The aim of the work was to study the primary bioenergetic mechanisms of hypoxia formation in the myocardial tissue of experimental animals depending on the differentiated physical characteristics of vibration (frequency and duration) and their combination. The study of the functional states of native mitochondria in the composition of the tissue homogenate was carried out using the polarographic method and a galvanic-type closed-oxygen sensor in a 1-ml thermostatic cuvette in a salt incubation medium. The metabolic states of the mitochondria of the myocardium of experimental animals were modeled in vitro during the oxidation of endogenous substrates (before and after the administration of inhibitors of different links of the respiratory chain), with varying exogenous energy substrates (before and after the introduction of 2,4-DNP into the cell). In order to ensure synchronism of measurements in a short time, an incomplete cycle of metabolic states “endogenous respiration → rest → activity” was used. The results of multiple comparisons of variations in kinetic parameters revealed a reliable but multidirectional effect of the frequency of vibration on the rate of oxidation of substrates of the mitochondria of the heart of rabbits in different metabolic states. A change in the duration of exposure to vibration showed an increase in the oxidation rate of endogenous substrates and succinic acid at rest to 21–56 sessions by 17% and 24. 4%, respectively, while the oxidation rate of glutamate decreased to 56 sessions by 24. 5%. Comparison of the general variability of kinetic parameters with a combination of frequency and duration of vibration at different levels of variation showed that it was the interaction of factors that made the most important and significant contribution to the intergroup variability of oxidation rates of endogenous and exogenous substrates, identifying signs of the formation of bioenergetic hypoxia and allowing analysis of the primary physical transformation phenomena in the biological effect.
РезюмеЦелью работы явилось экспериментальное изучение активности системы энергопродукции ткани печени и почки кролика при неблагоприятном действии раз-личных режимов общей вибрации. Изучение энергозави-симых реакций нативных митохондрий печени и почки проводили полярографическим методом с помощью за-крытого мембранного электрода типа Кларка. Найде-но, что системы энергопродукции печени и почки вовле-каются в ответную реакцию организма на воздействие общей вибрации. Изменения функциональной активно-сти митохондрий паренхиматозных органов зависят от исследованных режимов вибрации (частоты и дли-тельности) и проявляются нарушением баланса между функциональной активностью ФАД-и НАД-зависимых звеньев дыхательной цепи. Повышение частоты и дли-тельности вибрации ведет к развитию низкоэнергети-ческого сдвига с активацией системы окисления янтар-ной кислоты и торможением НАД-зависимого звена дыхательной цепи митохондрий и сопровождается признаками дистрофии тканей. ВВЕДЕНИЕСреди техногенных производственных факторов одним из наиболее распространенных и значимых является вибрация, действующая как хронический стрессор, вызывающий напряжение адаптационно-компенсаторных систем организма и формирование вибрационной болезни [22, 51]. На вибрационное воздействие откликаются практически все гомео-статические системы организма, их ответная реак-ция зависит от энергии колебания [69, 58]. Действие вибрации на биологические структуры приводит к нарушению структурной организации тканей, что можно подтвердить определением значимых тка-невых биомаркеров [23, 74]. При этом максимально негативным эффектом обладает частота вибрации, резонансная к частоте данной ткани или органа, так как материальной основой резонанса в биологиче-ских объектах являются масса и ее упругие свойства. Диапазон резонансных частот для тканей и органов теплокровных животных и человека находится в диа-пазоне от 1 до 200 Гц [67, 70, 72].Вибрация порождает гидродинамические силы, вызывающие колебания центрального и перифери-ческого внутрисосудистого давления и изменяю-щие кровенаполнение паренхиматозных органов, периферический крово-и лимфоотток, значитель-ные изменения ультраструктуры клеток мышечного слоя артерий, деградацию регуляции их тонуса. Ге-нерализованная эндотелиальная дисфункция, усу-губляемая активизацией прооксидантной системы [57], ведет к микро-и макроангиопатиям, наруше-нию гемодинамики и микроциркуляции и заверша-ется формированием синдрома регенераторно-пластической недостаточности (висцеропатии) на уровне паренхиматозных органов. The review presents an analysis of literature on the study of nervous system changes in experimental animals exposed to vibration. The hypoxic type of cellular metabolism against the background of vibration is the result of vascular mechanoreceptors stimulation and spasm of blood vessels as well as intravascular pressure phase fluctuations and impaired blood and lymph outflow. Hemodynamic disorders and microangiopathy in the central nervous system can reach the level of cerebral vascular impairment and capillary-trophic insufficiency of the brain. Changes in calcium homeostasis at the cellular and tissue levels is a key mechanism of neuronal destruction occurring alongside with impaired blood supply to the nervous tissue in patients with vibrational disease. Long-term exposure to vibration results in reduced tissue respiration in the brain structures of experimental animals, that is most pronounced in the cortex and leads to a pathological change of spontaneous electrical activity of brain structures. Acute vibration stress stimulated the synthesis and excretion of serotonin not only in the hippocampus and hypothalamus but also in the cerebellum, which explains the disturbances of the oculomotor reactions observed in vibration exposure in the control system of the vertical vestibular ocular reflex. A decrease in the total number of neurons as well as an increase in the number of astroglia cells against the background of paravascular tissue edema were revealed against the background of changes in neurotransmitters levels. An increase in the concentration of the biomarker of structural and functional damage to brain tissue specific protein S-100B accompanies the development of professional sensorineural hearing loss and dysfunction of cerebral level vegetative regulation.

Нередки полимедикаментозные отравления, а также случаи преднамеренного самоотравления подростков. опасна не только токсикогенная фаза отравления различными химическими соединениями, но и ее отдаленные последствия. в качестве последствий острых отравлений у детей описаны симптомы цереброастении, минимальной мозговой дисфункции, сосудистые расстройства, нарушения терморегуляции, судорожный синдром, нарушения зрения, гепато-и нефропатии, дисфункция коры надпочечников. вышеизложенные факты свидетельствуют о необходимости совершенствования токсикологической помощи, как при проведении реанимационных мероприятий, так и в фазу отдаленных последствий, скорейшее устранение которых сохраняет возможность развития ребенка, соответственно возрасту. Изучение общих закономерностей химико-биологического взаимодействия токсиканта с органами и тканями животных в эксперименте позволяет выявлять адаптивные и компенсаторные механизмы, обеспечивающие устойчивость основных функций организма к повреждающему действию ксенобиотиков. Изучение молекулярных и клеточных механизмов действия экзогенных химических факторов различной природы определяет точки-мишени для воздействия фармакологических средств. Учитывая то, что обязательным патогенетическим компонентом процесса интоксикации является гипоксия, то в данном исследовании на модели отравления этиленгликолем в качестве защитных фармакологических средств были использованы прямые антигипоксанты 2-этилтиобензимидазола гидробромид (метапрот) и 5-этокси-2-этилтиобензимидазол (этомерзол). Цель исследования. оценить антигипоксический, антиоксидантный и актопротекторные эффекты метапрота и этомерзола в модели отравления этиленгликолем. Abstract.
