In this in vitro study, we assessed the antibacterial efficacy of four endodontic sealers-resin AH26, EndoRez, calcium hydroxide (Apexit), and pure zinc oxide-against Enterococcus faecalis. The agar diffusion test was employed to evaluate the antibacterial efficacy of the sealers in vitro, with distilled water serving as a control. The sealers were prepared following the manufacturer's instructions and placed in wells of 50 agar plates, each inoculated with 15 samples of Kocuria rhizophila and Staphylococcus aureus. Inhibition zones were assessed after 72, 120, and 168 hours of anaerobic incubation at 37°C for 196 hours. Kruskal-Wallis and Friedman tests were used for data analysis. Positive control plates exhibited bacterial growth in all specified periods. AH26 demonstrated significantly higher antibacterial effectiveness against both bacterium types compared to the other sealers (P<0.01). Pure zinc oxide exhibited moderate antibacterial activity, while Apexit and EndoRez showed the lowest activity against S. aureus and no activity against K. rhizophila. AH26 had the highest antibacterial effect, and EndoRez had the lowest (P<0.05). In terms of inhibiting bacterial growth, the effectiveness of root canal sealers was ranked as follows: AH26 > Pure Zinc Oxide > Apexit/EndoRez.
Introduction. Dental caries is one of the most common infectious diseases of the oral cavity, and is an inflammatory disease caused by several factors. Interleukin-1β (IL-1β) is a major mediator of acute inflammation and essential for the development of specific immune responses. The objective of this study was to assess the levels of secretory IgA (s-IgA) and IL-1β in the saliva of smokers with dental caries, and to discover the correlation between these parameters and dental caries. Methods. Saliva samples were collected from 30 smokers, age range 21–70 years, with dental caries, in addition to 18 healthy non-smoker volunteers, age range 21–65 years. s-IgA and IL-1β levels in the saliva samples were estimated by enzyme-linked immunosorbent assay (ELISA). Results. The mean saliva IgA levels between smokers with dental caries group and healthy subjects were not significantly different (p=0.077), while the saliva levels of IL-1β were higher in the smokers with dental caries group, with a significant difference of p<0.05. No significant associations were found between s-IgA and IL-1β levels, and other parameters such as age, sex, body mass index (BMI) and C-reactive protein (CRP) (p>0.05). There were highly positive associations and significant differences between IL-1β and CRP levels in the two groups under the study (p=0.006). Conclusions. Our study revealed a significant increase in IL-1β levels in saliva of smokers with dental caries, and a positive association between IL-1β levels and caries disease. There is no significant relationship between elevated IL-1β levels and s-IgA in smokers with dental caries.

Objective: to assess micro-shear bond strength (μSBS) of two universal adhesives (UA) having two etching aggressiveness applied to dentin in a self-etch mode, immediately and after thermal ageing.Methods: the occlusal enamel of twenty-eight sound human premolar teeth were removed to expose a flat dentin surface for bonding procedures. Teeth were randomly assigned to one of two groups according to the adhesive; the first group (Peak-Universal: PU) has intermediate etching aggressiveness (1.2 pH) and the second group (All-Bond-Universal: ABU) has ultra-mild etching aggressiveness (3.2 pH). After bonding procedures as per manufacturer instructions, a nanohybrid composite resin was placed and light cured. All specimens were stored in demineralized water at 37˚C for 24 hours. Half of the specimens were evaluated for μSBS testing after 24 hours of water storage at 37˚ C and the other half were evaluated after thermal ageing using10000 thermal-cycles and sixmonths water storage, then observed by SEM. μSBS in (MPa) were compared with analysis of variance. Kolmogorov-Smirnov and Shapiro-Wilk tests were applied to assess normality of distributions (P < 0.05 was considered as significant).Results: PU showed statistically significant higher mean μSBS than AU, immediately and after thermal ageing. Thermal ageing and water storage did not significantly influence PU while bond degradation was significant in AU.Conclusion: with regard to the adhesive systems used in this study, when self-etch mode was used, UA with aggressive pH (intermediate) revealed better bonding effectiveness and stability against thermal aging and water storage than those with ultra-mild pH.Clinical relevance: Self-etch mode has a positive effect on the dentin bond durability of universal adhesives, as was seen with the previous generation of self-etch adhesives.
