Human chorionic gonadotropin (hCG) has been used to induce ovulation and as a luteotrophic agent in cattle. However, the effect of hCG therapy on the functional status of the equine corpus luteum (CL) is unclear. This study aimed to characterize the hemodynamic and secretory function of early CL of mares treated with different doses of hCG at distinct stages of the estrous cycle. Mares were assigned to nine experimental groups (n ¼ 6 mares/ group) according to dose of hCG and time of treatment. A single injection of one of three different doses of hCG (750, 1,500, or 2,500 IU) was performed in one of three distinct stages of the estrous cycle: preovulatory follicle !35 mm, day of ovulation (D0), or 48 hours after ovulation (D2). In addition, a control group treated with NaCl 0.9% was included in the study. The end points evaluated daily from D0 to D8 were area of the CL, luteal vascularity, number of colored pixels and total pixel intensity, and concentrations of plasma progesterone (P4). No effect (P > .1) of dose or time of treatment was observed for any end point, within each day. Luteal area did not differ throughout the days (P > .1), whereas Doppler parameters and concentrations of plasma P4 presented a progressive increase (P < .05) after ovulation in all groups. Secretory function and luteal hemodynamic were not affected (P > .1) by hCG dose and time of treatment. In conclusion, hCG therapy during estrus or early diestrus, at the doses tested, did not improve P4 secretion or luteal blood flow.
Improved understanding of the molecular mechanisms underlying ascending equine placentitis holds the potential for the development of new diagnostic tools and therapies to forestall placentitis-induced preterm labor. The current study characterized the equine placental transcriptome (chorioallantois; CA and endometrium; EN) during placentitis (placentitis group, n = 6) in comparison to gestationally-matched controls (control group, n = 6). Transcriptome analysis identified 2953 and 805 differentially expressed genes in CA and EN during placentitis, respectively. Upstream regulator analysis revealed the central role of toll-like receptors (TLRs) in triggering the inflammatory signaling, and consequent immune-cell chemotaxis. Placentitis was associated with the upregulation of matrix metalloproteinase (MMP1, MMP2, and MMP9) and apoptosis-related genes such as caspases (CASP3, CASP4 and CASP7) in CA. Also, placentitis was associated with downregulation of transcripts coding for proteins essential for placental steroidogenesis (SRD5A1 and AKR1C1), progestin signaling (PGRMC1 and PXR) angiogenesis (VEGFA, VEGFR2 and VEGFR3), and nutrient transport (GLUT12 and SLC1A4), as well as upregulation of hypoxia-related genes (HIF1A and EGLN3), which could explain placental insufficiency during placentitis. Placentitis was also associated with aberrant expression of several placenta-regulatory genes such as, PLAC8, PAPPA, LGALS1, ABCG2, GCM1, and TEPP, which could negatively affect placental functions. In conclusion, our findings revealed for the first time the key regulators and mechanisms underlying placental inflammation, separation, and insufficiency during equine placentitis, which might lead to the development of efficacious therapies or diagnostic aids by targeting the key molecular pathways.
O principal fator a limitar o processo de criopreservação de sêmen equino está relacionado à própria espécie, por apresentar uma grande variabilidade nas características dos ejaculados pós-descongelação. O objetivo desta pesquisa foi avaliar a viabilidade espermática pós-descongelação, do sêmen de garanhões das raças Quarto de Milha e Mangalarga Marchador utilizando-se dois diluentes (Tris e Botu-crio®) na criopreservação. Para tanto, foram analisadas as características físicas do sêmen fresco, funcionalidade da membrana espermática pelo Teste Hiposmótico (HOST), motilidade total e vigor pelo Teste de Termoresistência (TTR), motilidade progressiva pelo sistema computadorizado CASA (Computer Assisted Sêmen Anlyses) e integridade da membrana espermática, do acrossoma e funcionalidade de mitocôndria por microscopia de epifluorescência. O diluente Botu-crio® preservou melhor os valores de motilidade e vigor e a integridade de membrana plasmática, pós-descongelamento. Não houve diferença significativa entre as raças quanto à motilidade total e vigor no sêmen fresco, bem como para a análise desses parâmetros no Teste de Termoresistência pós-descongelamento. Garanhões da raça Quarto de Milha apresentaram percentual maior de defeitos maiores na análise de patologia espermática.

The present study characterized the luteal status and the dynamic of the conceptus during the first 20 days of gestation in mares with different ages and degrees of endometrial degeneration. Total area of the corpus luteum (CL), luteal vascularity, CL area with blood signals, progesterone concentrations (P4), embryonic vesicle diameter, number of embryonic location changes, embryonic fixation position and uterine contractility were evaluated. In Experiment 1, mares ≤6 years of age (Young group, 5.6 ± 0.2 years, n = 7 mares) and mares ≥15 years of age (Old group, 17.2 ± 0.9 years, n = 6 mares) were used to investigate the effect of age. In Experiment 2, the luteal and embryonic parameters were compared between mares with minimal (Mild group, endometrial category I, n = 9 mares) and severe (Severe group, endometrial category III, n = 7 mares) endometrial degeneration. The Old and Severe groups had greater (p ≤ 0.04) total CL area and reduced luteal vascularity (p ≤ 0.04) than the Young and Mild groups, respectively. However, P4 levels and CL area with blood signals were similar (p ≥ 0.8) between the groups. A negative effect of age (p < 0.01), but not of endometrial degeneration (p = 0.6), was found for the embryonic vesicle diameter. The conceptus mobility was high (p > 0.1) until day 14 of gestation in the Severe group, while a reduced number of changes of the embryo location was detected earlier (p < 0.05) in the Old group. In conclusion, the newly formed CL of aged mares and mares with severe endometrial degeneration suffered a structural remodelling to safeguard the local blood supply and the continuous P4 output during early gestation. Moreover, an earlier reduction of the embryonic mobility and a delayed development of the conceptus were associated with advanced age, regardless of the degree of endometrial degeneration.
The aim of the current project was to characterize the luteal vascularity and the plasma concentrations of progesterone (P4), prolactin (PRL) and 13,14-dihydro-15-keto-PGF (PGFM) in mares with luteal disturbances during early and mid-diestrus. In Experiment 1, twenty-one mares were treated with 2 mL of 0.9% NaCl, or 1 mg Dinoprost, or 10 mg Dinoprost on day two after ovulation (Control-D2, 1/10PGF-D2 and PGF-D2 groups, respectively; n = 7 mares/group). In Experiment 2, similar treatments were performed eight days post-ovulation using a different cohort of 21 mares (Control-D8, 1/10PGF-D8 and PGF-D8 groups, respectively; n = 7 mares/group). Blood samples were collected hourly and power-Doppler examinations of the corpus luteum (CL) were performed every 6 h from H0 (moment immediately before treatment) to H48. Data collection was also done once a day from D0 (day of ovulation) to D20. In Experiment 1, the PGF-D2 and 1/10PGF-D2 groups had lower increase of plasma concentration of P4 until H48 and reduced maximum P4 concentrations on D8-D11 than mares from the Control-D2 group. However, no differences among groups were detected for luteal vascularity during early and mid-diestrus. In Experiment 2, complete and partial luteolysis were detected in mares from the PGF-D8 and 1/10PGF-D8 groups, respectively. Luteal vascularity and plasma P4 concentrations differed among Control-D8, PGF-D8 and 1/10PGF-D8 groups on H48. Partially regressed CLs (1/10PGF-D8 group) generated more Doppler signals than completed regressed CLs (PGF-D8 group) between D10 and D13. In both experiments, a transient increase in PRL activity was observed in parallel to the PGFM pulse in mares receiving 1 or 10 mg Dinoprost. The use of prostaglandin on D2 at conventional or 1/10 of the dose impaired the luteal development in mares. Moreover, the low dose of prostaglandin lead to partial regression of mature CLs. The blood supply was reduced in partially regressed CLs, but not in CLs undergoing impaired luteogenesis.
