Response of Toyonoka strawberry to AgNO3 was studied. Types and combinations of plant growth regulators had significant effects on shoot regeneration efficiency. Explants cultured for 10 days in shoot regeneration medium in the presence of AgNO3 not only enhanced shoot regeneration efficiency but also expedited the initiation of adventitious buds. Highest regeneration (87.38%) and number of shoots per explant (11.67) were achieved in shoot regeneration media containing 1.5 mg·L–1 TDZ, 0.4 mg·L–1 IBA and 1.0 mg·L–1 AgNO3. Half-strength MS containing 1.0 mg·L–1 AgNO3 was an optimum medium for rooting. AgNO3 advanced root emergence and increased percent rooting, root length, dry weight and activity. Lower concentrations of AgNO3 inhibited ethylene production and promoted shoot regeneration and growth. It had a significant stimulatory effect on chlorophyll, soluble protein contents and antioxidant enzyme activities. Chlorophyll and soluble protein contents, superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activities were increased in the presence of AgNO3 and reached maximum at 1.0 mg·L–1 AgNO3. Root water content, superoxide free radicals (O2.-), malondialdehyde (MDA) content, proline accumulation and IAA-oxidase activity in leaves were increased while (IAA) content was decreased in the presence of AgNO3. A peltate-leaved species of Begonia was recently collected from a limestone hill in central Guangxi, China. Based on a careful study of literature, herbarium material, and plants both in the wild and in cultivation reveals that it is a new species of Begonia sect. Coelocentrum, which we hereby describe as B. ufoides. In addition to a full taxonomic account, color plates and line drawings are provided to aid in identification. Begonia ufoides may be confused with B. umbraculifolia Y. Wan & B.N. Chang in southwestern Guangxi and B. phuthoensis H.Q. Nguyen in northern Vietnam, both of which are peltate-leaved calciphilous species in sect. Coelocentrum, but the new species is clearly distinguishable in many morphological characters. A table comparing salient features of the three species is presented.
The objective of this study was to establish an efficient and reproducible in vitro plant regeneration for Citrullus lanatus cv. Zaojia. To achieve optimal conditions for adventitious shoot induction, five explants (entire cotyledons, distal cotyledons, proximal cotyledons, cotyledonary node A and cotyledonary node B) were tested on MS medium supplemented with different concentrations and combinations of growth regulators (0 to 0.2 mg/L IAA and 1.0 to 5.0 mg/L BA), the results showed that entire cotyledons cultured in MS + BA (2.0mg/L) + IAA(0.2mg/L) achieved the highest regenerated rate (89.67%) and the optimal protocol screened in this experiment had 7.69 ± 0.10 shoots per explants. Adventitious shoots were able to elongate both on MS medium with 0.2 mg/L KT and 0.2 mg/L NAA; IBA 0.3mg/L was found to be effective in the production of root. Acclimatized plantlets transferred to pot resumed growth, and their stems and leaves elongated and expanded in one month.
