Neste trabalho nós apresentamos uma revisão sobre a síntese em fase sólida de metabólitos secundários, tais como alcalóides, poliaminas, esteróides, terpenos e flavonóides, descritos na literatura a partir de 2000. Um número considerável de rotas sintéticas elegantes e desafiadoras, usando suportes sólidos, será discutido.In this manuscript, we disclose solid-phase organic syntheses (SPOS) of small-molecules of some secondary metabolites, such as alkaloids, polyamines, steroids, terpenes, and flavonoids, described in the literature since 2000. A number of elegant, efficient and challenging syntheses on solid support will be presented.

A Química Combinatória (QC) é atualmente uma das mais promissoras ferramentas para a descoberta e o desenvolvimento INTRODUÇÃOApesar do crescente desenvolvimento e dos evidentes avanços científico e tecnológico da indústria farmacêu-tica, existem ainda inúmeras doenças que necessitam de novos fármacos, com melhor perfil terapêutico para o seu tratamento, como por exemplo SIDA, Alzheimer, câncer, malária, entre outras (Wermuth, 1996). Assim, a busca por novos agentes terapêuticos ainda se faz necessária nos tempos de hoje.Historicamente, a pesquisa para a descoberta de novas substâncias biologicamente ativas vem seguindo processo seqüencial, em que a síntese dos compostos ocorre de maneira artesanal, isto é, uma molécula de cada vez, escolhendo-as a partir de substâncias de origem natural ou sintética e, depois de exaustivas etapas de isolamento, purificação e caracterização, o candidato a protótipo de fármaco é submetido a testes para detecção de eventuais atividades farmacológicas (Gedeck, Willett, 2001). Este processo clás-sico é amplamente empregado em laboratórios de pesquisa de instituições de ensino e empresas farmacêuticas, mas apresenta como desvantagem o fato de ser relativamente lento e gerar reduzido número de derivados diferentes.Atualmente, o planejamento racional de fármacos fundamenta-se basicamente no conhecimento da estrutura molecular dos receptores, o que possibilita elaborar substâncias com perfis farmacológicos mais definidos. Para tanto, as hipóteses são elaboradas fundamentando-se no conhecimento das propriedades físico-químicas dos compostos ativos e seus respectivos sítios moleculares de ação (Folkers, Kubinyi, 1997). Entretanto, como muitas vezes a estrutura dos receptores não é conhecida admite-se que a gênese de moléculas potencialmente bioativas seja realizada sem levar em conta sua natureza (Villar, Koehler, 2000).

Over the past years, polymer-supported reagents have been extensively studied and used in various applications. One class of such reagents is called "scavengers," which can be used to easily eliminate compounds in a solution. The present work describes the production of a resin that can be used for scavenging ketones and aldehydes using low-cost reagents and simple reaction steps, named here Amb15-Iso. This resin is obtained by reacting a low-cost commercial sulfonyl resin, Amberlyst-15, with isoniazid, a drug used for the treatment of tuberculosis. Acetone and isobutyraldehyde were used as carbonyl compound models. The reactions were monitored in-line by ATR-FTIR and results showed that the polarity of the solvent influences the kinetics of the production of the resin and water proved to be the fastest solvent. For the scavenging of acetone and isobutyraldehyde, two factors showed to have an impact in the amount of compounds captured: the polarity of the solvent and the solubility of water in the solvent. The capacity of scavenging acetone in water varied from 0.11 to 0.28 mmol per gram of resin, depending on the initial acetone concentration. The equilibrium of this reaction was modeled and the equilibrium constant was calculated to be 0.63 6 0.07 L mol 21 . The resin was also recycled and tested in a second round of scavenging and results showed that there was not much difference between the new resin and the recycled one, proving that the Amberlyst-15 could be reused for a second cycle of scavenging.
