Field undisturbed tension-free monolith lysimeters and 15 N-labeled urea were used to investigate the fate of fertilizer nitrogen in paddy soil in the Taihu Lake region under a summer rice-winter wheat rotation system. We determined nitrogen recovered by rice and wheat, N remained in soil, and the losses of reactive N (i.e., NH 3 , N 2 O, NO 3 − , organic N and NH 4 + ) to the environment. Quantitative allocation of nitrogen fate varied for the rice and wheat growing seasons. At the conventional application rate of 550 kg N ha −1 y −1 (250 kg N ha −1 for wheat and 300 kg N ha −1 for rice), nitrogen recovery of wheat and rice were 49% and 41%, respectively. The retention of fertilizer N in soil at harvest accounted for 29% in the wheat season and for 22% in the rice season. N losses through NH 3 volatilization from flooded rice paddy was 12%, far greater than that in the wheat season (less than 1%), while N leaching and runoff comprised only 0.3% in the rice season and 5% in the wheat season. Direct N 2 O emission was 0.12% for the rice season and 0.14% for the wheat season. The results also showed that some dissolved organic N (DON) were leached in both crop seasons. For the wheat season, DON contributed 40-72% to the Nleaching, in the rice season leached DON was 64-77% of the total N leaching. With increasing fertilizer application rate, NH 3 volatilization in the rice season increased proportionally more than the fertilizer increase, N leaching in the wheat season was proportional to the increase of fertilizer rate, while N 2 O emission increased less in proportion than fertilizer increase both in the rice season and wheat season.
Polyphenols in whole grain wheat have potential health benefits, but little is known about the expression patterns of phenolic acid biosynthesis genes and the accumulation of phenolic acid compounds in different-colored wheat grains. We found that purple wheat varieties had the highest total phenolic content (TPC) and antioxidant activity. Among phenolic acid compounds, bound ferulic acid, vanillic, and caffeic acid levels were significantly higher in purple wheat than in white and red wheat, while total soluble phenolic acid, soluble ferulic acid, and vanillic acid levels were significantly higher in purple and red wheat than in white wheat. Ferulic acid and syringic acid levels peaked at 14 days after anthesis (DAA), whereas p-coumaric acid and caffeic acid levels peaked at 7 DAA, and vanillic acid levels gradually increased during grain filling and peaked near ripeness (35 DAA). Nine phenolic acid biosynthesis pathway genes (TaPAL1, TaPAL2, TaC3H1, TaC3H2, TaC4H, Ta4CL1, Ta4CL2, TaCOMT1, and TaCOMT2) exhibited three distinct expression patterns during grain filling, which may be related to the different phenolic acids levels. White wheat had higher phenolic acid contents and relatively high gene expression at the early stage, while purple wheat had the highest phenolic acid contents and gene expression levels at later stages. These results suggest that the expression of phenolic acid biosynthesis genes may be closely related to phenolic acids accumulation.
Fusarium oxysporum f. sp. cubense (Foc), the causal agent of Fusarium wilt (Panama disease), is one of the most devastating diseases of banana (Musa spp.). The Foc tropical race 4 (TR4) is currently known as a major concern in global banana production. No effective resistance is known in Musa to Foc, and no effective measures for controlling Foc once banana plants have been infected in place. Early and accurate detection of Foc TR4 is essential to protect banana industry and guide banana planting. A real-time fluorescence loop-mediated isothermal amplification assay (RealAmp) was developed for the rapid and quantitative detection of Foc TR4 in soil. The detection limit of the RealAmp assay was approximately 0.4 pg/µl plasmid DNA when mixed with extracted soil DNA or 103 spores/g of artificial infested soil, and no cross-reaction with other relative pathogens were observed. The RealAmp assay for quantifying genomic DNA of TR4 was confirmed by testing both artificially and naturally infested samples. Quantification of the soil-borne pathogen DNA of Foc TR4 in naturally infested samples was no significant difference compared to classic real-time PCR (P>0.05). Additionally, RealAmp assay was visual with an improved closed-tube visual detection system by adding SYBR Green I fluorescent dye to the inside of the lid prior to amplification, which avoided the inhibitory effects of the stain on DNA amplification and makes the assay more convenient in the field and could thus become a simple, rapid and effective technique that has potential as an alternative tool for the detection and monitoring of Foc TR4 in field, which would be a routine DNA-based testing service for the soil-borne pathogen in South China.
To evaluate the physiological responses of wheat to zinc (Zn) fertilizer application under drought stress, pot, and field experiments were conducted on wheat plants grown under different soil moistures and treated with soil and foliar Zn applications. Photosynthetic characteristics, antioxidant content, Zn element concentration, and the transcription level of genes involved in antioxidant biosynthesis were analyzed. Zn application increased SPAD and Fv/Fm of wheat flag leaves, while decreased lipid peroxidation levels and H2O2 content. Zn application increased the antioxidant content (ascorbate, reduced glutathione, total phenolic, and total flavonoid) of wheat flag leaves, and enhanced the relative expression levels of two antioxidant enzyme genes, four ascorbate–glutathione cycle genes, and two flavonoid biosynthesis pathway genes under drought stress. Soil Zn application increased grain yield and Zn concentration by 10.5 and 15.8%, 22.6 and 9.7%, and 28.2 and 32.8% under adequate water supply, moderate drought, and severe drought, respectively. Furthermore, foliar application of Zn in the field increased grain yield and grain Zn concentration under both adequate water supply and rain-fed conditions. Zn plays a role in alleviating wheat plant drought stress by Zn-mediated increase in photosynthesis pigment and active oxygen scavenging substances, and reduction in lipid peroxidation. Furthermore, Zn fertilizer could regulate multiple antioxidant defense systems at the transcriptional level in response to drought.
Two-year (June 2003-May 2005 precipitation samples were collected from three monitoring sites with similar economy development level in the Yangtze River Delta Region of China to investigate the spatial-temporal variation of inorganic N wet deposition. The results showed that the Yangtze River Delta Region had higher inorganic N wet deposition than the northwestern, northern, or southern China. There was spatial variation of inorganic N wet deposition. The positive relationship between inorganic N deposition and precipitation suggested that rainfall amount might be an important factor influencing the wet deposition of inorganic N. Deposition of inorganic N occurred mainly in the spring and summer season (March-August; 70% of the annual total), which was in accord with seasonal distribution of precipitation. However, a negative logarithmic relation between rainfall and inorganic N concentration in rainwater indicated the dilution effect of rainwater on inorganic N concentration. Distinguished variation of NH 4 + /NO 3 − ratio in wet deposition existed in the different time scale. NH 4 + /NO 3 − ratio evidently decreased from 6 in 1980s to 1.2 in 2003/ 2005 and greatly varied between 0.3 and 9.9 within a year. NH 4 + /NO 3 − ratio peaked in accordance with nitrogen-fertilizing time during crop growing season. Annual alternate appearance of the 15 N-enriched and 15 N-depleted periods coincided with the temporal variation of NH 4 + /NO 3 − ratio, which was closely related to the timing of fertilization and seasonal climate changes, suggesting the effect of NH 4 + sources in the wet deposition.
Little information is available describing the effects of exogenous H2S on the ABA pathway in the acquisition of drought tolerance in wheat. In this study, we investigated the physiological parameters, the transcription levels of several genes involved in the abscisic acid (ABA) metabolism pathway, and the ABA and H2S contents in wheat leaves and roots under drought stress in response to exogenous NaHS treatment. The results showed that pretreatment with NaHS significantly increased plant height and the leaf relative water content of seedlings under drought stress. Compared with drought stress treatment alone, H2S application increased antioxidant enzyme activities and reduced MDA and H2O2 contents in both leaves and roots. NaHS pretreatment increased the expression levels of ABA biosynthesis and ABA reactivation genes in leaves; whereas the expression levels of ABA biosynthesis and ABA catabolism genes were up-regulated in roots. These results indicated that ABA participates in drought tolerance induced by exogenous H2S, and that the responses in leaves and roots are different. The transcription levels of genes encoding ABA receptors were up-regulated in response to NaHS pretreatment under drought conditions in both leaves and roots. Correspondingly, the H2S contents in leaves and roots were increased by NaHS pretreatment, while the ABA contents of leaves and roots decreased. This implied that there is complex crosstalk between these two signal molecules, and that the alleviation of drought stress by H2S, at least in part, involves the ABA signaling pathway.
