El objetivo del presente estudio fue determinar la sobrevivencia in vitro y la fertilidad in vivo de espermatozoides recuperados de la cola del epidídimo de toros criollos post mortem. Se colectaron 18 pares de testículos de toros criollos adultos del camal Municipal El Collao-Ilave, posterior a su sacrificio. Cada par de testículos fueron transportados al laboratorio y almacenados aleatoriamente en refrigeración a 5°C durante 0(n=6), 12(n=6) y 24(n=6) h. Al final de cada periodo, los espermatozoides fueron colectados y evaluados antes de la congelación y después de la descongelación en motilidad total, motilidad progresiva, test hiposmótico e integridad de acrosoma, congelados con dilutor Tris-yema de huevo (10%)-glicerol (7%) en pajillas de 0,25 mL con 30x106 espermatozoides motiles/pajilla. La prueba de fertilidad in vivo se realizó en 13 vacas, evidenciando que las características espermáticas evaluadas se preservaron antes de la congelación, pero a la descongelación fueron afectados por efecto de la refrigeración. Producto del proceso de la congelación y los espermatozoides congelados-descongelados, tuvieron capacidad de fertilizar. Por tanto, los espermatozoides colectados son congelables y pueden aplicarse en técnicas reproductivas asistidas.
El objetivo del estudio fue evaluar el efecto del tipo de yema de huevo (codorniz-YC vs gallina-YG) sobre la calidad del semen de paco (Piaractus brachypomus) (volumen, color, concentración, motilidad, vitalidad y fertilidad) congelado. Se utilizaron 18 reproductores (15 machos y 3 hembras) cuya espermiación y ovulación, respectivamente, fue inducida con extracto de hipófisis de carpa. El semen fue colectado mediante masaje del abdomen del pez. El semen fresco fue evaluado y luego diluido con una solución crioprotetora que incluyó YC o YG. El semen diluido fue congelado y descongelado a los 30 días para la evaluación pos-descongelación y prueba de fertilidad. Se encontró una notoria disminución de los parámetros seminales por efecto de la congelación. La motilidad fue menor para la YC comparado a la YG (14.8 vs. 18.8%; p<0.05), mientras que la vitalidad (31 vs. 28.8%), tiempo de activación (1.4 vs. 1.3 min) y la fertilidad (18.4 vs. 20.4%) fueron similares entre grupos. En conclusión, la motilidad fue mejorada con la adición de yema de huevo de gallina, mientras que la vitalidad, tiempo de activación y fertilidad no fueron influenciados por el tipo de yema.
Trout production in Peru has export potential. However, there are no fry available to maintain production throughout the year and there is also a lack of egg production at certain times of the year. Therefore, the alternative is to cryopreserve semen for the non-reproductive season. The objective of the study was to determine the influence of freezing temperature on the quality of rainbow trout semen. The semen of 12 reproductive males was collected in the facilities of the Chucuito Research and Production Center of the National University of the Altiplano of Puno, which underwent a macroscopic, microscopic pre and post thaw seminal evaluation and the fertility rate was measured. Due to the effect of three freezing temperatures (-80 °C, -100 °C, -120 °C), cryopreservation had a decline curve of -20 °C / min. The fresh seminal parameters were similar to those reported by other researchers. While freezing had unfavorable effects on semen quality, the best results for activation time (51.33 sec) and vitality (35.33%) were obtained with -100 °C, but the higher motility was obtained with -120 °C (36.33%). Regarding fertility, the higher rate was obtained with -100 °C (70.97%), followed by -80 °C and -120 °C in which 68.86% and 64.34% were obtained, respectively. In conclusion, the results suggest that the tolerable freezing temperature of rainbow trout semen is around -100 °C, which is shown as a favorable alternative for the reproductive management of rainbow trout under natural hypobaric conditions of the Peruvian highlands
