Настоящая работа посвящена изучению эффектов лейтрагина, аналога эндогенного гексапептида динорфина 1-6, на экспрессию провоспалительных цитокинов и интерферонов I типа в экспериментальной фатальной модели острого респираторного дистресс-синдрома (ОРДС) у мышей C57BL/6Y. Экспрессию интерлейкина-1β (IL-1β), интерлейкина-6 (IL-6), фактора некроза опухоли (TNF-α), интерферонов α (IFN-α) и β (IFN-β) в легких оценивали методом ПЦР в реальном времени. Индукция ОРДС α-галактозилцерамидом и липополисахаридом E. coli приводила к многократному повышению экспрессии цитокинов в легких. Введение лейтрагина в сочетанном режиме, внутримышечная инъекция плюс ингаляция, приводило к статистически значимому снижению уровней мРНК цитокинов в легких уже через 3 ч после начала введения. При этом средний уровень мРНК IL-6, ключевого цитокина в развитии тяжелого ОРДС, снижался в 4,7 раза (p<0,01) до значений, близких к наблюдаемым у интактных животных. С учетом особой роли повышенных концентраций IL-6 в развитии тяжелых форм COVID-19 использование лейтрагина для подавления «цитокинового шторма» может быть эффективным подходом к лечению коронавирусной инфекции.
This study aims to investigate effects of leutragin, an opioid peptide analogue of endogenous dynorphin 1-6, on animal survival in an experimental model of “cytokine storm” and fatal acute respiratory distress syndrome (ARDS) in C57Bl/6Y mice under different administration regimens. The aforementioned factors cause a severe course of COVID-19, which explains the current interest in seeking new treatments for ARDS. It was shown that both the prophylactic (before ARDS induction) and therapeutic (after ARDS induction) administration of leutragin in a combined mode — intramuscular injection plus inhalation leads to a statistically significant increase in the survival rate of animals. Compared to the control, leutragin significantly reduced the risk of death in animals with ARDS. The discovered prophylactic effect of leutragin deserves special attention due to its potential in preventing the onset of the disease and impeding the development of severe lung damage, thus reducing the risk of ARDS and fatal outcomes. Thus, the use of leutragin can be seen as a new effective approach to the treatment and prevention of respiratory diseases associated with a “cytokine storm” and ARDS, including the coronavirus infection COVID-19.
Одним из факторов развития заболеваний желудочно-кишечного тракта (ЖКТ), описанных в литературе, является приём нестероидных противовоспалительных средств (НПВС). Энтероколит является частым осложнением приема НПВС. Экспериментальное моделирование заболеваний ЖКТ, максимально приближенных по происхождению и механизму развития соответствующего заболевания у человека, является актуальной проблемой современной медицины. В работе представлены результаты сравнительного экспериментального биомоделирования НПВС-индуцированного энтероколита у лабораторных крыс. Установлено, что при внутрижелудочном введении ацетилсалициловой кислоты у животных выявлены лишь повреждения желудка. Введение декскетопрофена как пероральным способом, так и внутримышечным вызывает эрозивно-язвенные повреждения желудка и кишечника. Развитие повреждений не зависит от способа введения препарата. It was found that the intragastric administration of acetylsalicylic acid resulted mainly in stomach damage. The introduction of dexketoprofen -both orally and intramuscularly -causes erosive and ulcerative lesions of the stomach and intestines. The type of damage showed no correlation with the type of administration. For the fi rst time, the results of enterocolitis biomodelling using dexketoprofen are presented.
With the advent of endonuclease methods of genome editing, particularly CRISPR/Cas9, it has become possible to obtain genetically modified rabbits by microinjection of zygotes. These highly effective human disease models can be used for various purposes. The present review aims to consider modern achievements in the creation of rabbit biomodels of human diseases using the technologies of genetic editing. It is concluded that Russian laboratories should intensify research in the development of genetically modified rabbits that can be used for various biomedical studies and biomodelling.
This study was aimed at developing an experimental model of fatal acute lung injury and acute respiratory distress syndrome (ARDS) based on the intratracheal administration of bacterial lipopolysaccharide (LPS) in combination with muramylpeptide and Freund’s complete adjuvant to C57Bl/6Y mice sensitized with α-galactosylceramide. The developed model is characterized by diffuse alveolar damage to the lungs and high mortality rates, as well as by a multifold increase in the mRNA level of interleukin-6 in the lungs. The model can be used for assessing the efficacy of drug candidates in the treatment of acute lung injury and ARDS, including in COVID-19.
