Flowering time, maturity, and plant morphology have considerable effects on the adaptation and grain yield of soybean (Glycine max). The identification of novel genes and an understanding of their molecular basis are critical to improve soybean productivity. In this study, we cloned a flowering time related APETALA2-like gene GmTOE4a and generated GmTOE4a-overexpressing lines in the cultivar Williams 82. The transgenic lines exhibited late flowering both under long day and short day conditions, and repressed the flowering-related genes, including GmFT2a, GmFT5a, GmAP1, and GmLFY, whereas the flowering repressors GmFT4 and miR156 were upregulated. Interestingly, GmTOE4a was also mediated by photoperiod via maturity genes E3 and E4, which encode photoreceptors in soybean. Further, miR172-mediated GmTOE4a, which regulates flowering in soybean, is different in Arabidopsis in that it is dependent on the CONSTANS-like gene GmCOL1a. In addition to its effect on flowering time, GmTOE4a regulated plant morphology, increased stem thickness, and reduced plant height, internode length and leaf size, which are important agronomic traits that enhance the capacity to resist lodging and increase soybean yield. This is useful information to understand the molecular mechanism of flowering time and plant morphology in soybean and will greatly influence soybean yield improvement.
We studied the development of excitatory synapses in cultured neurons dissociated from the cortices of rat embryos at the 18th day of gestation (E18) and rat pups at birth (P0). Between 7 and 14 days in vitro (DIV), large increases in the amplitudes and frequencies of the spontaneous excitatory postsynaptic currents (EPSCs) of both cultured E18 and P0 neurons were observed. The EPSCs of E18 neurons were mediated primarily by alpha-amino-3-hydroxy-5-methyl-4-iso-xazole-propionic acid (AMPA) receptors at 7 DIV and by both N-methyl-D-aspartate (NMDA) and AMPA receptors at 14 DIV. Consistently, immunostaining indicated significant increases in the proportion of the clusters of NR1, an NMDA receptor subunit, which were associated with the accumulation of synaptophysin, a presynaptic marker, in cultured E18 neurons between 7 and 14 DIV. The proportion of NR1 clusters residing in synaptic regions and the proportion of synapses that colocalized with NR1 clusters in 7-day-old P0 neurons were not different statistically from those found in 7-day-old E18 neurons. However, cultured P0 neurons at 7 DIV displayed clear EPSCs mediated by NMDA receptors. Our results suggest that the targeting of NMDA receptors to synaptic regions lag behind the synaptic clustering of AMPA receptors during the in vitro development of cultured rat E18 cortical neurons. The results further suggest that the cortical neurons at P0 differ from those at E19 in certain cellular properties; as a result, the currents mediated by the synaptic NMDA receptors in 7-day-old P0 neurons are larger than those mediated by the synaptic NMDA receptors in 7-day-old E18 neurons.
Backgroud: Breast milk is the optimal food for infant growth and development. The purpose of this study was to evaluate the association between breast milk macronutrient composition with dietary pattern among lactating women. Methods: A total of 220 lactating women from 2011 to 2012 in Changsha, a city of south-central China, was recruited using a multi-stage sampling method. Breast milk was collected, and the protein, fat, lactose, total dry matter, and energy contents of breast milk were measured. A 24 h recall method on three consecutive days was used to collect the dietary information of lactating women and an exploratory factor analysis was performed was to identify dietary patterns. The association between the concentration of a breast milk component and dietary pattern was assessed using a multivariable linear regression model. Results: Three major dietary patterns were classified. Lactating women with dietary pattern 1 mainly ate fresh vegetables and fresh legumes. Those with dietary pattern 2 mainly ate red meat, cereals and eggs, and those with dietary pattern 3 mainly ate fungi and algae, dries legumes and soy milk. Pattern 2 was positively associated with the concentration of protein (B = 0.07, 95% CI 0.00, 0.15), total dry matter (B = 0.20, 95% CI 0.02, 0.38) and energy (B = 1.66, 95% CI 0.03, 3.30) in breast milk. Morever, lactation period was negatively associated with the protein and total dry matter concentrations and positively associated with lactose. Conclusions: The results show the lactation period was an important factor affecting milk composition and a dietary pattern with high intake of red meat, cereals, and eggs was associated with higher protein, total dry matter, and energy contents in breast milk. These findings show that the dietary patterns of lactating women can affect breast milk macronutrient composition and provide a foundation for improving child health.
The "all carbon" organic solar cells (OSCs) based on the homocyclic molecule tetraphenyldibenzoperiflanthene (DBP) as a donor and C60 as an acceptor were comprehensively characterized. The optimized planar-mixed heterojunction device with a DBP:C60 mixture ratio of DBP : C60 (1 : 2) exhibited a power conversion efficiency of 4.47%. To understand why DBP possesses such advantageous characteristics, the correlations of the morphology, molecular stacking, carrier dynamics and performance of DBP:fullerene-based devices have been systematically studied. First, the face-on stacked DBP molecules could enhance both the absorption of light and the charge carrier mobility. Second, DBP : C60 (1 : 2) thin films with optimized domain sizes and partially interconnected acceptor grains led to the most balanced carrier mobility and the lowest bimolecular recombination in devices. Finally, the DBP molecules were found to stack closely using grazing incidence wide-angle X-ray scattering measurements, with a π-π stacking spacing of 4.58 Å, indicating an effective molecular orbital overlap in DBP. The study not only reveals the promising characteristics of DBP as a donor in OSCs but the clear correlations of the thin-film nano-morphology, molecular stacking, carrier mobility and charge recombination found here could also provide insights into the characterization methodology and optimization of the small molecule OSCs.
