Резюме. Чума была причиной трех пандемий и привела к гибели миллионов людей. Чума -типичный зоо-ноз, и ее возбудитель -Yersinia pestis, циркулирует в популяциях диких грызунов, обитающих в природных очагах чумы на всех материках, кроме Австралии. Передача чумы осуществляется укусами блох. Циркуля-ция Y. pestis в природных очагах чумы обеспечивается целым рядом факторов патогенности. В обзоре рас-сматривается один из них -активатор плазминогена Pla. Этот белок является одним из представителей омп-тинов -семейства протеаз наружных мембран патогенных энтеробактерий, обеспечивающих колонизацию отдельных органов и даже генерализацию инфекции в результате успешного противостояния врожденному иммунитету хозяина. Описаны история открытия, генетический контроль, условия биосинтеза, выделение, очистка и физико-химические свойства активатора плазминогена. Высокоочищенные препараты активатора плазминогена утрачивают свою ферментативную активность, а ренатурация в присутствии липоолигосаха-рида Y. pestis восстанавливает энзиматические свойства Pla. Этот фактор патогенности отсутствует у наибо-лее древней филогенетической группы чумного микроба, bv. caucasica, а предшественник остальных групп Y. pestis subsp. microtus получил в результате горизонтального переноса изоформу Pla, близкую по свойствам омптинам менее вирулентных энтеробактерий. Затем в ходе микроэволюции была отобрана «классическая» изоформа Pla с повышенной протеолитической активностью, характерная для всех высоковирулентных для человека штаммов Y. pestis subsp. pestis. «Классическая» изоформа Pla Y. pestis функционально подобна актива-торам плазминогена млекопитающих, превращающих плазминоген в плазмин путем ограниченного протео-лиза. Протеазу Pla, активирующую плазминоген, а также деградирующую основной ингибитор плазмина -α2-антиплазмин и, соответственно, определяющую способность чумного микроба лизировать фибриновые сгустки, препятствующие его распространению после укуса инфицированными блохами или подкожного заражения, принято рассматривать в качестве основного фактора Y. pestis, обеспечивающего генерализацию инфекционного процесса. Pla-опосредованная способность Y. pestis избирательно связываться с внеклеточ-ным матриксом и базальными мембранами может способствовать последующему гидролизу этих структур плазмином хозяина и преодолению патогеном тканевых барьеров. Активатор плазминогена Y. pestis также ги-дролизует C3 компонент комплемента, человеческий антимикробный пептид -кателицидин LL-37 и такие цитокины как фактор некроза опухолей α, интерферон γ, интерлейкин 8 и протеин 1 хемотаксиса моноцитов. Основной эндогенный ингибитор инициации свертывания крови TFPI, также высокочувствителен к про-теолитическому действию Pla, причем эффективность инактивации TFPI гораздо выше, чем эффективность активации плазминогена. В обзоре обсуждается возможность использования Pla в качестве молекулярной мишени для профилактики и лечения чумы.
The causative agent of plague, Yersinia pestis, is a highly virulent bacterial pathogen and a potential bioweapon. Depending on the route of infection, two prevalent forms of the disease – bubonic and pneumonic, are known. The latter is featured by a high fatality rate. Mortality in untreated bubonic plague patients reaches up to 40-60%, whereas untreated pneumonic plague is always lethal. The development of the infectious process in susceptible host is accounted for by a whole set of pathogenicity factors in plaque pathogen displaying various functional modalities being expressed depending on stage of infectious process, providing their coordinated expression. Knocking out any of such factors, in turn, may not either affect microbe virulence or lead to its attenuation. A search for new Yersinia pestis pathogenicity factors and subsequent development of highly effective subunit and live attenuated plague vaccines inducing development of pronounced cellular and humoral immune reactions, and/or assessment of their potential use as molecular targets for plague therapy still remain a pressing issue, as both currently licensed plague vaccines do not meet the WHO requirements, whereas strains of plague microbe isolated in Madagascar are resistant to all drugs recommended for plague antibacterial therapy. Here we summarize an impact of described and newly discovered pathogenicity factors into the virulence of Y. pestis strains and their protective anti-plague activity. An effect of loss of genes encoding regulatory proteins as well as mutations in the genes for various transport systems of Y. pestis on attenuation of virulent strains is described as well. Perspectives for introducing characterized antigens into prototype subunit vaccine as well as some other obtained mutants into prototypes of living attenuating vaccines were assessed. The use of antibiotics for plague treatment has been embraced by the World Health Organization Expert Committee on Plague as the ‘gold standard’ treatment. However, concerns regarding development of antibiotic-resistant Y. pestis strains accounted for further exploring alternatives to plaque therapy. Several research groups continue working on seeking for other alternative approaches, e.g. treatment with inhibitors of pathogenicity factors. Preliminary data attempting to treat plague patients with pathogenicity factor inhibitors are summarized. Аnti-virulence drugs targeting key microbial factors represent new promising therapeutic options in fight against antibiotic-resistant bacteria.
Резюме. Чума -зоонозное заболевание, вызванное грамотрицательной бактерией Yersinia pestis, которая, как правило, передается человеку от зараженных грызунов через укус инфицированной блохи. Данный микро-организм убил больше людей, чем все войны в истории человечества. Циркуляция Y. pestis в природных оча-гах чумы обеспечивается целым рядом факторов патогенности различной функциональной направленности. В обзоре рассматривается один из них -капсульный антиген чумного микроба (F1 или Caf1). Описаны исто-рия открытия, генетический контроль, условия биосинтеза, выделение, очистка и физико-химические свой-ства капсульного антигена, а также обсуждается его вклад в патогенез чумы и его использование в качестве основного компонента чумных вакцин. Визуально аморфная при световой микроскопии капсула Y. pestis при электронной микроскопии высокого разрешения представляет собой структуру, сформированную из отдель-ных фимбриеподобных тяжей длиной до 200 нм, расходящихся в разные стороны от поверхности бактерии. При температуре 37°С образуется в 800-1000 раз больше капсульного антигена, чем при 28°С. Гены, кодирую-щие белок Caf1 с молекулярной массой субъединицы 17,6 kDa, состоящей из 170 аминокислот, локализованы в caf1 опероне на плазмиде pFra. На основе анализа первичной нуклеотидной последовательности caf1 оперо-на было установлено его филогенетическое родство с кластерами генов, кодирующих секретируемые с помо-щью шаперон/ашерных систем пилевые адгезины не только других энтеробактерий, но и еще шесть адгезинов чумного микроба. Размножение клеток Y. pestis внутри макрофагов является обязательным этапом патогенеза чумы, а вирулентность чумного микроба коррелирует не с устойчивостью к захвату фагоцитами, а со способ-ностью выживать и размножаться в фаголизосомах фагоцитарных клеток за счет подавления антибактери-альных функций фагоцитов. Капсула, образованная из агрегатов Caf1, защищает клетки Y. pestis от захвата интактными фагоцитами хозяина, препятствует инициации альтернативного пути активации комплемента. Молекулярный ашер Caf1A, ответственный за закрепление капсульного антигена на поверхнос ти бактери-альной клетки, имеет высокий аффинитет к человеческому интерлейкину 1β. Caf1 может вступать в конку-ренцию с интерлейкинами 1α, 1β и 1ra за связывание с рецепторами на лимфоидных клетках, препятствуя развитию адекватного иммунного ответа. Иммунодиагностика чумы построена на выявлении Caf1 или анти-Caf1-антител, так как этот антиген Y. pestis видоспецифичен. Покрывающий поверхность микробных клеток капсульный антиген -важнейший составной элемент всех современных чумных вакцин. Показана его ве-дущая роль в создании напряженного иммунитета у белых мышей, крыс, приматов и человека. Однако в ор-ганизме иммунного животного могут накапливаться бескапсульные (Caf1 -) варианты Y. pestis, сохранившие вирулентность на уровне исходных штаммов «дикого типа».
