Background: Cladophialophora bantiana is a dematiaceous fungus that causes phaeohyphomycosis, a generic term used to describe a variety of unusual mycoses caused by fungi that have melanin in their cell wall. C. bantiana targets the central nervous system, commonly causing localized brain infections that may result in disseminated infections. In Brazil, minimal phaeohyphomycosis data are available, and information about C. bantiana infections in animals, especially canines, is scarce. Thus, the aim of this study was to describe the clinical and pathological aspects of systemic phaeohyphomycosis caused by C. bantiana in a dog.Case: A 1-year-old female Pit Bull presented with weight loss, reduced appetite, and a history of cutaneous lesions on the right thoracic limb; however, clinical evolution was not reported. The dog had reportedly given birth recently. Physical examination revealed thinness, pale ocular and oral mucosa, submandibular lymph nodes, and enlarged popliteal lymph nodes. The animal died after convulsive crises during hospitalization. At necropsy, white-yellowish multifocal nodules were observed in the liver and right kidney. The brain featured left cerebral hemisphere asymmetry with blood vessel congestion in the leptomeninges and an irregular brownish focal area on the surface of the right occipital cortex. Cross-sections of the formalin-fixed brain exhibited compression of the left lateral ventricle and the presence of grayish and friable multifocal areas in the gray matter of the left parietal and right occipital cortices. Fragments of the lesions were collected for histopathological and microbiological examination. Histologically, the lesions were similar, characterized by hepatitis, nephritis, and granulomatous and necrotizing meningoencephalitis, multifocal to coalescing, accentuated, chronic, and associated with numerous pigmented fungi. Fontana-Masson–stained fungi exhibited a strong black color. In cleared and unstained histological slides, brownish pigmentation was observed in the cytoplasm and walls of the fungi. C. bantiana was identified via microbiological cultivation.Discussion: A diagnosis of phaeohyphomycosis caused by C. bantiana was made based on the characteristic morphology of the microscopic lesions and confirmed via isolation in microbiological culture. As numerous species cause phaeohyphomycosis, specific confirmation of the etiologic agent using several diagnostic techniques is necessary. In histopathological examinations, pigmented fungal organisms are easily seen among lesions. However, in some cases, the pigment is not apparent in the tissues. FM staining is necessary to demonstrate the presence of the melanin in fungi. As in most phaeohyphomycosis cases, it was not possible to determine the primary portal of entry. However, the lesion on the right thoracic limb probably favored the penetration of the agent. In addition to cerebral lesions, severe lesions in the hepatic and renal parenchyma were observed, which are characteristic of systemic mycosis. Infection and clinical diseases are usually associated with immunocompromised; here, the gestation period may have had an immunosuppressive effect, favoring the proliferation and dissemination of the agent. It was concluded that phaeohyphomycosis caused by C. bantiana produced severe systemic lesions in the brain and organs of the abdominal cavity. Although uncommon, phaeohyphomycosis caused by Cladophialophora bantiana should be included as a differential diagnosis for other canine diseases that present with similar clinical symptoms.Keywords: canine, fungal diseases, dematiaceous fungi, Cladosporium trichoides, Xylohypha bantiana, melanin, Fontana-Masson
<p>As neoplasias mamárias em gatas representam até 40% dos tumores encontrados nessa espécie, sendo o terceiro tipo mais incidente, seguido das neoplasias hematopoiéticas e cutâneas. Existem vários estudos sobre neoplasias de mama em cadelas, entretanto há poucos estudos relacionados a neoplasmas de mama em gatas, apesar da incidência do tumor mamário na espécie felina ser inferior a metade da incidência na espécie canina, a percentagem de neoplasias malignas é muito superior. O objetivo desse trabalho é descrever um caso de carcinoma cribriforme metastático em uma gata sem raça definida, com 10 anos de idade, que foi encaminhada para atendimento apresentando nódulos ulcerados na região das mamas inguinais há mais de seis meses. O diagnóstico de carcinoma cribiforme foi estabelecido com base nos achados anatomopatológicos.</p>
This study discusses the epidemiological, clinical, and pathological characteristics of two outbreaks of ocular infection in ostriches (Struthio camelus) caused by the trematode Philophthalmus sp. The outbreaks occurred in the states of Ceará and Rio Grande do Norte in Northeast Brazil. Ostrich farming was categorized as semi-intensive farming with free access to the weir and river. The clinical symptoms for infection included nictitating membrane inflammation, epiphora (tearing), eyelid edema, ocular congestion, eyeball destruction, emaciation, and death, and the infection period ranged from 8 to 16 months. Macroscopically, thickening of the eyelids and conjunctiva were observed, with orange parasites infestation. Histologically, the lesions were characterized as lymphoplasmacytic and heterophilic, multifocal to coalescing, moderate, chronic conjunctivitis associated with hyperplasia and ectasia of the Harderian gland, and numerous parasites. The parasites were oval and flattened with an oral suction cup, acetabulum, and thick cuticle containing spicules. The diagnosis of philophthalmiasis was based on epidemiological, clinical, and pathological findings associated with the morphological characteristics of the parasites present in the conjunctival sacs which were similar to the trematode Philophthalmus sp. Philophthalmiasis can occur in ostriches raised on urban and rural properties in Northeastern Brazil, probably associated with the contact of birds with contaminated water, and is characterized by severe conjunctivitis that can result in the loss of the eyeball and death of the animals.
<p>Descreve-se um caso de onfaloflebite em uma bezerra de cinquenta dias de idade atendida na Clínica Médica e Cirúrgica de Grandes Animais. O animal apresentava sinais clínicos de diarreia e onfalite. Foi instituída terapia antimicrobiana e anti-inflamatória, além de correção da desidratação, porém o animal não apresentou melhora e foi a óbito, sendo posteriormente encaminhado ao Laboratório de Patologia Animal para realização de necropsia. Na necropsia foram observadas lesões absedativas no parênquima pulmonar, hepático e artrite séptica. O exame histopatológico revelou múltiplos abscessos no pulmão e fígado, com centro necrótico e miríades de agregados bacterianos circunscritos por acentuado infiltrado inflamatório neutrofílico e confirmando o quadro de septicemia, em consequência da onfaloflebite. O diagnóstico foi realizado através de coleta de dados epidemiológicos, clínicos e patológicos.</p>
Background: Melanosis is a blackened pigmentation resulting from the accumulation of melanocytes in tissues that are not normally pigmented. This change in the color of the organs occurs due to the agglomeration of melanocytes originating from abnormal migration during embryogenesis and does not cause dysfunction to the affected organ. Although melanosis frequently occurs in several species and affects several organs such as the brain and spinal cord leptomeninges, involvement in the thalamus region is unusual. The objective of this work was to report two cases of thalamic melanosis in goats, determining the pathological and histochemical aspects that assist in the diagnosis of this condition.Cases: Two cases of thalamic melanosis in goats were diagnosed. In both cases, the animals had no nervous history disease and clinical signs. The cause of death in cases 1 and 2 was established based on anatomopathological findings and clinical signs being diagnosed with mycoplasmosis and asphyxia, respectively. After fixing and making cross-sections of the brain, a focal lengthy blackened area was observed on the thalamus surface in both cases. Microscopically, lesions in the brain were similar in both cases and exclusively affected the thalamus. These cells had abundant cytoplasm, well delimited with brownish granular pigment. The nuclei were difficult to visualize and in some cells, it was rounded, well-defined, morphologically compatible with melanocytes. Melanocytes were mainly distributed around neurons and often distended the perivascular space of multiple blood vessels. In Fontana Masson staining, the granules in the cytoplasm of these cells stained strongly black. The Prussian Blue, Periodic Acid- Schiff's, Von Kossa, and Giemsa stains were negative, and the pigment remained brown. In the unstained slides, assembled after the deparaffinization and clarification process, it was observed the permanence of cells with blackish-brown pigment in the cytoplasm. In immunohistochemistry, strong immunostaining of pigmented cells with the Anti-MelanA antibodies was observed in both cases.Discussion: The diagnosis of thalamic melanosis in goats was carried out based on the characteristic pathological findings, in which melanin pigments were demonstrated and identified through HE, Fontana-Masson staining, and unstained slides and confirmed by the IHC. The use of complementary histochemical techniques was fundamental for the classification of the pigment as melanin, demonstrating to be an accessible and reliable tool for the diagnosis of pathological processes that lead to the accumulation of pigments and or material in the tissues. The occurrence of melanin in the thalamus may be associated with a failure in the migration of melanoblasts, which would go to the optical pathways or to the thalamus. This erratic migration of melanoblasts can be explained by the fact that the forebrain is the embryogenic origin of the optic and diencephalon pathways. Macroscopically, thalamic melanosis must be differentiated mainly from neoplastic processes such as melanoma and hemangiosarcoma, pigmented fungus infections, Phalaris angusta poisoning, listeriosis, neurocutaneous melanosis, and neuromelanin. It was concluded that thalamic melanosis is an uncommon alteration in goats and although it has been diagnosed as an incidental necropsy finding, should be included in the differential diagnosis of diseases that affect the central nervous system, especially those that have a color change associated with the deposition of pigments in the tissues. We described the clinical and anatomopathological findings observed in a case of nasal leiomyosarcoma in a five-year-old male Quarter Mile horse, whose main complaints were decreased sports performance and bilateral purulent nasal discharge. The nodule was observed in the nasal cavity, obstructing the left nostril and associated with purulent drainage. The nodule was of irregular shape and yellow color, measuring 19.4 cm × 6.9 cm × 4.3 cm in size, with coalescent multifocal areas that were brownish, friable, opaque, and fetid. When cut, the surface was compact, grayish-white, and smooth with yellow, friable, irregular multifocal areas, measuring 1-3.2 cm in diameter. Histopathological examination showed spindle-shaped neoplastic cells, which was negative on Masson’s trichromic stain. A diagnosis of leiomyosarcoma was established based on the morphotintorial aspects of neoplastic cells and confirmed through immunohistochemistry, with positive immunostaining for antibodies 1A4, HHF35, desmin, and S100. Leiomyosarcoma primarily affects the nasal cavity of horses and should be included in the differential diagnosis of diseases that affect the nasal cavity and cause nasal obstruction associated with dyspnea.
This study describes the epidemiological, clinical, and pathological aspects of spontaneous and experimental poisoning by nitroxinil at 34% concentration in goats. The outbreak occurred on a farm in the municipality of Prata, Paraíba state. Nitroxinil was administered to a herd of 120 goats, of which 18 presented with anorexia, vocalization, abdominal distension, weakness, staggering, and falls. Necropsy of three goats revealed that the main lesion was acute liver injury. Histologically the liver showed centrilobular necrosis associated with hemorrhage and hepatocyte degeneration. In the kidneys, tubular nephrosis with granular cylinder formations was observed. The lungs showed multifocal to coalescent areas of moderate interalveolar edema and vascular congestion. Experimental poisoning was carried out in two goats, with the same medication and doses administered on the farm. The experimental goats showed clinical signs and macroscopic and histological changes similar to the spontaneously poisoned goats. The diagnosis of nitroxinil poisoning was made based on epidemiological, clinical, and pathological data, and confirmed by experimental poisoning. The administration of nitroxinil in high doses, associated with high ambient temperature and physical exercises, can cause poisoning with high lethality in goats.
Spontaneous and experimental poisoning by Tephrosia cinerea in the northeastern semiarid region of Brazil has only been described in sheep. Pathologically, such poisoning leads to ascites and centrilobular liver fibrosis. However, these effects require an experimental study in goats. This study aimed to determine the goats’ susceptibility to the ingestion of T. cinerea and the minimum toxic dose, describing the main clinical and anatomopathological findings. Poisoning was reproduced experimentally in one sheep that received 10g/kg of the ground plant and in two goats, the first receiving a dose of 5g/kg and the second receiving 10g/kg of the ground plant. The sheep presented abdominal distension 34 days after beginning the ingestion of the plant, developing sternal decubitus, breathing difficulty, opisthotonos, mandibular trismus, salivation, dysphagia, vocalization, and pedaling movements on the 50th day of the experiment. Fluid accumulation was observed in the abdominal cavity and liver via necropsy, with an irregular, slightly whitish capsular surface. Histologically, the main lesions observed in the liver were moderate fibrosis, marked sinusoidal distension, accompanied by marked hemorrhage, sometimes forming bridges between the centrilobular regions, associated with a dissociation of hepatocyte cords. There were discrete Alzheimer’s type II astrocytes in the gray matter in the region of the occipital cortex in the nervous system. Goat 2 showed apathy, drowsiness, and weight loss; on the 62th day, lateral decubitus evolved to sternal decubitus, with a rotation of the neck towards the flank. At necropsy, marked edema was observed on the face and dewlap, and a slight accumulation of liquid; slightly yellowish material was observed in the abdominal cavity. There were discrete blackened areas on the capsular surface in the liver. Histologically, the liver showed mild centrilobular fibrosis associated with mild dissociation of hepatocyte cords and mild vacuolar degeneration of the hepatocyte cytoplasm. Goat 1 showed no clinical signs; at necropsy, discrete multifocal areas were observed in the liver on the capsular surface. Histologically, diffuse intracytoplasmic vacuolar degeneration of hepatocytes was detected. The clinical picture and anatomopathological findings differ between the species, proving the lower susceptibility of goats to Tephrosia cinerea ingestion (compared to sheep), with differences in the pathogenesis and epidemiological aspects of poisoning.
