The genome of the pea aphid Acyrthosiphon pisum lacks genes thought to be crucial in other insects for recognition, signaling and killing of microbes.

Acute toxicity of thirty lectins was tested against the pea aphid Acyrthosiphon pisum (Harris) (Homoptera, Aphididae: Macrosiphini). Activity was measured on artificial diets containing moderate concentrations of lectins (10–250 μg/ml) by scoring mortality and growth inhibition over the whole nymphal period (7 days at 20°C). Most of the proteins tested exhibited low toxicity, but some induced significant mortality; these included the lectins from jackbean (Concanavalin A), amaranth, lentil and snowdrop. There was no direct correlation between toxicity and sugar specificity of the lectin; however, many mannose‐binding lectins were toxic towards A. pisum. Concanavalin A was also tested on five other aphid species (Aphis gossypii, Aulacortum solani, Macrosiphum euphorbiae, Macrosiphum albifrons and Myzus persicae) at concentrations between 10–1500 μg/ml. Mortality was very variable from one species to another. Strong growth inhibition invariably occurred within this concentration range, although dose‐response curves differed substantially between aphid species. The peptidase complement of A. pisum's digestive tract was also investigated, as well as the oral toxicity of some protease inhibitors (PIs) to this aphid. Most protein PIs were inactive, and no part of the digestive tract contained detectable amounts of endo‐protease activity. This is in contrast to the strong amino‐peptidase activity which was shown to occur predominantly in the midgut and crop portions of the digestive tract. The potential of lectins in transgenic crops to confer Host‐Plant Resistance to aphids is discussed.

A green biotype of the pea aphid, A. pisum, from Lusignan (France), showed very poor performance on the standard Akey and Beck diet. Significant improvement occurred after reduction of the osmotic pressure of the diet and modification of the amino acid component, according to the results of carcass analysis. A further improvement was obtained through an optimalization of the aromatic amino acid level: the tyrosine deficiency induced by its poor solubility was overcome by an excess of phenylalanine and the inclusion of a soluble tyrosine compound, β-alanyltyrosine. A new diet was therefore formulated for this biotype, on which adults were more than twice as heavy as on the standard diet.

The feeding behaviour of cassava mealybug (Phenacoccus manihoti) was analysed by the electrical penetration graph technique (EPG, DC‐system) and by light microscopy. This study confirmed a typical phloem‐feeding behaviour with an aphid‐like predominance of extracellular pathways of stylets. Similarities of EPGs from mealybugs with those of aphids and whiteflies allowed adoption of standard pattern labelling. The main differences with aphid EPGs consisted of fewer but longer intracellular punctures (pd duration of 20 s vs 5–7 s for aphids), longer times to the first phloem‐ingestion period and a lower motility of stylets within the phloem searching process. Comparison of the feeding behaviour on 6 plant types, including two occasional hosts (Talinum and poinsettia), one cassava hybrid and three common cassava cultivars (Manihot esculenta), allowed differentiation of penetration profiles. Intracellular punctures were shorter on occasional hosts, on which phloem finding and subsequent ingestion were markedly delayed although a great variability within plants persisted. Chemical analysis of some secondary plant substances revealed that none of the plants tested contained detectable amounts of alkaloids, and that cyanides were restricted to true hosts (cassavas and Manihot hybrid). Levels of total flavonoids did not differ between hosts and non‐hosts, in contrast to those of phenolic acids. Preliminary correlations with EPG patterns are discussed. Résumé Le comportement alimentaire de la cochenille du manioc (Phenacoccus manihoti) a été étudié par électrographie de pénétration (EPG, analyse en courant continu) et par microscopie optique. Cette étude a permis de confirmer le caractère typiquement phloémophage de l'alimentation de cet Homoptère, présentant une large prédominance du trajet extracellulaire des stylets, comme cela est également observé chez les pucerons. Les similitudes des tracés EPG effectués sur cochenille et sur pucerons ou aleurodes ont permis d'adopter la même typologie du signal. Les principales différences avec les pucerons concernent le nombre moins important et la durée plus longue des ponctions intracellulaires (20 s en moyenne contre 5 à 7 s chez les pucerons), l'allongement du temps d'accès au phloème et une moins grande mobilité des stylets lors de la phase de recherche du phloème. La comparaison du comportement alimentaire sur 6 plantes, parmi lesquelles deux hôtes occasionnels (Talinum et poinsettia), un hybride et trois variétés courantes de manioc (Manihot esculenta), a permis de différencier plusieurs profils de pénétration. Les ponctions intracellulaires se sont révélées plus courtes sur hôtes occasionnels, qui induisent également, malgré la variabilité observée, un allongement net de la période de localisation du phloème et donc un retard à l'ingestion. Four Bacillus thuringiensis ␦-endotoxins, Cry3A, Cry4Aa, Cry11Aa, and Cyt1Aa, were found to exhibit low to moderate toxicity on the pea aphid, Acyrthosiphon pisum, in terms both of mortality and growth rate. Cry1Ab was essentially nontoxic except at high rates. To demonstrate these effects, we had to use exhaustive bufferbased controls.

Dickeya dadantii (Erwinia chrysanthemi) is a phytopathogenic bacterium causing soft rot diseases on many crops. The sequencing of its genome identified four genes encoding homologues of the Cyt family of insecticidal toxins from Bacillus thuringiensis, which are not present in the close relative Pectobacterium carotovorum subsp. atrosepticum. The pathogenicity of D. dadantii was tested on the pea aphid Acyrthosiphon pisum, and the bacterium was shown to be highly virulent for this insect, either by septic injury or by oral infection. The lethal inoculum dose was calculated to be as low as 10 ingested bacterial cells. A D. dadantii mutant with the four cytotoxin genes deleted showed a reduced per os virulence for A. pisum, highlighting the potential role of at least one of these genes in pathogenicity. Since only one bacterial pathogen of aphids has been previously described (Erwinia aphidicola), other species from the same bacterial group were tested. The pathogenic trait for aphids was shown to be widespread, albeit variable, within the phytopathogens, with no link to phylogenetic positioning in the Enterobacteriaceae. Previously characterized gut symbionts from thrips (Erwinia/Pantoea group) were also highly pathogenic to the aphid, whereas the potent entomopathogen Photorhabdus luminescens was not. D. dadantii is not a generalist insect pathogen, since it has low pathogenicity for three other insect species (Drosophila melanogaster, Sitophilus oryzae, and Spodoptera littoralis). D. dadantii was one of the most virulent aphid pathogens in our screening, and it was active on most aphid instars, except for the first one, probably due to anatomical filtering. The observed difference in virulence toward apterous and winged aphids may have an ecological impact, and this deserves specific attention in future research.Dickeya dadantii (syn. The common symptoms for D. dadantii infection, as well as other so-called pectinolytic soft rot Erwinia, are characterized by the rapid disorganization of parenchymatous tissues, mainly driven by pectic enzymes (38).
