Cytotoxicity Profile of Endodontic Sealers Provided by 3D Cell Culture Experimental Model

Abstract: The aim of the present study was to evaluate the cytotoxic effects of five endodontic sealers (AH Plus, Endomethasone N, EndoSequence BC, MTA Fillapex and Pulp Canal Sealer EWT) using a three-dimensional (3D) cell culture model. A conventional bi-dimensional (2D) cell culture model was used as reference technique for comparison. Balb/c 3T3 fibroblasts were cultured in conventional bi-dimensional cell culture and in rat-tail collagen type I three-dimensional cell culture models. Then, both cell cultures were in… Show more

“…8 The unfavorable results of MTA Fillapex in cell culture are reported even when compared to that of resin epoxy-based material such as AH Plus. 30 The results are observed in traditional two-dimensional (2D) and three-dimensional (3D) systems, 31 and are corroborated by results observed in our in vitro evaluation as follows: a greater reduction was observed in the cellular metabolism by MTA Fillapex when compared to that of the Control or AH Plus while using less-diluted extracts. The AH Plus exhibits less cytotoxicity with increases in dilution, and this is in agreement with the results of a previous study, which indicated that a greater dilution of AH Plus after 24 h caused minor cytotoxic effects toward L929 cells.…”

“…In this study, the cell viability induced by MTA-HP was lower than that of its precursor in some dilutions during the initial periods; however, it had a better result compared to white MTA-Ang when the former was more diluted. It should be noted that in vitro studies represent the first level of research and are important for understanding biological risks before using these materials in vivo (16). However, tests with cell cultures have limitations, especially when using two-dimensional cell cultures, as in this study (8,16).…”

“…It was created combining a material with excellent biocompatibility and bioactive potential such as MTA with synthetic resins with good physical properties. However, this blend has shown cytotoxicity [4,5]. The Brasseler Industry (Savannah, GA, USA) introduced the EndoSequence Root Repair Material; a premixed bioceramic material indicates to pulp capping/pulpotomy, perforation repair, apexification, and root-end filling.…”
