Avaliação das variáveis analíticas do método de Folin-Ciocalteu para determinação do teor de taninos totais utilizando como modelo o extrato aquoso de folhas de psidium guajava L.

Abstract: Psidium guajava L. LEAVES AQUEOUS EXTRACT AS A MODEL. The main analytical variables of a modified Folin-Ciocalteu method were studied by UV-Vis and gradient HPLC-PDA methods, using purified (PC) and technical grade (TGC) casein. Rutin and an aqueous extract of Psidium guajava L. leaves were used as models. The best results were ascribed to TGC. Certainly PC bonds the polyphenols of the P. guajava extract better than TGC, but TGC afforded better precision. A lack of specificity occurred when rutin was analyzed … Show more

“…5,11,14,15,25 In a former work we asked whether casein and hidepowder were able to bind P. guajava tannins in the presence of flavonoids in a specific way. 8 This question can be extended to PVPP because some of the main features of the casein-tannin and hide-powder-tannin binding are also noticeable in PVPP cases. Alike these protein substrates, the influence of hydrogen bonds and hydrophobic interactions are also manifested in the PVPP molecule, namely, in the pyrrolidone moiety and in the vinyl chain, respectively.…”

“…6,7 It was recently demonstrated that methods using hide-powder and casein are non-specific when flavonoids are also present in the reaction milieu. 5,8 The lack of specificity was also related to other methods intended for the tannin assay, including gravimetric and HPLC-UV methods. 9,10 The capacity of insoluble cross-linked povidone (PVPP) to bind polyphenols arises in this context as a seldom explored analytical alternative.…”

“…The concentration of total phenols was determined colorimetrically (absorbance at 760 nm) using the Folin-Ciocalteu method (Verza et al 2007). The proanthocyanidin content was determined according to the method of Price et al (1980).The results were expressed in milligram catechin equivalents per gram of dry matter.…”

“…The total phenolic content was determined according to the European Pharmacopoeia spectrophotometric method (European Pharmacopoeia, 2007;Verza et al, 2007). Stock solution was prepared from lyophilized CE (0.15 g) or EAF (0.03 g) of powered sample mixed with 250 ml of water.…”
