Fidelity and efficiency of pre-mRNA splicing are critical for generating functional mRNAs, but how such accuracy in 5′ splice site (SS) selection is attained is not fully clear. Through a series of yeast genetic screens, we isolated alleles of prp28 that improve splicing of suboptimal 5′SS substrates, demonstrating that WT-Prp28p proofreads, and consequently rejects, poor 5′SS. Prp28p is thought to facilitate the disruption of 5′SS–U1 snRNA pairing to allow for 5′SS–U6 snRNA pairing in the catalytic spliceosome; unexpectedly, 5′SS proofreading by Prp28p is dependent on competition with the stability of the 5′SS:U6 duplex, but not the 5′SS:U1 duplex. E404K, the strongest prp28 allele containing a mutation located in the linker region between adenosine triphosphatase (ATPase) subdomains, exhibited lower RNA-binding activity and enhanced splicing of suboptimal substrates before first-step catalysis, suggesting that decreased Prp28p activity allows longer time for suboptimal 5′SS substrates to pair with U6 snRNA and thereby reduces splicing fidelity. Residue E404 is critical for providing high splicing activity, demonstrated here in both yeast and Drosophila cells. Thus, the subdomain linker in Prp28p plays important roles both in splicing efficiency across species and in proofreading of 5′SS.
Transcription and pre-mRNA splicing are coupled to promote gene expression and regulation. However, mechanisms by which transcription and splicing influence each other are still under investigation. The ATPase Prp5p is required for pre-spliceosome assembly and splicing proofreading at the branch-point region. From an open UV mutagenesis screen for genetic suppressors of prp5 defects and subsequent targeted testing, we identify components of the TBP-binding module of the Spt–Ada–Gcn5 Acetyltransferase (SAGA) complex, Spt8p and Spt3p. Spt8Δ and spt3Δ rescue the cold-sensitivity of prp5-GAR allele, and prp5 mutants restore growth of spt8Δ and spt3Δ strains on 6-azauracil. By chromatin immunoprecipitation (ChIP), we find that prp5 alleles decrease recruitment of RNA polymerase II (Pol II) to an intron-containing gene, which is rescued by spt8Δ. Further ChIP-seq reveals that global effects on Pol II-binding are mutually rescued by prp5-GAR and spt8Δ. Inhibited splicing caused by prp5-GAR is also restored by spt8Δ. In vitro assays indicate that Prp5p directly interacts with Spt8p, but not Spt3p. We demonstrate that Prp5p's splicing proofreading is modulated by Spt8p and Spt3p. Therefore, this study reveals that interactions between the TBP-binding module of SAGA and the spliceosomal ATPase Prp5p mediate a balance between transcription initiation/elongation and pre-spliceosome assembly.
Abstract. In order to solve the problem of shortage of water resources in northern Shaanxi, we selected rectangular large loop source transient electromagnetic method with high water affinity, and radioactive measurement method which can delineate the water storage structure, comprehensive geophysical prospecting methods to look for groundwater. Algorithm has established a forward model, and compared all-time apparent resistivity in late-time apparent resistivity is better than late. We can find out the exact location of the groundwater and thus improving wells rate by comparatively using these two kinds of geophysical prospecting method. Hydrogeology drilling confirmed water inflow of a single well can be up to 40, it can fully cover native Domestic and Agricultural water, and provide an important basis for groundwater exploration.
The metal source of gold deposits in the Jiaoxibei area, eastern China, has been investigated by many researchers, but no consensus has been reached so far. In this study, three typical gold deposits, the Xinli, Jiaojia, and Dayingezhuang deposits, were selected for trace element analysis of gold and pyrite to constrain the metal source. Pyrite from these three deposits has similar morphological and compositional characteristics, and can be divided into three types: Py1 with euhedral to subhedral textures, Py2 with subhedral to anhedral textures with micro‐fractures, and Py3 with subhedral to anhedral textures and intergrowing polymetallic sulfides. Among them, Py2 and Py3 were formed in the main ore‐forming stage and they are the dominant host minerals of visible gold. In these deposits, visible gold occurs mainly in micro‐fractures or as inclusions in Py2 and Py3. Most of the pyrite has extremely low concentration of invisible gold, indicating that visible gold in the Jiaoxibei district is not a product of the remobilization of invisible gold from earlier pyrite. Both Py2 and Py3 are characterized by low Co concentration of <100 ppm and Co/Ni ratio of <1, which are similar to those of pyrite in sedimentary rocks. Therefore, ore‐forming metals of these gold deposits in the Jiaoxibei district may originate mainly from a sediment‐related metal source.
Bioturbation plays a critical role in sediment mixing and biogeochemical cycling between sediment and seawater. An abundance of bioturbation structures, dominated by Thalassinoides, occurs in carbonate rocks of the Cambrian Series 2 Zhushadong Formation in the Dengfeng area of western Henan Province, North China. Determination of elemental geochemistry can help to establish the influence of burrowing activities on sediment biogeochemical cycling, especially on changes in oxygen concentration and nutrient regeneration. Results show that there is a dramatic difference in the bioturbation intensity between the bioturbated limestone and laminated dolostone of the Zhushadong Formation in terms of productivity proxies (Baex, Cu, Ni, Sr/Ca) and redox proxies (V/Cr, V/Sc, Ni/Co). These changes may be related to the presence of Thalassinoides bioturbators, which alter the particle size and permeability of sediments, while also increase the oxygen concentration and capacity for nutrient regeneration. Comparison with modern studies shows that the sediment mixing and reworking induced by Thalassinoides bioturbators significantly changed the primary physical and chemical characteristics of the Cambrian sediment, triggering the substrate revolution and promoting biogeochemical cycling between sediment and seawater.
Rock weathering is the main source of element geochemical cycle, which has a very important impact on the environment. Three well-developed basalt weathering profiles in Changbai Mountain area are selected in this study, and the samples of parent rock, parent material layer, sedimentary layer, and leaching layer are systematically collected. The systematic study showed that the basalt in the study area experienced moderate chemical weathering under warm and humid climate conditions, with leaching of some major elements such as silicon and aluminum. The distribution of the rare earth elements (REE) in the weathering products recalls that of an alkali-basalt, with an evident fractionation between light- and heavy-REE. Such a feature is therefore referable to the parent volcanic rock, although with some degree of leaching.
