Polyvinyl alcohol-glutaraldehyde as solid-phase in Elisa for Schistosomiasis

Abstract: Soluble adult Schistosoma mansoni antigen preparation (SWAP) was covalently fixed onto polyvinyl alcohol-glutaraldehyde discs and an enzyme linked-immunosorbent assay (ELISA) was set up. The best conditions for the assay were established and it was found that small amount of antigen such as 1.5 micrograms was required. A comparison between this procedure and the conventional ELISA was proceeded. A reliable method of antigen immobilization was achieved and the low prices of the employed reagents are economicall… Show more

“…In our laboratories, several supports have been proposed for biomolecules immobilization: Dacron [42,43]; ferromagnetic Dacron [44,45]; polyvinyl alcohol-glutaraldehyde network [46][47][48]; polyaniline-Dacron composite [49]; plasticized filter paper with polyvinyl alcohol-glutaraldehyde [50].…”

“…In our laboratories other procedures for antigen covalent immobilization on supports have been proposed such as Dacron (11), ferromagnetic Dacron (12,13), interpenetrated network of PVA-glutaraldehyde (14)(15)(16)(17), filter paper plasticized with PVA-glutaraldehyde (18), and a polyaniline-Dacron composite (19). The optimal values of F1 antigen amount, conjugate dilution and serum titer established for the present method (1.25 µg/disc; 1:4,000 and 3,200, respectively) can be compared to those found for PVA-glutaraldehyde (1.3 µg/disc; 1:6,000 and 1:12,600), plasticized filter paper (1.25 µg/disc; 1:12,000 and 1:6,400), and polyaniline-Dacron (2 µg/disc; 1:4,000 and 1:800).…”

“…29 Solid phase immunoassay supports have been cast from glutaraldehyde crosslinked PVOH, illustrating that antibodies can be immobilised effectively onto glutaraldehyde treated PVOH. 30 Here we investigated for the first time the sensitive quantitation of clinically relevant protein biomarkers in FEP microcapillaries with covalently immobilised capture antibodies, which involved carrying out sandwich ELISA in MCF devices with antibodies immobilised covalently using a variety of crosslinking chemistries. To produce a suitable surface for covalent antibody immobilisation to the unreactive and hydrophobic FEP microcapillaries, the inner surface of the microcapillaries was firstly coated with a layer of PVOH.…”
