SummaryEmerging evidence indicates that natural killer (NK) cells may contribute to liver injury in patients with hepatitis B virus (HBV) infection. Because HBV infection progresses through various disease phases, the cytolytic profiles of peripheral and intrahepatic NK cells in HBV-infected patients remain to be defined. In this study, we comprehensively characterized intrahepatic and peripheral NK cells in a cohort of HBV-infected individuals, and investigated their impact on liver pathogenesis during chronic HBV infection. The study population included 34 immuneclearance (IC) patients, 36 immune-tolerant (IT) carriers and 10 healthy subjects. We found that the activity of peripheral NK cells from IC patients was functionally elevated compared to IT carriers and controls, and NK cell activation was indicated by an increased expression of CD69, CD107a, interferon (IFN)-g and tumour necrosis factor (TNF)-a. Further analysis showed that the increased activity of both peripheral and hepatic NK cells was correlated positively with liver injury, which was assessed by serum alanine aminotransferase levels (ALT) and the liver histological activity index (HAI). Interestingly, the frequency of peripheral NK cells was reduced in IC patients (especially those with higher HAI scores of 3-4), but there was a concomitant increase in hepatic NK cells. The functionally activated NK cells are enriched preferentially in the livers of IC patients and skew towards cytolytic activity that accelerates liver injury in chronic hepatitis B (CHB) patients.

Tilapias are important aquaculture fishes that have been introduced widely all over the world, often carrying their monogenean parasites with them. An extensive investigation on monogeneans of invasive tilapias was conducted in 19 natural water sources in south China between July 2015 and December 2017. We found nine known species of monogeneans, i.e., Enterogyrus coronatus, E. malmbergi, Cichlidogyrus cirratus, C. halli, C. sclerosus, C. thurstonae, C. tilapiae, Scutogyrus longicornis, Gyrodactylus cichlidarum, and one unknown Gyrodactylus species. In addition to reporting ten new hosts and four new geographical records, we observed new morphological characteristics of these species. Observation on living specimens of Enterogyrus spp. demonstrated that these two species have characteristic opisthaptoral retraction capacities, while the opisthaptor glands were not observed in our specimens of E. coronatus and E. malmbergi. The morphological differences of the accessory piece of the male copulatory complex between C. cirratus and C. mbirizei (character for species differentiation) could result from the observation at different perspectives, which indicates that C. mbirizei is likely a synonym of C. cirratus. A more detailed structure of the sclerotized parts of Cichlidogyrus spp. and S. longicornis were revealed by scanning electron microscopy. As was the case for the monogeneans found on alien tilapias from other geographic regions, the present study confirmed the high potential of these monogeneans to establish populations in new habitats.

Object counting in aquaculture is an important task, and has been widely applied in fish population estimation, estimation of lobster abundance and scallop stocks, and so forth. However, underwater object counting is challenging for biologists and marine scientists because of the diversity of backgrounds of the lake or ocean, the uncertainty of the object motion, and the occlusion between objects. With the rapid development of sensor, computer vision, and acoustic technologies, advanced and efficient counting methods are available in aquaculture. We reviewed underwater object counting methods in aquaculture, provided a survey including more than 50 articles in the recent 10 years, and analyzed the pros and cons of the counting methods and the applicable scenarios of those methods. Finally, the major challenges and future trends of underwater object counting in aquaculture are discussed.
