Glutathione S-transferases (GSTs) are the enzymes that defend cells against the damage mediated by oxidant and electrophilic carcinogens. GSTπ (GSTP1) is a member of the GST family and the hypermethylation GSTP1 CpG island DNA is detected in human hepatocellular carcinoma (HCC) tissues, which contributes to the negative expression of GSTP1 mRNA and protein. GSTP1 expression is considered to be an early event in HCC. Stat3, a member of the signal transduction and activator of transcription (Stat) family, is important for promoting the proliferation, survival and other biological processes of cells triggered by cytokines and growth factors. Activated Stat3 may participate in oncogenesis. Previous studies have demonstrated that overexpression of phosphorylated Stat3 is important in the proliferation of HCC cells, suggesting that disturbance of the Stat3 pathway may be an early event. We hypothesize that the suppression of GSTP1 expression in HCC cells increases Stat3 activation. In order to test this hypothesis, HepG2 cells were genetically modified to transiently express high levels of GSTP1. The transient expression of GSTP1 specifically downregulated epidermal growth factor (EGF)-mediated tyrosine phosphorylation of Stat3, and subsequently suppressed the transcriptional activity of Stat3. By contrast, GSTP1 RNAi was able to lead to an increase in the phosphorylation of Stat3. In addition, overexpression of GSTP1 was capable of reducing the survival of HepG2 cells and inducing cell cycle arrest. This inhibition was mediated by a direct interaction between GSTP1 and Stat3. Overall, our results suggest that GSTP1 is important in the regulation of the transcriptional activity of Stat3, and that it is also a regulator of the cell cycle via EGF signaling.
This article documents the addition of 139 microsatellite marker loci and 90 pairs of singlenucleotide polymorphism sequencing primers to the Molecular Ecology Resources Database. Loci were developed for the following species: Aglaoctenus lagotis, Costus pulverulentus, Costus scaber, Culex pipiens, Dascyllus marginatus, Lupinus nanus Benth, Phloeomyzus passerini, Podarcis muralis, Rhododendron rubropilosum Hayata var. taiwanalpinum and Zoarces viviparus. These loci were cross-tested on the following species: Culex quinquefasciatus, Rhododendron pseudochrysanthum Hay. ssp. morii (Hay.) Yamazaki and R. pseudochrysanthum Hayata. This article also documents the addition of 48 sequencing primer pairs and 90 allele-specific primers for Engraulis encrasicolus. et al.
c-aminobutyric acid (GABA) is an important bioactive regulator, and its biosynthesis is primarily through the a-decarboxylation of glutamate by glutamate decarboxylase (GAD). The procedures to obtain GABA by bioconvertion with high activity recombinant Escherichia coli GAD have been seldom understood. In this study, Escherichia coli GAD (gadA) was highly expressed (about 70-75% of total protein) as soluble protein in Escherichia coli BL21(DE3) containing pET28a-gadA, which was induced by 0.4 mM IPTG in LB medium, and maximal GABA-forming activity of the recombinant GAD was 40 U/mL at a concentration (0.15 mM) of pyridoxal phosphate (PLP) and a concentration (0.6 mM) of Ca 2? at optimal pH of 3.8. The optimal concentration (7.5 mM) of Mn 2? can also improve the activity of recombinant enzyme, but the co-effect of Ca 2? and Mn 2? exhibited antagonism effect when added simultaneously. LB and 0.1% (w/v) lactose were selected as culture medium and inducer, respectively. The relative activity was markedly higher activated by Ca 2? (174%), Mn 2? (164%) than that by other seven bivalent cations. Finally, the yield of GABA was high of 94 g/L detected by paper chromatography or HPLC in 1 L reaction system with 30 mL crude GAD (12 U/mL). By entrapping Escherichia coli glutamate decarboxylase into sodium alginate and carrageenan gel beads, the activity of immobilized GAD (IGAD) remained 85% during the initial five batches and the activity still remained 50% at the tenth batch, these results indicated that the recombinant Escherichia coli GAD was feasible for the future industrial production of GABA.
Abstract2D organic–inorganic perovskites are an emerging class of materials with great potential for optoelectronics since a wide variety of large functional chromophores can be regularly incorporated. Among this new type of materials, hybrid perovskite systems incorporating strong electron acceptor molecules are considered as a promising approach to designing a new type of functional 2D perovskites for optoelectronics. In this work, a rare example of organic–inorganic 2D perovskite incorporating strong acceptors such as naphthalene diimide (NDI) building blocks between inorganic sheets is presented. This hybrid architecture forms highly air‐stable thin films with a structure consisting of inorganic perovskite monolayers of metal‐halide octahedra separated by bilayers of NDI‐based organic cations. The presence of strong electron‐accepting moieties in this multifunctional donor–acceptor hybrid heterostructure leads to a rare type II heterojunction in which the excitons can be efficiently dissociated via the electron‐transfer process and in which holes and electrons can be easily confined in the inorganic and organic sublayers, respectively. Such an ultimate p–n heterojunction shows improved photoconduction properties with a photocurrent multiplied by ≈40 under white‐light illumination in comparison to a similar 2D perovskite structure containing optically and electrically inert alkyl chains as organic components.
In this study, we consider a scheduling problem with truncated exponential sum-of-logarithm-processing-times based and position-based learning effects on a single machine. We prove that the shortest processing time (SPT) rule is optimal for the makespan minimization problem, the sum of the θth power of job completion times minimization problem, and the total lateness minimization problem, respectively. For the total weighted completion time minimization problem, the discounted total weighted completion time minimization problem, the maximum lateness minimization problem, we present heuristic algorithms (the worst-case bound of these heuristic algorithms are also given) according to the corresponding single machine scheduling problems without learning considerations. It also shows that the problems of minimizing the total tardiness, the total weighted completion time and the discounted total weighted completion time are polynomially solvable under some agreeable conditions on the problem parameters.
