For evaluation of effects of release-active antibodies to CD4 on cultured lymphocytes from human peripheral blood, we measured intracellular content of lck-kinase cell-based ELISA. In cells treated with release-active antibodies to CD4, the content of intracellular lck-kinase significantly (p<0.01) decreased in comparison with the control (purified water processed in a similar way). Phytohemagglutinin had no effect on the concentration of lck-kinase in cells. The decrease in the content of CD4-associated lck protein suggests that the preparation enhanced intracellular coupling of lck-kinase with T-cell receptor and potentiated T-cell immune response.

Москва, Российская Федерация РезюмеСиндром старческой астении -один из главных гериатрических синдромов, тесно связанный со старением человека. Патогенез старческой астении изучен недостаточно, однако значительный интерес вызывает роль воспалительного старения как ключевого механизма развития этого синдрома. В обзоре рассматривается вклад про-и противовоспалительных цитокинов, ассоциированных с системой врожденного иммунитета, а также ряда других факторов воспаления в развитие синдрома старческой астении и его основных фенотипических проявлений. Обсуждается неодназначная роль воспалительного старения в патогенезе старческой астении. С одной стороны, воспалительное старение является одним из ключевых признаков синдрома астении, однако непонятно, чем оно является -патогенетическим фактором развития старческой астении или следствием развития этого синдрома. С другой стороны, при старческой астении происходит нарушение врожденного иммунитета, выражающееся в снижении способности продуцировать про-и противовоспалительные цитокины под действием лигандов TLR, что является значимым предиктором смертности. Изменение экспрессии генов, кодирующих ACE2 и молекулы врожденного иммунитета (TLR7, ИФН-α, ИЛ-1β, ФНО), у пожилых пациентов с COVID-19 Российский геронтологический научно-клинический центр Федерального государственного автономного образовательного учреждения высшего образования «Российский национальный исследовательский медицинский университет имени Н.И. Инфекция, вызванная SARS-CoV-2, создала значительную нагрузку на систему здравоохранения стран по всему миру. В связи с этим в условиях продолжающейся циркуляции SARS-CoV-2 изучение возможных предикторов неблагоприятного течения и исхода заболевания, а также развития тромботических осложнений является актуальной и важной задачей. Цель исследования -изучение экспрессии генов, кодирующих АСЕ2, рецептор врожденного иммунитета TLR7, цитокины ИФН-α, ИЛ-1β и ФНО, в лейкоцитах периферической крови у пациентов пожилого возраста с COVID-19, а также ассоциации экспрессии указанных генов с риском развития тромботических осложнений, оцениваемых по уровню D-димера. Материал и методы. В исследование включено 50 пациентов пожилого возраста (65-77 лет) с подтвержденным диагнозом COVID-19, группа сравнения составила 15 человек (возраст 65-69 лет) без COVID-19. В качестве исследуемого материала использовали лей коцитарную массу, полученную из образцов периферической крови. Экспрессию генов оценивали методом полимеразной цепной реакции в реальном времени с обратной транскрипцией.Результаты. Показано значительное снижение экспрессии гена ACE2 в основной группе относительно группы сравнения. Оценка молекул врожденного иммунитета показала повышение экспрессии генов TLR7, IFNA, а также генов TNF и IL1B по отношению к группе сравнения. Особую задачу исследования представляло сравнение исследуемых показателей с учетом прокоагулянтной активности, оцениваемой по уровню D-димера.Показано, что высокий уровень D-димера (> 1 мг/л) у пациентов с COVID-19 ассоциирован с более низким уровнем экспрессии гена IFNA и повышением уровня экспрессии генов провоспалительных цитокинов TNF и IL1B по сравнению с подгруппой с низким уровнем D-димера (< 1 мг/л). В группе с низким уровнем D-димера экспрессия генов TNF и IL1B не отличалась от группы сравнения.Заключение. Повышение уровня экспрессии гена TLR7 в сочетании со снижением экспрессии гена IFNA в подгруппе с высоким уровнем D-димера может свидетельствовать о дисбалансе факторов врожденного иммунитета у пациентов с COVID-19. Выявленное повышение экспрессии генов провоспалительных цитокинов TNF и IL1B ассоциировано с высоким риском тромбообразования (оцениваемому по уровню D-димера) и может быть одним из маркеров неблагоприятного исхода COVID-19.
Неоднозначная роль факторов врожденного иммунитета в нарушении фертильной функции у мужчин при варикоцелеФедеральное государственное автономное образовательное учреждение высшего образования «Российский национальный исследовательский медицинский университет имени Н.И. Проблема мужского бесплодия, вызванного варикоцеле, особенно актуальна, поскольку данная патология наиболее распространена у мужчин. Частота ее встречаемости, по данным разных авторов, достигает 10-30 %, а возникающие на этом фоне отклонения параметров спермограммы от нормы наблюдаются более чем у 40 % больных. Изучение механизмов системы врожденного иммунитета, участвующих в развитии патологического процесса при варикоцеле, представляет собой актуальную задачу и привлекает внимание все большего числа ученых в мире.Цель исследования -изучение роли факторов врожденного иммунитета при мужском бесплодии, вызванном варикоцеле.Материал и методы. В исследование были включены 60 доноров эякулята. Основную группу составили 27 пациентов с диагнозом «варикоцеле». Группу сравнения составили 33 здоровых донора.Полученный биоматериал разделяли на фракции сперматозоидов и семенной жидкости методом центрифугирования в градиенте плотностей, из сперматозоидов выделяли нуклеиновые кислоты и методом полимеразной цепной реакции в реальном времени определяли уровень экспрессии генов TLR2, TLR4, HSP60, HSP70, HSP90, IL1B, IL18, TNFA. Концентрацию пептида HSP70 в семенной жидкости определяли методом твердофазного иммуноферментного анализа, концентрацию цитокинов ИЛ-1β, ИЛ-18 и ФНОα -методом мультиплексного иммунофлуоресцентного анализа.Результаты. В ходе комплексного анализа показателей системы врожденного иммунитета в сперматозоидах и семенной жидкости пациентов с варикоцеле было продемонстрировано повышение уровней экспрессии генов TLR2 и TLR4 в сперматозоидах пациентов с варикоцеле в 6,9 и в 4,18 раза соответственно, гиперэкспрессия генов белков теплового шока HSP60, HSP70 и HSP90 -лигандов TLR2/4, увеличение уровней экспрессии генов цитокинов TNFA, IL1B, IL18 и содержание их белковых продуктов в семенной жидкости по сравнению с группой здоровых доноров эякулята. Продемонстрировано, что дисбаланс факторов врожденного иммунитета в эякуляте мужчин с варикоцеле коррелирует с нарушениями функциональных параметров сперматозоидов.Заключение. В ходе исследования выявлена неоднозначная роль факторов врожденного иммунитета в эякуляте мужчин при варикоцеле. Toll-подобные рецепторы сперматозоидов необходимы для распознавания патогенов и обеспечения локальной иммунной защиты, а белки теплового шока выполняют множество функций, обеспечивающих сперматогенез и оплодотворение. Однако гиперэкспрессия этих факторов ассоциирована со снижением как фертильных свойств сперматозоидов, так и их количества в эякуляте.
