To investigate the case about room air pollution by microorganism from some region of Gannan Medical University and the effect of disinfectant made from traditional chinese medicine on room air. By flat natural precipitation method, the six sampling zones which were classroom, microorganism teaching laboratory, teacher’s office, the corridor of out-patient department, in-patient department hall and department of dermatology were selected to observe the case about room air pollution before disinfectant and the disinfection effect after disinfected with disinfection maken from traditional chinese medicine which named KZY-2 composed by Anemarrhena asphodeloides and Rhizoma coptidis, effect of disinfectant of KZY-2 on teaching laboratory was compared with ultraviolet ray by counting colonies of bacteria and fungi respectively. Total number of colony of the sampling zones from most to least was the corridor of out-patient department, department of dermatology, in-patient department hall, classroom, teaching laboratory, teacher’s office, number of fungal colony was department of dermatology, the corridor of out-patient department, in-patient department hall, teaching laboratory, classroom, teacher’s office, the average number of colony from the sampling sites of hospital (6764.3 cfu/m3) was significantly higher than campus (2190.7 cfu/m3),P<0.01. Total number of colony of the sampling sites was significantly decreased after disinfected with KZY-2 and ultraviolet ray. The sterilizing rate of KZY-2 was average 82.9% and up to 90.1%, the sterilizing rate of ultraviolet ray was average 63.5%,the disinfection effect between KZY-2 and ultraviolet ray was differential (P<0.05). These results indicate that KZY-2 and ultraviolet ray all have degermation on air and the disinfection effect of KZY-2 is superior to ultraviolet ray.
To investigate the efficacy of the compound preparation named KZY-2 which include the effective group formula of anemarrhena asphodeloides bunge and rhizoma coptidis on the fluconazole and ifraconazo resistant strains of Candida albicans respectively in vitro. The fluconazole and ifraconazo resistant strains of Candida albicans were sieved by drug sensitive scrip test and the diameter of inhibition zone was measured adopting the method of agar plate diffusion, reference to the M27-A recommended by the United States National Laboratory Standards Committee (NCCLS), the minimal inhibitive concentration(MIC,mg/m1) of KZY-2 against the the standard strain(ATCC32354) and the fluconazole and ifraconazo resistant strains of candida albicans was measured respectively. Results showed that the 12 fluconazole and ifraconazo resistant strains which named RC1 to RC12 were sieved from the 20 strains Candida albicans isolated from clinic, the diameters of inhibited zone of KZY-2 against ATCC32354 was 32mm and against RC1~RC12 was between 12mm and 28mm, the MIC of KZY-2 against ATCC32354 was 312.5μg/m1 and against RC1~RC12 was between 625μg /m1 and 5000μg /m1. These results indicate that KZY-2 has good antifungi efficacy on the standard strain and the fluconazole and ifraconazo resistant strains of Candida albicans in vitro.
To investigate the efficacy of the compound preparation of traditional Chinese drug which is named KZY-2) on system fungi reside in faecal specimen of pigeon, the 66 faecal specimen of pigeon were collected from columbary of local park and residents.Each specimen which weigh 2.0g was mixed with 10ml sterile saline and the supernatant which volume was 1ml was incubated with the same volume drug which concentration were 200mɡ/ml,100mɡ/ml and 50mɡ/ml respectively at 37°C.After 24h,48h and 72h, the isolated positive rates of Cryptococcus neoformans and Candida albicans were calculated. Candida albicans was identified by gram staining, budding development, chlamydospore formation, sugar fermented test,sugar assimilated test and Cryptococcus neoformans was identified by gram staining,sugar fermented test,sugar assimilated test,urease tests,caffeic acid test, Hiss capsule staining.Results showed that the isolated positive rates of Cryptococcus neoformans and Candida albicans were 27.3%(18/66) and 16.7%(11/66) without drug action, KZY-2 could reduce the isolated positive rates of Cryptococcus neoformans and Candida albicans(P<0.05 and P<0.05).The drug concentration and action time could significantly affect the isolated positive rate which was negative correlation between them, higher drug concentration and longer incubation time,more lower the positive rate, especially the isolated positive rates of Cryptococcus neoformans are reduced 77.7%(6.1/27.3)which incubated with 200mɡ/ml drug after 72h compared with the control group.These results indicate The KZY-2 have good germicidal efficacy on Cryptococcus neoformans and Candida albicans isolated from faecal specimen of pigeon.
