Prostaglandin A1 Inhibits Replication of Classical Swine Fever Virus

Abstract: Prostaglandins (Pgs) have been shown to inhibit the replication of several DNA and RNA viruses. Here we report the effect of prostaglandin (PgA1) on the multiplication of a positive strand RNA virus, Classical Swine Fever Virus (CSFV) in PK15 cells. PgA1 was found to inhibit the multiplication of CSFV. At a concentration of 5 µg/ml, which was nontoxic to the cells, PgA1 inhibitis virus production in 99%. In PgA1 treated cells the size and number of characteristic Classical Swine Fever focus decreased in amount

“…Chemical analysis of these PGs indicated that the presence of a carbonyl group at the unsaturated (α, β) cyclopentane ring might be responsible for this property 21,22 . PGs inhibit production of MAYV 23 and other viruses, including Sendai 24 , Vesicular stomatitis virus (VSV) 25,26 , human immunodeficiency virus (HIV) 27 , Rotavirus 28 and Classical swine fever virus 29 .…”

“…Prostaglandin A 1 (PGA 1 ) is capable of blocking the replication of a wide variety of RNA and DNA viruses, including CSFV. 9 , 10 , 11 Despite the importance of BVDV for animal health, the effect of PGA 1 on BVDV replication has not been examined. Therefore, we evaluate the effect of PGA 1 on the replication of cpBVDV in MDBK cells, focusing on changes in BVDV-induced vacuolization.…”

“…Although prostaglandins (PGs) are capable of inhibiting the replication of a variety of RNA and DNA viruses, their mode of action is not entirely clear. 9 , 10 , 11 In some systems, PGAs exert potent antiviral activity by inducing the synthesis of proteins from the heat shock protein 70 (HSP70) family, through cycloheximide-sensitive activation of a heat-shock trigger factor. 13 …”

“…MG132 [119], a proteasome inhibitor, could attenuate the CSFV replication. (9) Other molecules also have anti-CSFV effects, such as Prostaglandin A1 [120,121], the phage-displayed E2-binding peptides [122], ceramide (C6) (activator of the protein phosphatase 1 pathway) [123], and quercetin (inhibitor of HSP70 function) [88]. More and more anti-CSFV drug candidates will be discovered with the increase of antiviral research.…”
