Zusammenfassung: Eine Aminosäure: 2-Oxosäure-Aminotransferase aus Lupinus angustifolius wird 21fach angereichert und charakterisiert. Ornithin und Lysin sind Substrate dieses Enzyms. Diese beiden Aminosäuren werden von den Transaminasen aus Lupinus angustifolius und Phaseolus aureus ROXB. im Sinne einer -Desaminierung umgesetzt. Lysin wird von Transaminasen aus Pseudomonas fluorescens und Flavobacterium flavescens dagegen in -Stellung desaminiert. In etiolierten Erbsenkeimlingen lassen sich Decarboxylasen für Ornithin und Lysin nachweisen. Sie fuhrten y-Phthalimido-butterslure uber eine groRere Zahl von Zwischenstufen in 8-Phthalimido-a-keto-valeriansaure Uber, aus der das Hydrochlorid von I durch Hydrolyse gewonnen werden konnte.
Al-Piperidein-carbonsaure-(6) (111) wird durch Oxydation von 6-Hydroxy-aamino-capronsaure (I) mit Chromsaure erhalten. Sie geht im physiologischen pH-Bereich eine reversible, temperaturabhangige Dimerisierung zur Tetrahydroanabasindicarbonsaure VI ein. -Al-Piperidein-carbonsaure-(6) entsteht bei der enzymatischen Oxydation von Hexahomoserin durch c-Hydroxy-a-aminosauredehydrogenase aus Neurospora crassa und von Lysin mit pflanzlicher Diaminoxydase.a-Acetaminoadipinslure-6semkaldehyd (Derivat des Hydrolyseprodukts von 111) bildet sich durch enzymatische Transaminierung von Na-Acetyl-L-lysin.A1-Pyrrolin-und A1-Piperideincarbonsauren sind Intermediarprodukte des Aminosaurestoffwechsels. A~-Pyrrolin-carbonsaure-(5) (Pyr-5) 1) und der mit dieser Verbindung im Gleichgewicht stehende Glutarninsaure-y-semialdehyd nehmen eine zentrale Stellung in den gegenseitigen biologischen Umwandlungen von Glutaminsaure, Ornithin und Prolin ein: Pyr-5 entsteht durch Reduktion aus Glutaminsaure, durch Transaminierung aus Ornithin und durch Dehydrierung aus Prolin2).Die Bedeutung der A1-Piperideincarbonsauren ist weitgehend ungeklart. Vielleicht spielen Al-Piperidein-carbonsPure-(6) (Pip-6) und ihre nichtcyclische Form, der a-Amino-adipinsaure-6-semialdehyd, eine der Pyr-5 entsprechende Rolle im Stoffwechsel von a-Amino-adipinsaure, Lysin und Pipecolinsaure. Hierfiir sprechen verschiedene Befunde.w-Hydroxy-a-aminosauren werden durch ein DPN-abhbgiges Enzym aus Neurospora crnssa dehydriert3). Als Dehydrierungsprodukt von c-Hydroxy-a-amino-capronsaure ist Pip-6 zu erwarten.a-Amino-adipinsaure kann offensichtlich eine Vorstufe der Biosynthese des Lysins sein, denn im Nahrstoffbedarf bestimmter Lysin-bediirftiger Mutanten von Neurospora crussn kann 1) Abkiirzungen: a-AA = a-Amino-adipinsaure, DPN@ = Diphosphopyridinnucleotid, DPNH = reduzierte Form von DPN@, Pip-2 = AI-Piperidein-carbonsaure-(2), Pip-6 = A]-Piperidein-carbonsaure-(6), (Pip-6)~ = dimere Pip-6, Pyr-2 = Al-Pyrrolin-carbonsaure-(2), Pyr-5 = A1-Pyrrolin-carbonsiure-(5).
