История вопроса В 1957 г. была сформулирована гипотеза, согласно которой проводником биологического действия соматотропного гормона (СТГ) в организме является циркулирующий в крови «сульфирующий фактор», содержание которого в крови увеличивается под действием СТГ. Несколько лет спустя в крови людей была обнаружена инсулиноподобная активность, которая не подавлялась антителами к инсулину. Позднее проводники биологического действия СТГ стали называться соматомединами [12, 13]. В периферических тканях именно ИФР-I обеспечивает практически все физиологические эффекты соматотропного гормона. Очистка и исследование веществ, определяющих неподавляемую антителами инсулиноподобную активность крови, закончились выделением двух пептидов и установлением их аминокислотных последовательностей [21, 32, 33]. Они проявили высокое сходство с проинсулином и были названы инсулиноподобными ростовыми факторами в связи с их способностью стимулировать поглощение глюкозы мышечной и жировой тканью аналогично инсулину. Рецепторы и сигнальный поток Выделены два вида инсулиноподобных факторов роста: ИФР-I и ИФР-II. Они представляют собой одноцепочечные полипептиды, состоящие из 70 и 67 аминокислотных остатков соответственно. Степень сходства между двумя этими гормонами достигает 62%, причем аминокислотные остатки в каждом из них на 50% идентичны таковым в инсулине.
