Hypocotyl, cotyledon and zygotic embryo explants from two Tunisian Cucumis melo L. cultivars Beji and Maazoun, cultured on the MS medium added with 2,4-D (0.25-1 mg l )1 ) and BA (0.10-0.50 mg l )1 ), produce calluses with somatic embryos after 3 weeks of culture. For Beji c.v. the highest percentage (62.50%) of embryogenesis was observed for cotyledons. The average embryo number per callus was 10.40. Embryogenesis induction for zygotic embryos reached 33.50% with 29 embryos per callus. The embryogenesis ability of hypocotyls did not exceed 12.50% (2.50 embryos per callus). Somatic embryogenesis for Maazoun c.v. explants was less efficient. Embryos formation was observed only for cotyledons (29%) and zygotic embryos (25%). Cotyledonary staged embryos, when transferred to hormone free MS medium, germinated. The maximum germination rates were 51.50 and 44.50%, respectively for Maazoun and Beji c.v. The highest percentage (36.50%) of survival plants was noted for Beji c.v. Regenerants were diploids (2n = 2x = 24) and morphologically similar to their parents issued from seeds.

The aim of this study was to evaluate the in vitro morphogenic potential of genipap (Genipa americana L.) zygotic embryos. Seeds obtained from ripe fruits had their zygotic embryos excised and inoculated in MS medium with 4.44µM of 6-benzylaminopurine (BAP) and supplemented with 0.0; 1.07; 2.14 and 3.21µM of naphthalene acetic acid (NAA). The potential of explants regeneration and the shoot length and number of leaves in plantlets were evaluated. The in vitro regeneration of genipap is possible from the conversion of zygotic embryos in a MS medium with 4.44µM BAP supplemented with 3.21µM NAA.

An efficient procedure has been developed for inducing somatic embryogenesis and regeneration of plants from tissue cultures of oil palm (Elaeis guineensis Jacq.). Thin transverse sections (thin cell layer explants) of different position in the shoot apex and leaf sheath of oil palm were cultivated in Murashige and Skoog (MS) (Physiol Plant 15:473-497, 1962) medium supplemented with 0-450 µM picloram and 2,4-D with 3.0% sucrose, 500 mg L −1 glutamine, and 0.3 gL −1 activated charcoal and gelled with 2.5 gL −1 Phytagel. Embryogenic calluses were evaluated 12 wk after inoculation. Picloram (450 µM) was effective in inducing embryogenic calluses in 41.5% of the basal explants. Embryogenic calluses were maintained on a maturation medium composed of basal media, plus 0.6 µM NAA and 12.30 µM 2iP, 0.3 gL −1 activated charcoal, and 500 mg L −1 glutamine, with subcultures at 4-wk intervals. Somatic embryos were converted to plants on MS medium with macro-and micronutrients at half-strength, 2% sucrose, and 1.0 gL −1 activated charcoal and gelled with 2.5 gL −1 Phytagel.
