Lysosomotropic composition of dialdehyde dextran and amphotericin B had a greater therapeutic effect in mice with systemic candidiasis compared to free amphotericin B. This composition normalized glucocorticoid function of the adrenal glands and decreased the severity of liver destruction at late terms of granulomatous inflammation.

Эффективным подходом к исследованию ревматоидного артрита (РА) является создание и использование моделей животных. Экспериментальное изучение различных аспектов формирова-ния и течения РА, включая ранний РА, позволяют приблизиться к пониманию этиопатогенетических механизмов заболевания человека. В обзоре рассматриваются известные в настоящее время модели заболевания. Использование этих моделей для тестирования потенциальных терапевтических пре-паратов поможет прогнозировать их эффективность. Abstract. The establishment and application of animal models represent effective tools for research in rheumatoid arthritis (RA) pathogenesis. Animal models that replicate various mechanisms reflecting all aspects of RA, including early RA pathology, have provided important insights into studying etiology and pathogenetic mechanisms of RA in humans. This review article was compiled in order to give an introduction to the current state of RA models. Application of these experimental disorders for testing potential therapeutic approaches will help to make better predictions for drug efficiency in human RA. VARV-CrmB is a TNF binding protein of variola virus. VARV-CrmB protein was previously shown to be active as a TNF-antagonist in a number of in vivo and in vitro models. Here we investigated the epicutaneous effect of recombinant VARV-CrmB protein using an experimental model of muTNFinduced migration of skin leukocytes as well as colony forming activity of bone marrow cells (BMC). Epiсutaneous applications of muTNF enhanced the number of cells migrating from skin flaps of BALB/c mice, whereas subsequent applications of VARV-CrmB protein in 30 min after muTNF, abolished that effect. Epicutaneously applied muTNF influenced the activity of committed hematopoietic progenitors causing a reduction of erythroid (BFUe+CFUe) colonies and increase of granulocyte-macrophage (CFU-GM) colonies in the colony-forming tests. VARV-CrmB, applied in combination with muTNF, demonstrated an ability to reverse this effect, namely, to increase BFUe+CFUe and reduce CFU-GM back to the control levels. Taking together, these data demonstrate the TNF-blocking properties of VARV-CrmB in vivo at epicutaneous applications. As effective TNF antagonist VARV-CrmB protein might be conceded as a beneficial candidate for future research and development of therapeutic approaches in the field of inflammatory skin diseases.
Резюме. В настоящем обзоре собственных экспериментальных данныъх приведены результаты изучения in vivo и in vitro эффектов TNF-связывающего белка вируса натуральной оспы (VARV-CrmB) на TNF-зависимые изменения функциональной активности мышиных и человеческих клеток. In vitro VARV-CrmB отменяет вызываемую TNF повышенную продукцию IL-1β и IL-6 мононуклеарными клетками человека и их повышенную окислительно-метаболическую активность. Рекомбинантный белок VARV-CrmB восстанавливает TNF-индуцированное снижение количества эритроидных колоний и оказывает корригирующие эффекты на колониеобразующую активность ГМ-предшественников мыши и человека. In vivo белок VARV-CrmB оказывает выраженный лечебный эффект на проявления LPS-индуцированного эндотоксического шока у SPF мышей Balb/C, достоверно увеличивая процент выживших животных. Белок VARV-CrmB уменьшает выраженность коллаген-индуцированного артрита на ранних сроках его формирования. Аппликативное воздействие белка VARV-CrmB нормализует TNF-индуцированное повышение миграционной и окислительно-метаболической активности лейкоцитов мышей и оказывает корригирующие эффекты на колониеобразующую активность гемопоэтических предшественников. Аппликативное воздействие VARV-CrmB снижает миграцию лейкоцитов из кожного лоскута в афферентной фазе контактной реакции на ДНХБ и показатель «отека уха» в эфферентной фазе контактной реакции на ДНХБ. Полученные результаты демонстрируют TNF-блокирующие свойства белкаVARV-CrmB. Совокупность полученных нами данных позволяет рассматривать рекомбинантный вирусный белок VARV-CrmB как новый потенциальный TNF-антагонист, эффекты которого могут быть реализованы за счет его нейтрализующей способности TNF-индуцированной активации окислительно-метаболической, цитокин-продуцирующей и миграционной функций эффекторных клеток при терапии патологических воспалительных процессов.
