a-Keto-6-amino-valerianslure (I) bzw. a-Keto-e-amino-capronslure (11) werden durch Oxydation des im alkalischen Gelatinehydrolysat vorhandenen Prolins und Ornithins bzw. Lysins mit Wasserstoffperoxyd in Gegenwart von Cu2@ dargestellt und in Form kristalliner Sake sowohl mit Sauren als auch mit Metallen isoliert. Nur die Slure I wurde in freiem Zustand gewonnen. Nach den Analysen und den IR-Spektren basitzen Hydrochlorid, Hydrobromid und freie Ketosaura I offenkettige, das von ihr abgeleitete Kupfer(I1)-und Natriumsalz Pyrrolin-Struktur. Die Sake der Saure I1 weisen hingegen ausnahmslos eine cyclische Struktur auf, die bei Hydrohalogeniden dem Al-Piperidein, bei Metallsalzen dem A2-Piperidein entspricht. Die a-Keto-6-amino-valeriansaure ( I a) und die a-Keto-c-amino-capronsaure (I1 a) sind Intermediiirprodukte im Stoffwechsel der Amiiosiiurepaare Ornithin und Prolin bzw. Lysin und Pipecolinsiiure. In wll3rigen Msungen treten sie im Gleichgewicht mit k e n inneren Schiffschen Basen auf, d. h. mit der A'-Pyrrolin-und A1-Piperidein-carbonSiiure-(Z) (Ib, IIb), die katalytisch sowie auch enzymatisch m Prolin bzw. Rpecolinsa;Ure reduziert werden konnenl. 2). l a I b I1 a IIb Die beiden Aminoketoduren wurden zwn ersten Ma1 durch Desaminierung von 0 -N -Carbobenzoxy-L-ornithin und -blysin mittels einer Schlangengift-Oxydase als Hydrobromide erhalten 1). Splter wurden durch Hydrolyse synthetischer a-Keto-w-phthalimido-carbonsauren kristalline Hydrochloride gewonnen W, von denen das I entsprechende offenkettige Struktur besaI3, wahrend I1 der A1-Piperidein-carbon&iure-(2) entsprachs). Das Hydrochlorid I wurde neuerdings durch Oxalesterkondensation des PyrroIidons-(2) in zwei Stufen bereitets).

A chemical synthesis closely following a biochemical pathway discussed by several authors is described Die Entstehung mono-und bicyclischer Pyrrolidin-und Piperidinalkaloide aus den basischen Aminosauren Ornithin und Lysin ist durch Precursorversuche weitgehend gesichert 1). Die im folgenden mitgeteilte Synthese der moglichen Alkaloidvorstufen 1.5-Didehydro-norhygrin (5) und I .6-Dldehydro-isopelletierin (6) lehnt sich eng an einen von mehreren Autoren 2 4) fur die Tropanalkaloide vorgeschlagenen Riosyntheseweg an.
Al-Piperidein-carbonsaure-(6) (111) wird durch Oxydation von 6-Hydroxy-aamino-capronsaure (I) mit Chromsaure erhalten. Sie geht im physiologischen pH-Bereich eine reversible, temperaturabhangige Dimerisierung zur Tetrahydroanabasindicarbonsaure VI ein. -Al-Piperidein-carbonsaure-(6) entsteht bei der enzymatischen Oxydation von Hexahomoserin durch c-Hydroxy-a-aminosauredehydrogenase aus Neurospora crassa und von Lysin mit pflanzlicher Diaminoxydase.a-Acetaminoadipinslure-6semkaldehyd (Derivat des Hydrolyseprodukts von 111) bildet sich durch enzymatische Transaminierung von Na-Acetyl-L-lysin.A1-Pyrrolin-und A1-Piperideincarbonsauren sind Intermediarprodukte des Aminosaurestoffwechsels. A~-Pyrrolin-carbonsaure-(5) (Pyr-5) 1) und der mit dieser Verbindung im Gleichgewicht stehende Glutarninsaure-y-semialdehyd nehmen eine zentrale Stellung in den gegenseitigen biologischen Umwandlungen von Glutaminsaure, Ornithin und Prolin ein: Pyr-5 entsteht durch Reduktion aus Glutaminsaure, durch Transaminierung aus Ornithin und durch Dehydrierung aus Prolin2).Die Bedeutung der A1-Piperideincarbonsauren ist weitgehend ungeklart. Vielleicht spielen Al-Piperidein-carbonsPure-(6) (Pip-6) und ihre nichtcyclische Form, der a-Amino-adipinsaure-6-semialdehyd, eine der Pyr-5 entsprechende Rolle im Stoffwechsel von a-Amino-adipinsaure, Lysin und Pipecolinsaure. Hierfiir sprechen verschiedene Befunde.w-Hydroxy-a-aminosauren werden durch ein DPN-abhbgiges Enzym aus Neurospora crnssa dehydriert3). Als Dehydrierungsprodukt von c-Hydroxy-a-amino-capronsaure ist Pip-6 zu erwarten.a-Amino-adipinsaure kann offensichtlich eine Vorstufe der Biosynthese des Lysins sein, denn im Nahrstoffbedarf bestimmter Lysin-bediirftiger Mutanten von Neurospora crussn kann 1) Abkiirzungen: a-AA = a-Amino-adipinsaure, DPN@ = Diphosphopyridinnucleotid, DPNH = reduzierte Form von DPN@, Pip-2 = AI-Piperidein-carbonsaure-(2), Pip-6 = A]-Piperidein-carbonsaure-(6), (Pip-6)~ = dimere Pip-6, Pyr-2 = Al-Pyrrolin-carbonsaure-(2), Pyr-5 = A1-Pyrrolin-carbonsiure-(5).
