Antibacterial activity of endophytic fungi isolated from Croton lechleri (Euphorbiaceae)

Croton lechleri is a native species from the Amazon and used with relative frequency in folk medicine in Brazil and other countries. Diversity and antibacterial activity of endophytic fungi associated with this plant were studied here. Samples of leaves and stems were used and 575 endophytic fungi were isolated (307 from leaves and 268 from stems), comprising 284 morphotypes distributed in 13 genera and unknown. The most frequently isolated genera were Phomopsis (30.78%), Penicillium (21.57%) and Pestalotiopsis (16.70%). Diversity and richness of species were higher in leaf tissues. Fifty-five fungi presented antibacterial activity. The fungi with the highest activity were Phomopsis (6.34%), Penicillium (3.17%), and those unknown (5.28%). Penicillium sp. 9 showed the highest antibacterial activity against Enterococcus faecalis and Phomopsis sp. 8 and Phomopsis sp. 9 against Streptococcus pneumoniae and Staphylococcus aureus. Curvularia sp. 1 and a fungus that could not be identified (Unknown sp. 9), showed the highest antibacterial activity against Klebsiella pneumoniae and Escherichia coli, respectively. Only two fungi (Penicillium sp. 9 and Curvularia sp. 1) inhibited the five tested bacteria. Endophytic fungi of C. lechleri harbor a great diversity of endophytic fungi, which have the potential for producing antibacterial compounds.

Os microrganismos que vivem no interior de plantas e habitam de modo geral suas partes aéreas, como folhas e caules, mas não causam aparentemente nenhum dano a seus hospedeiros, são chamados de endofíticos. Além de exercerem diversas funções de importância para o hospedeiro, esses microrganismos são potencialmente úteis na agricultura e na indústria, sobretudo na farmacêutica e de defensivos agrícolas. Por se constituírem em potenciais substitutos de produtos químicos, ao exercerem ações de biocontrole e/ou promoção de crescimento de plantas, favorecendo a preservação do ambiente, vem sendo apontados como alternativa viável para sistemas de produção agrícola ecológica e economicamente sustentáveis. A obtenção de substâncias de interesse econômico, como enzimas, antibióticos e outros fármacos, a partir de microrganismos endofíticos, tem sido frequentemente relatada na literatura científica. Exemplo disso é o taxol, poderoso anticancerígeno que anteriormente era obtido apenas da exploração da planta Taxus brevifolia e, agora pode ser obtido de diferentes gêneros de fungos endofíticos. Avanços como este reforçam a grande potencialidade biotecnológica de tais microrganismos. Este trabalho visa apresentar um panorama sobre potenciais aplicações de microrganismos endofíticos na agricultura e na produção de substâncias de interesse econômico. AbstractEndophytic microorganisms are mainly fungi and bacteria that live inside plants, generally inhabiting aerial parts such as leaves and stems, without causing any apparent damage to their hosts. In addition to many important functions for the host, the endophytic microorganisms are potentially useful in agriculture and industry, especially in pharmaceuticals and agrochemicals. By forming themselves into substitutes for chemicals by exercising stock biocontrol and/or promotion of plant growth, favoring the environmental preservation, has been appointed as a viable alternative agricultural production systems for environmentally and economically sustainable. Obtaining substances of economic interest, such as enzymes, antibiotics and other drugs from endophytic microorganisms has often been reported in the scientific literature. One example is taxol, a powerful anti-cancer substance that was previously obtained only from the exploitation of the plant Taxus brevifolia, and can now be obtained from different genera of endophytic fungi. Advances like this reinforce the great biotechnological potential of such microorganisms. This study presents an overview of potential applications of endophytic microorganisms in agriculture and production of substances of economic interest.

The paper describes the isolation of ergosterol peroxide, monocerin, annularin I and annularin J compounds of Exserohilum rostratum biomass extracts. Compounds were isolated by silica gel column chromatography in polarity gradient and the structures were determined by nuclear magnetic resonance (NMR) and mass spectrometry (MS) methods. Monocerin, annularin I and annularin J compounds were tested against Escherichia coli, Pseudomonas aeruginosa and Bacillus subtilis bacteria and showed moderate activity. Annularin I and annularin J compounds are new natural products. Keywords: annularins, polyketides, E. rostratum, endophytic fungi IntroductionEndophytic microorganisms live in plants internal tissues without causing apparent disease to their hosts. 1 Endophyte microbial colonization can be demonstrated by isolating plant tissues disinfected through histological cut and by DNA amplification technique. 2 In tropical and temperate forests (which cover only 1.5% of the terrestrial surface), there are approximately 300,000 (three hundred thousand) plant species analyzed so far, homing more than 60% of the world's biodiversity. 3 Extreme diversity allows for the opportunity to discover new microorganisms that may be potential bioactive compounds producers for agriculture, industry and medicine. 4 There is a great interest in the study of endophytic microorganisms, including their application in pest biocontrol. Fungi are good secondary metabolite producers, and many have the same biological activity, 5-8 especially polyketides. For example, the polyketide citrinin isolated from Penicillium janthinellum completely inhibited the growth of Leishmania braziliensis.9 Some fungi species of Penicillium and Pestalotiopsis genera produce high diversity of polyketides with antimicrobial activity. 10,11In another study with a variety of endophytes isolated from Vigueira arenaria and Tithonia diversifolia fungal extracts, important antiparasitic and antimicrobial activities were observed in fungal extracts. 12 In the Nodulisporium sp. (Xylariaceae) fungus extract, which was isolated from the woody plant species Erica arborea (Ericaceae), nodulisporina D, E and F compounds were obtained, having antifungal, antibacterial and algaecide activity.13 From the endophytic fungus Ampelomyces sp., isolated from the medicinal plant Urospermum picroides (Asteraceae), 6-O-methylalaternina and altersolanol A compounds were isolated, which showed antimicrobial activity against Gram-positive pathogens Staphylococcus aureus, S. epidermidis and Enteorococcus faecalis.14 Brazil is estimated to be home of about 20% of the world's biodiversity and is considered the country in the world with the largest number of endemic species.15 About 270,000 plant species on this planet are estimated to harbor about 13.6 million unique endophytic fungal species, in addition to approximately 100,000 fungi species already known to date.
