Brucellosis is a zoonotic disease with common agent is Brucella abortus in bovine specie. The disease in bulls is usually asymptomatic and could have absence of anti-B. abortus antibody in serum. It was investigated of the occurrence of bovine brucellosis in bulls, which involved testing blood serum and seminal plasma from 177 bulls used for natural mating or as semen donors. Blood was collected from all the animals by venipuncture of the jugular vein to obtain the serum samples, while seminal plasma was obtained from the animals by electroejaculation. Samples were tested in a screening test (rose bengal test). In the second stage, the samples testing positive in the RBT were subjected to the confirmatory test (2-mercaptoethanol). None of the animals tested positive when blood serum was used. However, 5.06% of the bulls were considered reagent positive when seminal plasma was tested. It also sought to determine whether there was an association between the kind of production and the destination of animals with positive results. The calculated odds ratio indicated that bulls of the kind of production dairy/both (beef and dairy) had a higher risk of brucellosis infection. It is recommended that seminal plasma agglutination and seroagglutination techniques are applied concomitantly. Key words: Blood serum, males, natural mating, semen donor ResumoBrucelose é uma zoonose que nos bovinos tem como principal agente a bactéria Brucella abortus. A doença nos touros é geralmente assintomática e pode haver ausência de anticorpos anti-B. abortus no soro sanguíneo. O presente estudo teve como objetivo principal avaliar a ocorrência da brucelose bovina em reprodutores para isso foram empregados testes oficiais tendo como material o soro sanguíneo e o plasma seminal. Foram utilizados 177 machos bovinos destinados à monta natural ou doação de sêmen. O soro sanguíneo foi obtido através de venopunção da jugular e o plasma seminal através da eletroejaculação. As amostras foram submetidas ao teste de triagem (Antígeno acidificado tamponado) e na segunda etapa, apenas as amostras positivas na triagem foram submetidas ao exame confirmatório (2-mercaptoetanol). Não houveram animais positivos quando utilizado o soro sanguíneo. Para plasma seminal houve 5,06% de touros reagentes. Avaliou-se também se sistema produtivo ou a destinação do animal possuíam associação com resultados positivos. O cálculo de odds ratio demonstrou que os reprodutores bovinos do sistema produtivo misto/leite tem maior risco de infecção para brucelose. Recomenda-se que a técnica de sêmen plasma aglutinação seja feita em conjunto com a soroaglutinação. Palavras-chave: Soro sanguíneo, machos, monta natural, doador de sêmen

Control of brucellosis as a worldwide zoonotic disease is based on vaccination of animals and diagnosis of infected cases to be eradicated. Accurate and rapid detection of infected animals is of critical importance for preventing the spread of disease. Current detection of brucellosis is based on whole-cell antigens and investigating serum antibodies against Brucella lipopolysaccharide (LPS). The critical disadvantage is misdiagnosis of vaccinated animals as infected ones and also cross-reactions with other Gramnegative bacteria. Recombinant outer membrane protein 2b (Omp2b) of Brucella abortus was evaluated as a novel serodiagnostic target in comparison to conventional tests which are based on LPS. Recombinant Omp2b (rOmp2b) was expressed in Escherichia coli BL21 and purified by Ni 2+-based chromatography. rOmp2b was evaluated in an indirect enzyme-linked immunosorbent assay (ELISA) system for diagnosis of brucellosis, with sera from Brucella-infected mice along with negative sera and sera from mice which were inoculated with other Gram-negative species for assurance of specificity. Thereafter, cattle sera collected from different regions were assessed along with known negative and known positive serum samples. We found that Omp2b can discriminate between Brucella-infected animals and non-infected ones. Results for assessment of two hundred and fifty cattle sera by Omp2b-based indirect ELISA which were compared to Rose Bengal plate agglutination test (RBPT) and serum tube agglutination test (SAT) showed that our proposed procedure has the sensitivity of 88.5%, specificity of 100%, and accuracy of 90.8%. We suggest that recombinant Omp2b could be used as a protein antigen for diagnosis of brucellosis in domestic animals and can be evaluated for detection of human brucellosis.

RESUMOO objetivo do presente trabalho foi determinar a frequência de propriedades positivas (focos) e de fêmeas bovinas soropositivas para a brucelose no Estado de Roraima, bem como identificar fatores de risco. Foram utilizados dados da Agência de Defesa Agropecuária do Estado de Roraima (ADERR), coletados de suas quatro microrregiões, durante o período de janeiro de 2007 a julho de 2009. Durante esse período, foram examinadas 387 propriedades, e 9.087 soros de fêmeas bovinas com idade ≥ 24 meses foram submetidos ao diagnóstico sorológico da brucelose. Para a análise de fatores de risco, foi utilizada a regressão logística múltipla. Uma propriedade foi considerada foco quando apresentou pelo menos um animal soropositivo. Das propriedades investigadas, 106 (27,4%) apresentaram pelo menos um animal soropositivo e, dos animais analisados, 369 (4,1%) foram soropositivos. Os fatores de risco identificados foram: possuir mais de 51 animais no rebanho (odds ratio = 1,87; p = 0,015) e alugar pasto (odds ratio = 2,20; p = 0,003). Sugere-se que esforços sejam concentrados na intensificação da vacinação de bezerras no Estado de Roraima com o intuito de que seja alcançada uma prevalência compatível com as ações de teste e sacrifício de animais. Data from the Agency of Agricultural Protection in the state, collected from its four microregions during the January 2007 to July 2009 period, were used. During this period, 387 herds were examined, and 9,087 adult bovine female sera were submitted to the serological diagnosis of brucellosis. For risk factor analysis the multiple logistic regression was used. A herd was considered focus when presented at least one seropositive animal. Of the herds investigated, 106 (27.4%) had at least one seropositive animal, and of the animals examined, 369 (4.1%) were seropositive. Risk factors identified were: herd size larger than 51 animals (odds ratio = 1.87; p = 0.015) and pasture rental (odds ratio = 2.20; p = 0.003). It is suggested that efforts must be concentrated on the intensification of heifer vaccination in the State of Roraima in order to reach a prevalence value compatible with actions of test and sacrifice of animals.
