Analysis of the nucleotide sequence of the coat protein and 3'-untranslated region of two Brazilian Potato virus Y isolates

Abstract: Two Brazilian Potato virus Y (PVY) isolates were biologically characterized as necrotic (PVY-NBR) and common (PVY-OBR) based upon symptoms on test plants. Additional characterization was performed by sequencing a cDNA corresponding to the 3' terminal region of the viral genome. The sequence consisted of 195 nucleotides (nt) coding part of the nuclear inclusion body b (NIb) gene, 804 nt of the coat protein (CP) gene, and 328 nt (PVY-OBR) or 326 nt (PVY-NBR) of the 3'-untranslated region (UTR). Translation of th… Show more

“…Therefore, classification of potato isolates of PVY based on the 3 0 -UTR implies that this region cannot be used to demarcate strains of the isolates. Furthermore, it seems the unique 3 0 -UTR sequence of PVY is not related to the tuber or tobacco leaf necrosis symptoms, as also indicated by others [7,25]. One would expect there to be some level of correlation between the serology assays and CP sequences of PVY since the MAbs bind to the CP, however, as we have demonstrated in this paper, there is no direct correlation.…”

“…However, the composition of the recombinant strains in Brazil has not been studied on a systematic basis and the contribution of mutations and new recombination events occurring in Brazil has not been determined. Among the Brazilian isolates reported to date, there are some with strong similarities to PVY O and PVY N strains and some recombinants similar to PVY NTN (Inoue‐Nagata et al. , 2001; Sawazaki et al.…”
