Neste trabalho nós apresentamos uma revisão sobre a síntese em fase sólida de metabólitos secundários, tais como alcalóides, poliaminas, esteróides, terpenos e flavonóides, descritos na literatura a partir de 2000. Um número considerável de rotas sintéticas elegantes e desafiadoras, usando suportes sólidos, será discutido.In this manuscript, we disclose solid-phase organic syntheses (SPOS) of small-molecules of some secondary metabolites, such as alkaloids, polyamines, steroids, terpenes, and flavonoids, described in the literature since 2000. A number of elegant, efficient and challenging syntheses on solid support will be presented.

The erratic absorption of PG15 observed after oral dosing could explain the lack of anti-inflammatory dose dependency.

A fast, sensitive, and specific liquid chromatography-tandem mass spectrometry (LC-MS=MS) method was developed and validated for the simultaneous determination of quinine and doxycycline in pharmaceutical formulations. The LC-MS=MS method was carried out on a Sun Fire Waters C 18 column (50 mm  3.0 mm I.D.) and the mobile phase consisted of acetonitrile:0.1% formic acid (75:25, v=v), run at a flow rate of 0.45 mL=min (split 1:3). The injection volume was 10 mL for both standard and samples. The triple quadrupole mass spectrometer equipped with an electrospray source in positive mode (ESþ) was set up in multiple reaction monitoring mode (MRM), monitoring the transitions of 325.0 > 307.0 and 445.0 > 428.1, for quinine and doxycycline, respectively. The total analysis time was 2 min and the method was linear in the concentration range of 10-1500 ng=mL for both compounds. Method validation investigated parameters such as the specificity, linearity, precision, accuracy, and robustness, giving results within the acceptable range. Moreover, the proposed method was successfully applied for determination of quinine and doxycycline in nanocapsule formulations to support the quality control.

Um método rápido e simples de cromatografia líquida com detecção por ultravioleta foi desenvolvido e validado para quantificação, em plasma de rato, do composto (5Z,E)-3-[2-(4-clorofenil)-2-oxoetil]-5-(1H-indol-3-ilmetileno)-tiazolidina-2,4-diona (PG15). Uma coluna em fase reversa C18 foi utilizada para separação do analito, seguida por detecção em UV a 385 nm. O método utilizou a precipitação do PG15 a partir do plasma e eluição isocrática com metanol:água (90:10, v/v). O tempo total da corrida foi 7,5 min. O método proposto foi validado e mostrou-se linear entre 62,5 e 4000 ng mL -1 . A precisão intra-e inter-dia, expressa como desvio padrão relativo, foi menor que 15 e 10%, respectivamente, para todas as concentrações investigadas. A exatidão, medida através dos controles da qualidade, ficou entre 86,1-114,9 %. A aplicabilidade do método validado foi testada no estudo farmacocinético pré-clínico do PG15.A simple and rapid liquid chromatography-ultraviolet detection (LC-UV) method has been developed and validated for quantifying (5Z,E)-3-[2-(4-chlorophenyl)-2-oxoethyl]-5-(1H-indol-3-ylmethylene)-thiazolidine-2,4-dione (PG15) in rat plasma. A C18 reversed phase column provided chromatographic separation of the analyte which was followed by UV detection at 385 nm. The method involves precipitation of PG15 from plasma and isocratic elution with methanol:water (90:10, v/v). Total elution time was 7.5 min. The proposed method was validated and showed linear correlation in the range of 62.5 to 4000 ng mL -1 . The within-and between-day precision, expressed as the relative standard error, were found to be less than 15 and 10 %, respectively, for all the concentrations investigated. The accuracy, measured using the quality control samples, was in the range of 86.1-114.9 %. The applicability of the validated method was tested in a pre-clinical pharmacokinetic study of the thiazolidinone PG15.

1. The present work investigated the pharmacokinetic and tissue distribution as well as acute toxicity of a new chemical entity (NCE), the anticancer candidate LaSOM 65 in Wistar rats. 2. LaSOM 65 pharmacokinetics was investigated after intravenous (i.v., 1 mg/kg) and oral (p.o., 10 and 30 mg/kg) dosing. Tissue distribution was assessed after i.v. bolus dose. Acute toxicity was evaluated after i.v. (1, 2.5 and 5 mg/kg) and p.o. (50, 100 and 150 mg/kg) administration. 3. Short half-life (1.75 ± 0.71 h), a clearance of 0.85 ± 0.18 L/h/kg and a volume of distribution of 1.76 ± 0.24 L/kg were observed after i.v. dosing. The compound showed good bioavailability and linear pharmacokinetics after oral doses. The NCE distributes consistently in lung and fatty tissues, with penetration ratios of 2.7 and 1.4, respectively. The other tissues investigated presented smaller penetration ratios. Adverse clinical symptoms were observed only after i.v. administration, and regressed 3 h after dosing. Compared with controls, no statistical differences were found for serum analysis, body weight and relative organ weight, indicating no acute toxicological effects. 4. Overall, LaSOM 65 showed good pharmacokinetic characteristics and no signs of acute toxicity, indicating that it is a promising anticancer candidate.
