima, teste de frio, massa seca de raiz, de hipocótilo e de total de plântula, comprimento de raiz, comprimento de hipocótilo, comprimento total de plântula, emergência de plântulas, índice de velocidade de emergência e comprimento do hipocótilo na emergência. As variáveis avaliadas nas plântulas foram submetidas à análise de trilha e a percentagem de plântulas normais obtidas nos testes de sementes, submetidas à análise de regressão múltipla stepwise, considerando a emergência de plântulas como variável resposta. Foi realizada também a análise de fatores com todas as variáveis avaliadas. O teste de temperatura subótima foi o melhor estimador da emergência de plântulas da cultura, para todos os cultivares avaliados.
