OBJECTIVE: To assess microbicide function and macrophage viability after in vitro cellular infection by methicillin-sensitive/resistant Staphylococcus aureus in nourished rats and rats subjected to neonatal malnutrition. METHODS: Male Wistar rats (n=40) were divided in two groups: Nourished (rats suckled by dams consuming a 17% casein diet) and Malnourished (rats suckled by dams consuming an 8% casein diet). Macrophages were recovered after tracheotomy, by bronchoalveolar lavage. After mononuclear cell isolation, four systems were established: negative control composed exclusively of phagocytes; positive control composed of macrophages plus lipopolysaccharide; and two testing systems, macrophages plus methicillin-sensitive Staphylococcus aureus and macrophages plus methicillin-resistant Staphylococcus aureus. The plates were incubated in a humid atmosphere at 37 degrees Celsius containing 5% CO2 for 24 hours. After this period tests the microbicidal response, cytokine production, and cell viability were analyzed. The statistical analysis consisted of analysis of variance (p<0.05). RESULTS: Malnutrition reduced weight gain, rate of phagocytosis, production of superoxide anion and nitric oxide, and macrophage viability. Production of nitrite and interleukin 18, and viability of macrophages infected with methicillin-resistant Staphylococcus aureus were lower. CONCLUSION: The neonatal malnutrition model compromised phagocyte function and reduced microbicidal response and cell viability. Interaction between malnutrition and the methicillin-resistant strain decreased the production of inflammatory mediators by effector cells of the immune response, which may compromise the immune system's defense ability.

ObjetivoVerificar o efeito da administração intraperitoneal de glutamina, prévia a uma situação de estresse, sobre o número de leucócitos do sangue e sobre a taxa de fagocitose de macrófagos alveolares de ratos. MétodosRatos machos Wistar aos 90 dias de idade foram divididos em controle (C, n=15), estresse (C+E, n=16), glutamina (GLN, n=15) e glutamina e estresse (GLN+E, n=15) e mantidos em condições padronizadas. Os grupos GLN e GLN+E receberam, via intraperitoneal (v ip), L-glutamina (0,2g/kg de peso corporal) durante 10 dias. Seus pares receberam, v ip, L-glicina (0,4g/kg do peso corporal). Os animais C+E e GLN+E foram submetidos ao estresse agudo de contenção (40min, após 24h da última intervenção com o aminoácido). Leucócitos totais foram contados em hemocitômetro e os diferenciais por esfregaço sanguíneo. Para avaliação da taxa de fagocitose, os macrófagos alveolares (1x10 6 em meio de cultura RPMI) e fungos S. cerevisiae (1x10 7 ) foram incubados em estufa (37ºC, 5%CO 2 ) durante 1 hora. Foram utilizados os testes estatísticos análise de variância (ANOVA) e o teste de Tukey, valores expressos em média do percentual e desvio-padrão.

Biocompatibility is the ability of a material to perform specific tasks when applied to living tissues without causing damage or injuries to it. Thus, this study aimed at determining the in vivo and in vitro biocompatibility of Cissus sicyoides L. -Vitaceae hydroalcoholic extracts. A total of 30 rats (Rattus norvegicus Albinus Wistar), with ages ranging from 45 to 90 days and weighing between 170 and 260g were used. The animals were divided into 3 groups (A1, A2 and A3) with 6 animals each, for the in vivo test, which were sacrificed after 2, 4 and 6 days, respectively. For in vitro test, 12 animals were used to obtain the index of adherence and phagocytic ability of macrophages of rats from the control and the experimental groups. In results found for the in vivo test, it was concluded that the extract was biocompatible, whereas no significant changes were observed in the tissue. As to the in vitro test, the extract was not biocompatible, since the pure extract showed a low rate of adherence (7.1) and a high rate of phagocytosis (35.7), indicating a significant difference when compared to the control group. However, when diluted, the extract was shown to be harmless, due to an increase in the values of the adherence index at the following concentrations : 1/10 (61.4) and 1/100 (74.3) in the tests, which showed no significant differences when compared to the control group. After analyzing the data, it was concluded that since the infusion of the plant is a kind of dilution, its use does not cause any harm to the body. A new study is necessary at the moment to possibily demonstrate its effects on the long term.
