Zinc Pyrithione (ZnPT) dapat dianalisis kadarnya dengan berbagai metode, salah satunya dengan kromatografi cair kinerja tinggi (KCKT). Peneliti terdahulu telah mengembangkan metode analisis ZnPT dengan KCKT fase terbalik sebagai kompleks copper pyrithione (CuPT), tetapi setelah diaplikasikan di laboratorium, metode ini tidak memenuhi syarat kesesuaian sistem karena koefisien variasi area lebih dari 2%. Penelitian ini bertujuan untuk menentukan kondisi optimum untuk menentukan kadar ZnPT dalam sediaan sampo dengan metode KCKT melalui variasi perbandingan komposisi dan pH fase gerak, variasi laju alir, dan variasi suhu kolom. Dari hasil penelitian diperoleh bahwa kondisi optimum untuk analisis ZnPT dilakukan pada panjang gelombang 240 nm, menggunakan kolom Licrocart Lichrospher RP-18, fase gerak campuran larutan dapar fosfat 0,2 M pH 6,0 : asetonitril (60 : 40), laju alir 1,0 mL/menit, suhu kolom 25°C dan volume injeksi 10 µL. Hasil validasi metode analisis pada kondisi optimum mempunyai selektivitas yang baik dan menunjukkan hubungan yang linear antara luas area dibawah kurva dengan konsentrasi pada rentang 3,08-24,62 µg/mL dan koefisien korelasi (r 2 ) sebesar 0,995. Perolehan kembali hasil uji akurasi sebesar 99-101% pada konsentrasi CuPT diatas 12,31 µg/mL dan simpangan baku relatif hasil uji presisi sebesar 0,65%. Kondisi optimum ini efektif digunakan untuk analisis ZnPT dalam sediaan sampo dengan konsentrasi diatas 12,31 µg/mL..
