Οι μυκοτοξίνες είναι δευτερογενείς μεταβολίτες των μυκήτων που αναπτύσσονται στα τρόφιμα και εμφανίζουν μεγάλο ερευνητικό ενδιαφέρον λόγω των τοξικών ιδιοτήτων τους. Η αφλατοξίνη Β1 (ΑΦΒ1) και η ωχρατοξίνη Α (ΩΤΑ) έχουν καρκινογόνες, τερατογόνες, ανοσοτοξικές και ενδεχομένως νευροτοξικές ιδιότητες. Η ΑΦΒ1 χαρακτηρίζεται ως η πιο επικίνδυνη φυσικώς παραγόμενη χημική ένωση, ενώ η ΩΤΑ έχει συνδεθεί με την εμφάνιση νεφροπάθειας στους ανθρώπους. Στόχος της παρούσας διατριβής είναι η μελέτη της επίδρασης της ακτινοβολίας-γ και του όζοντος στη βιοσύνθεση της ΑΦΒ1 και της ΩΤΑ στην Κορινθιακή σταφίδα, με σκοπό να ελαχιστοποιηθεί ο ενδεχόμενος κίνδυνος για τους καταναλωτές. Αρχικά έγινε ανάπτυξη και επικύρωση μεθόδων προσδιορισμού της αφλατοξίνης Β1 και της ωχρατοξίνης Α στη σταφίδα, με χρήση στηλών ανοσοσυγγένειας για τον καθαρισμό - απομόνωση και υγρής χρωματογραφίας υψηλής πίεσης (HPLC) με ανιχνευτή φθορισμού (FD) για τον προσδιορισμό των τοξινών. Η ανάκτηση τηςμεθόδου για την ΑΦΒ1 ήταν 97,6%, ενώ το όριο ανίχνευσης (DL) και το όριο ποσοτικοποίησης (QL) ήταν 0,045ng/g και 0,091ng/g, αντίστοιχα. Επιπλέον, η ανάκτηση της μεθόδου για την ΩΤΑ ήταν 104%, ενώ το όριο ανίχνευσης (DL) και το όριο ποσοτικοποίησης (QL) ήταν 0,26ng/g και 0,51ng/g, αντίστοιχα. Στη συνέχεια μελετήθηκε η επίδραση της ακτινοβολίας-γ και του όζοντος στη βιοσύνθεση της ΑΦΒ1 από το μύκητα Aspergillus parasiticus και της ΩΤΑ από το μύκητα Aspergillus carbonarius. Έπειτα μελετήθηκε η επίδραση της ακτινοβολίας-γ και του όζοντος στη μείωση πρότυπης ΑΦΒ1 και πρότυπης ΩΤΑ, σε δείγματα σταφίδας. Tα αποτελέσματα έδειξαν πως μία ασφαλής δόση ακτινοβολίας-γ ή όζοντος, μπορεί να είναι αποτελεσματική στην αναστολή της βιοσύνθεσης της ΑΦΒ1 από το μύκητα Aspergillus parasiticus (65,3% και 90,9% μείωση, αντίστοιχα) και της ΩΤΑ από το μύκητα Aspergillus carbonarius (>81,9% και 61,6% μείωση, αντίστοιχα), όπως και στη μείωση των ίδιων των τοξινών, σε θρεπτικό υπόστρωμα σταφίδας.
