In Vitro Organogenesis from Root Explants of Passiflora miniata Mast., an Amazonian Species with Ornamental Potential

Abstract: range 2-9 µM) of 6-benzyladenine (BA); thidiazuron (TDZ) or kinetin (KIN). Plant growth regulators were not added to the control treatment. Root explants have showed a high regenerative potential. After 30 days of in vitro culture, the root explants showed several shoots formed direct and indirectly. TDZ provided the best response in the differentiation adventitious shoots, mainly in the presence of 6.8 µM. The cytokinins BA and KIN responded producing a reduced number of shoots. After 120 days, rooted regener… Show more

“…MS medium supplemented with 1.0 mg/l TDZ was the best treatment, producing the highest percentage of organogenesis formation of 76% and the highest mean number of shoots/explant of 17. This result is in harmony with that obtained by De Carvalho et al [53] who found that TDZ-induced direct organogenesis from leaf explants of Passiflora miniata.…”

“…It was reported that the maximum response on medium supplemented with 1.0 mg/l BA and 0.5 mg/l TDZ was observed, producing an average of 18.2 buds per culture after 24 weeks from date palm culture [ 19 ]. TDZ at 2.0 mg/l also could achieve shoot sprouting directly on leaf tissues of Urginea altissima [ 24 ], at 1.5 mg/l for Passiflora miniata [ 25 ] and at 1.0 mg/l for Aloe vera [ 26 ] and Arnebia euchroma [ 27 ].…”

“…In all three cultivars, TDZ alone or in combination with BA was found to be superior for direct organogenesis, so a new TDZ-BA combination was tested. TDZ at 2.0 mg/l could induce shoot in Urginea altissima leaf tissues (Baskaran et al, 2018), 1.5 mg/l in Passiflora miniata (Carvalho et al, 2019), and 1.0 mg/l in Aloe vera leaf tissues (Lavakumaran and Seran, 2014). TDZ at 0.5 and 5.0 µM was found to be optimum for inducing an average of 4-5 shoots per cotyledonary node in 93% of the cultures and 55 somatic embryos in 68% of the cultures by Chhabra et al (2008).…”
