The present study aimed to describe epidemiological, clinical, laboratorial, and pathological findings of polioencephalomalacia (PEM) in ruminants from the semi-arid region of Rio Grande do Norte, Brazil. A total of seven ruminants (five sheep, one cattle, and one goat) with pathological diagnosis of PEM were included. Four cases were associated with thiamine deficiency, on account of chronic ruminal acidosis caused by diets rich in carbohydrate, with mainly concentrates, ground soybean hulls, and melon. Three ruminants from an outbreak of petroleum poisoning presented macro and microscopic lesions consistent with changes of malacia and edema in deep structures of the brain, as described in ruminants with PEM associated with sulfur intoxication. Major macroscopic changes included congestion of cerebral vessels, edema, and herniation of the cerebellum. The most observed microscopic lesions, among all assessed cases, were laminar and segmental neuronal necrosis at different regions of the brain, spongiosis, nuclear pyknosis, and red nucleus neurons. Cerebrospinal fluid analysis revealed nonspecific alterations, requiring its association with epidemiological, clinical, and pathological findings, as the results described here are similar to those reported in toxic diseases with neurological manifestations, such as botulism. ResumoO presente trabalho objetiva descrever os achados epidemiológicos, clínicos, laboratoriais e patológicos de casos de polioencefalomalácia (PEM) em ruminantes na região semiárida do Rio Grande do Norte. Sete ruminantes (cinco ovinos, um bovino e um caprino) com diagnóstico patológico de PEM foram incluídos. Quatro casos foram associados à deficiência de tiamina devido acidose láctica ruminal crônica por oferta de alimentação rica em carboidratos, destacando-se os concentrados, casca de soja triturada e melão. Três ruminantes de um surto de intoxicação por petróleo apresentaram lesões macro e microscópicas condizentes com alterações de malácia e edema em estruturas profundas do encéfalo, descritas em ruminantes com PEM associada à intoxicação por enxofre. As principais alterações macroscópicas incluíram congestão dos vasos cerebrais, edema e conificação do cerebelo. Em todos os casos avaliados, as lesões microscópicas mais observadas foram a necrose neuronal laminar e segmentar em diferentes regiões do encéfalo, espongiose, picnose nuclear e a presença de neurônios vermelhos. A análise do líquido cefalorraquidiano revelou alterações inespecíficas, sendo necessário sua associação aos achados epidemiológicos, clínicos e patológicos, pois os resultados aqui descritos são semelhantes aos relatados em doenças tóxicas com manifestações neurológicas, como o botulismo. Pesq. Vet. Bras. 34(8) Lesions caused by excessive reaction to foot and mouth vaccine were observed in 1,815 out of 5,000 cattle slaughtered in a slaughterhouse under federal meat inspection. Such lesions resulted in condemnation of the affect parts of the carcasses in accordance with the Brazilian Federal Meat Inspection Regulations. The affected cattle had been vaccinated on May 2012 and then slaughtered in September of the same year. They were allotted to two groups with 1,500 (Groups 1) and 315 cattle (Group 2). Post vaccination lesions were characterized as salient, firm variable sized nodules multifocally distributed in the musculature of the lateral neck. Pus oozed at the cut surface of these nodules. Samples of 28 cattle were submitted for histopathological examination; main findings were pyogranulomas with central variable-sized clear spaces surrounded by marked infiltrate of viable and dead neutrophils, which were surrounded by epithelioid macrophages with vacuolated cytoplasm and occasional multinucleated giant cell. This inflammatory reaction was walled by abundant connective tissue infiltrated by lymphocytes and plasma cells. Samples of the lesion of six cattle were submitted to bacteriological culture with negative results. When affected carcasses went through dressing, there was an average of 1.8 and 2.0 kg of muscle tissue cut off from the carcasses respectively of Group 1 and 2. Economic losses in the farm of origin of the affected cattle was R$ 20,424.00, considering the price paid by kg of cattle for slaughter at the time of the occurrence. This amount at that time was enough to buy 29.17 weaned calves for fattening. If one considers theses 5,000 cattle vaccinated on the farm as the population at risk, the morbidity rate would be 0.36%. These results indicate that the losses due to vaccine reaction, even with no overt clinical signs, may cause important economic losses.

This study evaluated the effects of supplementation with a combination of probiotic microorganisms, added at different concentrations to the mineral mixture, on the production of serum antibodies against rabies virus in cattle vaccinated with a single dose of rabies vaccine. Forty-two male Nellore cattle, aged 12 months, were randomly divided into three groups (n = 14): the control group (CG) received 70 grams of mineral mixture/animal/day; and the 2 gram probiotic group (G2P) and 8 gram probiotic group (G8P) received 70 grams of mineral mix/animal/day with 2 and 8 grams added, respectively, of a combination of probiotic microorganisms (Lactobacillus acidophilus, Streptococcus faecium, Bifidobacterium thermophilum and Bifidobacterium longum). Individual antibody titers were determined using a neutralization in cell-based rapid fluorescent focus inhibition test (RFFIT) technique. One-way analysis of variance (one-way ANOVA) was used with contrasts using the Tukey method to determine whether the experimental groups differed within each time point, and the paired t-test was used to determine whether differences occurred between time points within each group. The level of significance was set at 5%. There were statistically significant differences between the mean serum concentrations of the CG and G8P groups at 30 and 60 days after the first vaccination, and at 60 days, 100% of the animals maintained minimum titers of protective antibodies only in the G8P group. There was also improvement in the production of antibodies in the G2P group compared with the CG after 30 and 60 days, but this difference was not statistically significant. In conclusion, increasing doses of probiotic added to the mineral mix beneficially affected the rabies humoral immune response, as determined by serum antibodies, and enabled the maintenance of minimum protective titers for a longer period in previously vaccinated cattle. ResumoEsse estudo avaliou o efeito da suplementação de uma associação de microrganismos probióticos, adicionados à mistura mineral em diferentes doses, na produção de anticorpos séricos antirrábicos em bovinos primovacinados. Quarenta e dois bovinos Nelore machos, com idade de 12 meses, foram divididos em três grupos (n=14): grupo controle (GC) recebeu 70 gramas de mistura mineral/animal/dia; grupos probiótico 2 gramas (G2P) e 8 gramas (G8P) receberam 70 gramas de mistura mineral/animal/dia adicionados respectivamente de 2 e 8 gramas de probióticos (Lactobacillus acidophilus, Streptococcus faecium, Bifidobacterium thermophilum e Bifidobacterium longum). Os títulos individuais de anticorpos antirrábicos foram determinados por meio da técnica de soroneutralização em células baseado no rapid fluorescent focus inhibition test (RFFIT). Os resultados obtidos foram comparados pelo teste t não pareado, com 5% de nível de significância.
