Oogram studies in mice infected with Schistosoma mansoni and treated with dexamethasone

Abstract: Mice infected with about 90 cercariae of Schistosoma mansoni (LE strain) were treated during five consecutive days with dexamethasone (50 mg/Kg, subcutaneously), starting on the 42th day of infection. Groups of five mice were then daily sacrificed from the first day after onset of treatment until the first day after. The perfusion of the portal system was performed and a piece of the intestine was processed for qualitative and quantitative oograms. This treatment carries to larger numbers of eggs in the tissue… Show more

“…Qualitative and quantitative oogram evaluation showed that arctiin did not affect oviposition kinetics or egg maturation. Similar effects on oogram were described with treatment of infected animals with dexamethasone (50 mg/kg) [34]. In addition, Aires et al [35] observed eggs in all development phases following administration of β-lapachone, however the percentage of immature eggs was significant lower, indicating a kinetic change in the egg development.…”

“…9 , 21, 44, 45 Our data are similar to results from other studies showing that glucocorticoids dampen the immune response to helminthic infection. 36,[46][47][48][49] We show that Dexa increased the time of adult female parasites remaining in the gastrointestinal tract, and the worms were fertile with release of eggs that were quantified in the feces. Dexa also led to increase in the burden of infective larvae recovered from the lungs of infected mice.…”

“…For the evaluation of quantitative oograms, the formula 'number of eggs/g of tissue ¼ number of eggs in intestinal fragment/weight of fragment in mg £ 1000' was employed (Cançado et al, 1965;Katz et al, 1966;Katz & Pellegrino, 1974;Hermeto et al, 1994). Additionally, in order to verify the number of eggs per female parasite in a day, the ratio of second-stage S. mansoni eggs per g intestine/number of female parasites recovered from the portal system was utilized (Pacheco et al, 2005(Pacheco et al, , 2008Mati et al, 2010).…”
