ЦЕЛЬ: оценить распространенность полиморфизмов С677Т, А1298С гена метилентетрагидрофолатре- дуктазы (MTHFR) у пациентов с перенесенным инфарктом миокарда (ИМ) и сахарным диабетом (СД) 2 типа. МАТЕРИАЛЫ И МЕТОДЫ: для решения поставленной цели обследовано 77 пациентов, сформировано 2 группы. Группа 1 (n=39) – пациенты с перенесенным ИМ в сочетании с СД 2 типа. Группа 2 (n=38) – паци- енты с перенесенным ИМ без СД 2 типа. Средний возраст исследуемых пациентов составил 59 лет. В иссле- дование не включались пациенты с наличием СД 1 типа, декомпенсации СД 2 типа, печёночной и почечной недостаточности, заболеваний щитовидной железы с нарушением функции, а также с наличием тяжёлых сопутствующих соматических и инфекционных заболеваний в стадии декомпенсации патологического процесса. Определение полиморфизмов С677Т, А1298С гена MTHFR осуществляли с помощью метода полимераз- ной цепной реакции с детекцией результатов в режиме реального времени с применением набора реа- гентов «Литех», Россия. Выделение геномной ДНК проводилось набором реагентов «ДНК-экспресс-кровь» («Литех», Россия). РЕЗУЛЬТАТЫ: по результатам молекулярно-генетического исследования пациентов 1 группы по по- лиморфизму С677Т гена MTHFR получены следующие результаты: генотип СС выявлен у 11 пациентов, генотипы СТ и ТТ – у 14 пациентов. Частота встречаемости аллеля С составила 46,2%, аллеля Т – 53,8%. По результатам генотипирования пациентов 1 группы по полиморфизму А1298С гена MTHFR генотип АА выявлен у 15 пациентов, генотип АС – у 14, СС – у 10 пациентов. Частота встречаемости аллеля А составила 56,4%, аллеля С – 43,6% Гомозиготный генотип СС полиморфизма С677Т гена MTHFR в группе 2 выявлен у 20 пациентов, гете- розигота СТ – у 15 пациентов. Гомозиготный генотип ТТ обнаружен у 3 пациентов. Аллель С встречалась в 72,4%, аллель Т – в 27,6%. В группе 2 генотип АА полиморфизма А1298С гена MTHFR обнаружен у 15 паци- ентов, гетерозигота АС – у 14, СС – у 9 пациентов. Частотное распределение аллелей составило: А-аллель встречалась в 57,9%, аллель С – в 42,1%. ВЫВОДЫ: изучено распределение генотипов и аллелей полиморфизмов С677Т, А1298С гена MTHFR у исследуемых пациентов. Получены достоверные различия по генотипам СС и ТТ, а также аллелям С и Т полиморфизма С677Т гена MTHFR между пациентами 1 и 2 групп. Достоверных различий по распределе- нию генотипов и аллелей полиморфизма А1298С гена MTHFR между исследуемыми пациентами получено не было.
ЦЕЛЬ: изучить частоту встречаемости генотипов и аллелей полиморфного маркера Lys198Asn гена эндотелина-1, определить уровень эндотелина-1 у пациентов с наличием хронической ишемической болезни сердца в сочетании и без сочетания с сахарным диабетом 2 типа.МАТЕРИАЛЫ И МЕТОДЫ: обследовано 135 пациентов, которые были разделены на 2 группы. Группа 1пациенты с наличием хронической ишемической болезни сердца (ИБС) и сахарного диабета (СД) 2 типа (n=65). Группа 2 -пациенты с хронической ИБС без СД 2 типа (n=70). Набор пациентов был осуществлен на базе кардиологического отделения учреждения здравоохранения «Гродненский областной клинический кардиологический центр».Определение полиморфизма Lys198Asn гена эндотелина-1 (EDN1) осуществляли с помощью метода полимеразной цепной реакции с детекцией результатов в режиме реального времени с применением набора реагентов производства «Литех», РФ. Выделение геномной ДНК человека проводилось набором реагентов «ДНК-экспресс-кровь». Амплификацию ДНК проводили на амплификаторе Rotor Gene-Q («Qiagen», Германия).Определение уровня эндотелина-1 в сыворотке крови выполняли методом иммуноферментного анализа с использованием наборов «Human EDN1» (Biomedica, Австрия) на анализаторе Sunrise TECAN.Статистический анализ проводился с помощью непараметрических методов в программе Statistica 10.0. Различия считались статистически значимыми при р<0,05.
